A number of natural and synthetic flavonoids have been assessed previously with regard to their effects on the activity of cyclin-dependent kinases (Cdk1 and -2) related to the inhibition of cell cycle progression. On the other hand, the Cdk5/p35 system is of major importance in neuronal migration phenomena and brain development, and its deregulation is implicated in neurodegenerative diseases, particularly Alzheimer's. Here we show that some natural flavonoids inhibit the activity of the Cdk5/p35 system in the micromolar range, while others are practically inactive. Ring B-unsubstituted and highly methoxylated flavones were inactive or gave irreproducible results, and 6-methoxyapigenin and 6-methoxyluteolin were the most potent Cdk5 complex inhibitors within this series, while the common flavonols kaempferol and quercetin showed intermediate behavior. The reported crystal structure of the Cdk5 complex with its activator p25 was used for docking studies, which also led to the identification of the two 6-methoxyflavones, kaempferol and quercetin, as well as the untested 6-methoxy derivatives of kaempferol and quercetin and the corresponding 6-hydroxy analogues as compounds exhibiting a good fit to the active site of the enzyme.

Effects of natural flavones and flavonols on the kinase activity of Cdk5.

The anti-apoptotic Bcl-xL is a promising agent to prevent neurodegeneration in Parkinson's disease, which is characterized by a demise of dopaminergic neurons. We linked Bcl-xL to a peptide that allows its delivery across biological membranes and the blood-brain barrier. We tested the fusion protein in two models of Parkinson's Disease. Cell-permeable Bcl-xL protected neuroblastoma cells from the selective neurotoxin 1-methyl-4-phenylpyridinium. Furthermore, its systemic application in aged mice protected dopaminergic neurons following administration of MPTP as revealed by counting of tyrosine hydroxylase-immunoreactive neurons in the substantia nigra pars compacta. Hence, we present that a cell-permeable form of an anti-apoptotic protein can be delivered to CNS neurons through its systemic application, and we provide the proof that the delivery of this protein to the CNS neurons effectively prevents neuronal cell death in models of chronic neurodegenerative diseases.

Aggregates of alpha-synuclein are the pathological hallmark of sporadic Parkinson's disease (PD), and mutations in the alpha-synuclein gene underlie familial forms of the disease. To characterize the formation of alpha-synuclein aggregates in living cells, we developed a new strategy to visualize alpha-synuclein by fluorescence microscopy: alpha-synuclein was tagged with a six amino acid PDZ binding motif and co-expressed with the corresponding PDZ domain fused to enhanced green fluorescent protein (EGFP). In contrast to the traditional approach of alpha-synuclein-EGFP fusion proteins, this technique provided several-fold higher sensitivity; this allowed us to compare alpha-synuclein variants and perform time-lapse imaging. A C-terminally truncated alpha-synuclein variant showed the highest prevalence of aggregates and toxicity, consistent with stabilization of the alpha-synuclein monomer by its C-terminus. Time-lapse imaging illustrated how cells form and accumulate aggregates of alpha-synuclein. A substantial number of cells also reduced their aggregate load, primarily through formation of an aggresome, which could itself be cleared from the cell. The molecular chaperone Hsp70 not only prevented the formation of aggregates, but also increased their reduction and clearance, underlining the therapeutic potential of similar strategies. In contrast to earlier assumptions build-up, reduction and clearance of alpha-synuclein aggregation thus appear a highly dynamic process.

Streptococcus pneumoniae is the most frequent cause of bacterial meningitis, leading to permanent neurological damage in 30% and lethal outcome in 25% of patients. The cholesterol-dependent cytolysin pneumolysin is a major virulence factor of S. pneumoniae. It produces rapid cell lysis at higher concentrations or apoptosis at lower concentrations. Here, we show that sublytic amounts of pneumolysin produce rapid bundling and increased acetylation of microtubules (signs of excessive microtubule stabilization) in various types of cells--neuroblastoma cells, fibroblasts and primary astrocytes. The bundling started perinuclearly and extended peripherally towards the membrane. The effect was not connected to pneumolysin's capacity to mediate calcium influx, macropore formation, apoptosis, or RhoA and Rac1 activation. Cellular cholesterol depletion and neutralization of the toxin by pre-incubation with cholesterol completely inhibited the microtubule phenotype. Pharmacological inhibition of Src-family kinases diminished microtubule bundling, suggesting their involvement in the process. The relevance of microtubule stabilization to meningitis was confirmed in an experimental pneumococcal meningitis animal model, where increased acetylation was observed. Live imaging experiments demonstrated a decrease in organelle motility after toxin challenge in a manner comparable to the microtubule-stabilizing agent taxol, thus proposing a possible pathogenic mechanism that might contribute to the CNS damage in pneumococcal meningitis.

Rapid microtubule bundling and stabilization by the Streptococcus pneumoniae neurotoxin pneumolysin in a cholesterol-dependent, non-lytic and Src-kinase dependent manner inhibits intracellular trafficking.

