The inner plexiform layer (IPL) of the vertebrate retina comprises functionally specialized sublaminae, representing connections between bipolar, amacrine and ganglion cells with distinct visual functions. Developmental mechanisms that target neurites to the correct synaptic sublaminae are largely unknown. Using transgenic zebrafish expressing GFP in subsets of amacrine cells, we imaged IPL formation and sublamination in vivo and asked whether the major postsynaptic cells in this circuit, the ganglion cells, organize the presynaptic inputs. We found that in the lak/ath5 mutant retina, where ganglion cells are never born, formation of the IPL is delayed, with initial neurite outgrowth ectopically located and grossly disorganized. Over time, the majority of early neurite projection errors are corrected, and major ON and OFF sublaminae do form. However, focal regions of disarray persist where sublaminae do not form properly. Bipolar axons, which arrive later, are targeted correctly, except at places where amacrine stratification is disrupted. The lak mutant phenotype reveals that ganglion cells have a transient role organizing the earliest amacrine projections to the IPL. However, it also suggests that amacrine cells interact with each other during IPL formation; these interactions alone appear sufficient to form the IPL. Furthermore, our results suggest that amacrines may guide IPL sublamination by providing stratification cues for other cell types.

Transient requirement for ganglion cells during assembly of retinal synaptic layers.

Neuronal function depends on the accurate wiring between pre- and postsynaptic cells. Determining the mechanisms underlying precision in neuronal connectivity is challenging because of the complexity of the nervous system. In diverse parts of the nervous system, regions of synaptic contact are organized into distinct parallel layers, or laminae, that are correlated with distinct functions. Such an arrangement enables the development of synapse specificity to be more readily investigated. Here, we present an overview of the developmental mechanisms that are thought to underlie the formation of synaptic layers in the vertebrate retina, a highly laminated CNS structure. We will contrast the roles of activity-dependent and activity-independent mechanisms in establishing functionally discrete sublaminae in the inner retina, where circuits involving many subtypes of retinal neurons are assembled precisely. In addition, we will discuss new optical imaging approaches for elucidating how retinal synaptic lamination occurs in vivo.

Cellular mechanisms underlying the precision by which neurons target their synaptic partners have largely been determined based on the study of projection neurons. By contrast, little is known about how interneurons establish their local connections in vivo. Here, we investigated how developing amacrine interneurons selectively innervate the appropriate region of the synaptic neuropil in the inner retina, the inner plexiform layer (IPL). Increases (ON) and decreases (OFF) in light intensity are processed by circuits that are structurally confined to separate ON and OFF synaptic sublaminae within the IPL. Using transgenic zebrafish in which the majority of amacrine cells express fluorescent protein, we determined that the earliest amacrine-derived neuritic plexus formed between two cell populations whose somata, at maturity, resided on opposite sides of this plexus. When we followed the behavior of individual amacrine cells over time, we discovered that they exhibited distinct patterns of structural dynamics at different stages of development. During cellular migration, amacrine cells exhibited an exuberant outgrowth of neurites that was undirected. Upon reaching the forming IPL, neurites extending towards the ganglion cell layer were relatively more stable. Importantly, when an arbor first formed, it preferentially ramified in either the inner or outer IPL corresponding to the future ON and OFF sublaminae, and maintained this stratification pattern. The specificity by which ON and OFF amacrine interneurons innervate their respective sublaminae in the IPL contrasts with that observed for projection neurons in the retina and elsewhere in the central nervous system.