The relation of alpha-synuclein (alphaS) aggregation to Parkinson's disease (PD) has long been recognized, but the mechanism of toxicity, the pathogenic species and its molecular properties are yet to be identified. To obtain insight into the function different aggregated alphaS species have in neurotoxicity in vivo, we generated alphaS variants by a structure-based rational design. Biophysical analysis revealed that the alphaS mutants have a reduced fibrillization propensity, but form increased amounts of soluble oligomers. To assess their biological response in vivo, we studied the effects of the biophysically defined pre-fibrillar alphaS mutants after expression in tissue culture cells, in mammalian neurons and in PD model organisms, such as Caenorhabditis elegans and Drosophila melanogaster. The results show a striking correlation between alphaS aggregates with impaired beta-structure, neuronal toxicity and behavioural defects, and they establish a tight link between the biophysical properties of multimeric alphaS species and their in vivo function.

Synaptic vesicles recycle repeatedly in order to maintain synaptic transmission. We have previously proposed that upon exocytosis the vesicle components persist as clusters, which would be endocytosed as whole units. It has also been proposed that the vesicle components diffuse into the plasma membrane and are then randomly gathered into new vesicles. We found here that while strong stimulation (releasing the entire recycling pool) causes the diffusion of the vesicle marker synaptotagmin out of synaptic boutons, moderate stimulation (releasing approximately 19% of all vesicles) is followed by no measurable diffusion. In agreement with this observation, synaptotagmin molecules labeled with different fluorescently tagged antibodies did not appear to mix upon vesicle recycling, when investigated by subdiffraction resolution stimulated emission depletion (STED) microscopy. Finally, as protein diffusion from vesicles has been mainly observed using molecules tagged with pH-sensitive green fluorescent protein (pHluorin), we have also investigated the membrane patterning of several native and pHluorin-tagged proteins. While the native proteins had a clustered distribution, the GFP-tagged ones were diffused in the plasma membrane. We conclude that synaptic vesicle components intermix little, at least under moderate stimulation, possibly because of the formation of clusters in the plasma membrane. We suggest that several pHluorin-tagged vesicle proteins are less well integrated in clusters.

Limited intermixing of synaptic vesicle components upon vesicle recycling.

Loss of dopaminergic neurons in the substantia nigra pars compacta and the resulting decrease in striatal dopamine levels are the hallmarks of Parkinson's disease. Tgfbeta and Gdnf have been identified as neurotrophic factors for dopaminergic midbrain neurons in vivo and in vitro. Haploinsufficiency for either Tgfbeta or Gdnf led to dopaminergic deficits. In this study we therefore analyzed the nigrostriatal system of aged Tgfbeta2 (+/-)/Gdnf (+/-) double-heterozygous mice. Unexpectedly, we found no morphological changes in the nigrostriatal system as compared with age-matched wild-type mice. There were no significant differences in the number of TH-positive midbrain neurons and no changes in the optical density of TH immunoreactivity in striata of Tgfbeta2 (+/-)/Gdnf (+/-) double-heterozygous mice. Moreover, we found no significant differences in the striatal levels of dopamine and its metabolites dihydroxyphenylacetic acid and homovanillic acid. Our results indicate that a combined haploinsufficiency for Tgfbeta2 and Gdnf has no impact on the function and the survival of midbrain DA neurons under normal aging conditions.

Unerwartet fanden wir keine morphologischen Ver&auml;nderungen im Nigrostriatal System im Vergleich zu Wildtyp-M&auml;usen altersbezogenen. Es gab keine signifikanten Unterschiede in der Anzahl der TH-positivem Mittelhirn Nervenzellen und keine &Auml;nderungen in die optische Dichte der TH Immunoreactivity in Striata von Tgfbeta2 (+/-) / Gdnf (+/-) Doppel-heterozygote M&auml;use. Dar&uuml;ber hinaus fanden wir keine signifikanten Unterschiede im Niveau Striatale Dopamin und seiner Metaboliten Dihydroxyphenylacetic S&auml;ure und Homovanillins&auml;ure. The dopamine transporter (DAT) mediates dopamine uptake in many brain areas and has been suggested to underlie dopamine release in the substantia nigra pars reticulata. This release modulates basal ganglia output and motor performance. It is triggered by glutamatergic subthalamic afferents. We show here that activation of (G(q)-coupled) group I metabotropic glutamate receptors are sufficient to induce dopamine release in rat substantia nigra pars reticulata slices. Release was measured by amperometry and inhibited by a DAT antagonist. In a DAT-expressing cell line, dopamine release could be induced by activation of G(q)-coupled muscarinic receptors. Release was measured by HPLC and inhibited by a DAT antagonist. In both paradigms, activation of protein kinase C (PKC) was necessary and sufficient for dopamine release, suggesting that G(q)-coupled receptors can induce DAT-mediated dopamine release through PKC, and providing a physiological role for the known regulation of DAT-mediated release by PKC. Activation of muscarinic receptors and PKC also induced release from a cell line expressing the norepinephrine transporter, suggesting that transport-mediated release might be relevant in further brain areas.