Zellul&auml;re Mechanismen der Pr&auml;zisions durch die Neuronen ihre synaptische Partner deiner Wahl wurden weitgehend basierend auf der Studie der Projektion Neuronen bestimmt. Im Gegensatz dazu ist wenig bekannt &uuml;ber, wie Interneuronen ihre lokalen Verbindungen, die in-vivo herzustellen. Hier untersuchten wir wie entwickelnden Amacrine Interneuronen selektiv anregen die entsprechende Region der synaptischen Neuropil der inneren Retina, die innere plexiforme Schicht (IPL). Erh&ouml;hungen (ON) und R&uuml;ckgang der Lichtintensit&auml;t (OFF) werden durch Schaltungen verarbeitet, die strukturell beschr&auml;nkt werden, um separate ON und OFF synaptischen Sublaminae innerhalb der IPL. Mit transgenen Zebrafisch, in der die Mehrheit der Amacrine Zellen fluoreszierendes Protein Ausdr&uuml;cken, haben wir festgestellt, dass die fr&uuml;hesten Amacrine abgeleitete neuritische Plexus zwischen zwei Zelle Bev&ouml;lkerungen gebildet deren Somata, am Ende der Laufzeit auf gegen&uuml;berliegenden Seiten des dieser Plexus residierte. Targeting of axons and dendrites to particular synaptic laminae is an important mechanism by which precise patterns of neuronal connectivity are established. Although axons target specific laminae during development, dendritic lamination has been thought to occur largely by pruning of inappropriately placed arbors. We discovered by in vivo time-lapse imaging that retinal ganglion cell (RGC) dendrites in zebrafish show growth patterns implicating dendritic targeting as a mechanism for contacting appropriate synaptic partners. Populations of RGCs labeled in transgenic animals establish distinct dendritic strata sequentially, predominantly from the inner to outer retina. Imaging individual cells over successive days confirmed that multistratified RGCs generate strata sequentially, each arbor elaborating within a specific lamina. Simultaneous imaging of RGCs and subpopulations of presynaptic amacrine interneurons revealed that RGC dendrites appear to target amacrine plexuses that had already laminated. Dendritic targeting of prepatterned afferents may thus be a novel mechanism for establishing proper synaptic connectivity.

In vivo imaging reveals dendritic targeting of laminated afferents by zebrafish retinal ganglion cells.

Epilepsy is a common disorder, typified by recurrent seizures with underlying neurological disorders or disease. Approximately one-third of patients are unresponsive to currently available therapies. Thus, a deeper understanding of the genetics and etiology of epilepsy is needed to advance the development of new therapies. Previously, treatment of zebrafish with epilepsy-inducing pharmacological agents was shown to result in a seizure-like phenotype, suggesting that fish provide a tractable model to understand the function of epilepsy-predisposing genes. Here, we report the first model of genetically linked epilepsy in zebrafish and provide an initial characterization of the behavioral and neurological phenotypes associated with morpholino (MO) knockdown of leucine-rich, glioma-inactivated 1a (lgi1a) expression. Mutations in the LGI1 gene in humans have been shown to predispose to a subtype of autosomal dominant epilepsy. Low-dose Lgi1a MO knockdown fish (morphants) appear morphologically normal but are sensitized to epilepsy-inducing drugs. High-dose Lgi1a morphants have morphological defects which persist into adult stages that are typified by smaller brains and eyes and abnormalities in tail shape, and display hyperactive swimming behaviors. Increased apoptosis was observed throughout the central nervous system of high-dose morphant fish, accounting for the size reduction of neural tissues. These observations demonstrate that zebrafish can be exploited to dissect the embryonic function(s) of genes known to predispose to seizure-like behavior in humans, and offer potential insight into the relationship between developmental neurobiological abnormalities and seizure.

Epilepsie ist eine h&auml;ufige Erkrankung, versinnbildlicht durch wiederkehrende Anf&auml;lle mit zugrunde liegenden neurologischen St&ouml;rungen oder Krankheiten. Etwa ein Drittel der Patienten sind reagieren nicht auf die derzeit verf&uuml;gbaren Therapien. Daher ist ein tieferes Verst&auml;ndnis der Genetik und &Auml;tiologie der Epilepsie erforderlich, um die Entwicklung neuer Therapien. Behandlung von Zebrafish mit Epilepsie-induzierende pharmakologische Agenten wurde zuvor eine Beschlagnahme-&auml;hnlichen Ph&auml;notyp, darauf hindeutet, dass Fisch als steuerbar Modell zum Verst&auml;ndnis der Funktion der Epilepsie-pr&auml;disponierenden Gene dienen zur Folge gezeigt. Wir berichten hier, das erste Modell der genetisch verkn&uuml;pften Epilepsie im Zebrafisch und bieten eine erste Charakterisierung von der Verhaltens- und neurologischen Ph&auml;notypen zugeordnete Morpholino (MO) Niederschlag von Leucin-reiche, Gliom-inaktiviert 1a (lgi1a) Ausdruck. Mutationen im LGI1-Gen beim Menschen nachweislich f&uuml;r ein Untertyp von autosomal dominante Epilepsie pr&auml;disponieren. Niedrig dosierte Lgi1a MO Niederschlag Fisch (Morphants) erscheinen morphologisch normalen aber Epilepsie-induzierende Medikamente sensibilisiert sind. Mutations in the LGI1 gene predispose to a hereditary epilepsy syndrome and is the first gene associated with this disease which does not encode an ion channel protein. In zebrafish, there are two paralogs of the LGI1 gene, lgi1a and lgi1b. Knockdown of lgi1a results in a seizure-like hyperactivity phenotype with associated developmental abnormalities characterized by cellular loss in the eyes and brain. We have now generated knockdown morphants for the lgi1b gene which also show developmental abnormalities but do not show a seizure-like behavior. Instead, the most striking phenotype involves significant enlargement of the ventricles (hydrocephalus). As shown for the lgi1a morphants, however, lgi1b morphants are also sensitized to PTZ-induced hyperactivity. The different phenotypes between the two lgi1 morphants support a subfunctionalization model for the two paralogs.

Reporter-based assays underlie many high-throughput screening (HTS) platforms, but most are limited to in vitro applications. Here, we report a simple whole-organism HTS method for quantifying changes in reporter intensity in individual zebrafish over time termed, Automated Reporter Quantification in vivo (ARQiv). ARQiv differs from current "high-content" (e.g., confocal imaging-based) whole-organism screening technologies by providing a purely quantitative data acquisition approach that affords marked improvements in throughput. ARQiv uses a fluorescence microplate reader with specific detection functionalities necessary for robust quantification of reporter signals in vivo. This approach is: 1) Rapid; achieving true HTS capacities (i.e., >50,000 units per day), 2) Reproducible; attaining HTS-compatible assay quality (i.e., Z'-factors of â‰¥0.5), and 3) Flexible; amenable to nearly any reporter-based assay in zebrafish embryos, larvae, or juveniles. ARQiv is used here to quantify changes in: 1) Cell number; loss and regeneration of two different fluorescently tagged cell types (pancreatic beta cells and rod photoreceptors), 2) Cell signaling; relative activity of a transgenic Notch-signaling reporter, and 3) Cell metabolism; accumulation of reactive oxygen species. In summary, ARQiv is a versatile and readily accessible approach facilitating evaluation of genetic and/or chemical manipulations in living zebrafish that complements current "high-content" whole-organism screening methods by providing a first-tier in vivo HTS drug discovery platform.

Automated reporter quantification in vivo: high-throughput screening method for reporter-based assays in zebrafish.

Reporter-basierte Assays zugrunde liegen viele Hochdurchsatz-screening (HTS) Plattformen, aber die meisten sind auf in-vitro-Anwendungen beschr&auml;nkt. Hier berichten wir &uuml;ber eine einfache ganz-Organismus-HTS-Methode zur Quantifizierung von &Auml;nderungen in der Intensit&auml;t der Reporter in einzelnen Zebrafisch im Laufe der Zeit genannt, automatisierte Reporter Quantifizierung in-vivo (ARQiv).