Neurotransmitter release is achieved through the fusion of synaptic vesicles with the neuronal plasma membrane (exocytosis). Vesicles are then retrieved from the plasma membrane (endocytosis). It was hypothesized more than 3 decades ago that endosomes participate in vesicle recycling, constituting a slow endocytosis pathway required especially after prolonged stimulation. This recycling model predicts that newly endocytosed vesicles fuse with an endosome, which sorts (organizes) the molecules and buds exocytosis-competent vesicles. We analyzed here the endosome function using hippocampal neurons, isolated nerve terminals (synaptosomes), and PC12 cells by stimulated emission depletion microscopy, photooxidation EM, and several conventional microscopy assays. Surprisingly, we found that endosomal sorting is a rapid pathway, which appeared to be involved in the recycling of the initial vesicles to be released on stimulation, the readily releasable pool. In agreement with the endosomal model, the vesicle composition changed after endocytosis, with the newly formed vesicles being enriched in plasma membrane proteins. Vesicle proteins were organized in clusters both in the plasma membrane (on exocytosis) and in the endosome. In the latter compartment, they segregated from plasma membrane components in a process that is likely important for sorting/budding of newly developed vesicles from the endosome.

Endosomal sorting of readily releasable synaptic vesicles.

Neurotransmitter-Freisetzung wird durch die Fusion der synaptischen Vesikel mit der neuronalen Plasmamembran (Exozytose) erreicht. Bl&auml;schen werden dann von der Plasmamembran (Endozytose) abgerufen. Es wurde vor mehr als 3 Jahrzehnten die Hypothese, dass sich Vesikel recycling, Endosomen an bilden einen langsamen Endozytose-Weg ben&ouml;tigt, vor allem nach l&auml;ngerer Stimulation. Dieses recycling Modell prognostiziert, dass neu Endocytosed Bl&auml;schen mit einem Endosom verschmelzen die sortiert (organisiert) die Molek&uuml;le und Knospen Exozytose-zust&auml;ndigen Vesicles. Wir analysierten hier die Endosom-Funktion mithilfe von Neuronen, isolierte Nervenenden (Synaptosomes) und PC12 Zellen durch stimulierte Emission Depletion Microscopy, Photooxidation EM und mehrere konventioneller Mikroskopie-Proben. &Uuml;berraschenderweise fanden wir, dass Endosomen sortieren einen schnellen Weg ist, die das recycling von der anf&auml;nglichen Bl&auml;schen &uuml;ber Stimulation, leicht l&ouml;sbare Pool freigegeben werden beteiligt zu sein schien. Im Einvernehmen mit den Endosomen-Modell &auml;nderte sich die Vesikel-Zusammensetzung nach Endozytose, mit der neu gebildeten Bl&auml;schen wird angereichertes Plasma Membranproteine. Vesikel-Proteine in Clustern sowohl in der Plasmamembran (auf Exozytose) organisiert und in das Endosom. Im letzteren Fach, sie getrennt von Plasmamembran Komponenten in einem Prozess, der wahrscheinlich ist wichtig f&uuml;r die Sortierung/Knospen von neu entwickelten Bl&auml;schen aus dem Endosom.

Neurotransmitter release relies on the fusion of synaptic vesicles with the plasma membrane of synaptic boutons, which is followed by the recycling of vesicle components and formation of new vesicles. It is not yet clear whether upon fusion the vesicles persist as multimolecular patches in the plasma membrane, or whether they segregate into individual components. Evidence supporting each of these two models has been suggested in recent years. Using diffraction-unlimited imaging (stimulated emission depletion, or STED) of native synaptic vesicle proteins, we have proposed that vesicle proteins remain in clusters on the neuronal surface. These clusters do not appear to intermix. We discuss here these findings in the context of previous studies on synaptic vesicle fusion, and we propose a recycling model which accounts for most of the recent findings on the post-fusion fate of synaptic vesicle components.

Nachweise f&uuml;r jedes dieser beiden Modelle wurde in den letzten Jahren vorgeschlagen. Mit Beugung-unbegrenzte Bildgebung (stimulierte Emission Ersch&ouml;pfung oder STED) native synaptische Vesikel Proteine, haben wir vorgeschlagen, dass Vesikel Proteine in Clustern auf der neuronalen Oberfl&auml;che bleiben. Diese Cluster erscheinen nicht zu vermischen. Hier diskutieren wir diese Erkenntnisse im Zusammenhang mit fr&uuml;heren Studien &uuml;ber synaptische Vesikel Fusion, und wir schlagen eine Wiederverwertung Modell welche Konten f&uuml;r den Gro&szlig;teil der j&uuml;ngsten Erkenntnisse &uuml;ber das post-fusion Schicksal der synaptischen Vesikel-Komponenten.
