Retroviruses and DNA viruses utilize cellular dNTPs as substrates for their DNA polymerases during viral replication in infected cells. However, because of S phase-dependent dNTP biosynthesis, the availability of cellular dNTPs significantly varies among cell types (e.g. dividing versus nondividing cells and normal versus tumor cells). Here we tested whether alterations in the dNTP utilization efficiency and dNTP binding affinity of viral DNA polymerases can switch viral infection specificity to cell types with different dNTP concentrations. We employed an HIV-1 reverse transcriptase (RT) mutant (Q151N), which is catalytically active only at high dNTP concentrations because of its reduced dNTP binding affinity. Indeed, the modified HIV-1 vector harboring the Q151N mutant RT preferentially transduced tumor cells containing higher cellular dNTP concentrations than primary cells (e.g. human lung fibroblasts (HLFs) and human keratinocytes). Although the wild type HIV-1 vector transduced both HLFs and tumor cells, the Q151N vector failed to transduce HLFs and keratinocytes but efficiently transduced tumor cells. Pretreatment of HLFs with deoxynucleosides, which increase cellular dNTP pools, enabled the mutant vector to transduce HLFs, suggesting that the transduction failure of the RT mutant vector to primary cells is because of inefficient reverse transcription in low cellular dNTP environments. We also observed that the Q151N vector expressing herpes simplex virus-thymidine kinase renders tumor cells sensitive to gancyclovir. This study validates a novel strategy in which modifications of viral DNA polymerases in various vector systems allow the delivery of target genes exclusively to tumor cells exploiting elevated cellular dNTP concentration as a tumor cell-specific host factor.

Modification of human immunodeficiency virus type 1 reverse transcriptase to target cells with elevated cellular dNTP concentrations.

Retroviren und DNA-Viren nutzen zellul&auml;re dNTPs als Substrate f&uuml;r ihre DNA-Polymerasen w&auml;hrend der Virusreplikation in infizierten Zellen. Jedoch schwankt wegen der S-Phase-abh&auml;ngige dNTP-Biosynthese, die Verf&uuml;gbarkeit von zellul&auml;ren dNTPs unter Zelltypen (z. B. im Vergleich zu nondividing Zellen und normal gegen Tumorzellen geteilt). Hier haben wir getestet, ob &Auml;nderungen in der dNTP Auslastung Effizienz und dNTP Bindungsaffinit&auml;t der viralen DNA-Polymerasen Virusinfektion Spezifit&auml;t Zelltypen mit verschiedenen dNTP Konzentrationen aktiviert werden k&ouml;nnen. Wir Besch&auml;ftigten eine HIV-1 Reverse Transkriptase (RT) Mutant (Q151N), die aufgrund ihrer reduzierten dNTP-Bindungsaffinit&auml;t katalytisch aktiven nur bei hohen dNTP Konzentration ist. In der Tat transduzierten der modifizierte HIV-1-Vektor beherbergen die Q151N Mutant RT bevorzugt Tumorzellen mit h&ouml;here zellul&auml;re dNTP-Konzentrationen als Prim&auml;rzellen (z. B. Vorbehandlung der HLFs mit Deoxynucleosides, die zellul&auml;re dNTP Pool erh&ouml;hen, aktiviert den Mutanten Vektor zur&uuml;ckwirken HLFs, darauf hindeutet, dass die Transduktion der RT-mutant-Vektor zu Prim&auml;rzellen aufgrund ineffizienter reverse Transkription in Umgebungen mit geringen zellul&auml;ren dNTP ausf&auml;llt. Wir beobachten auch, dass der Q151N-Vektor Ausdr&uuml;cken Herpes-Simplex-Virus-Thymidinkinase Tumorzellen empfindlich auf Gancyclovir rendert. Diese Studie best&auml;tigt eine neue Strategie, in der &Auml;nderungen der viralen DNA-Polymerasen in verschiedenen Vektor die Lieferung Zielgene ausschlie&szlig;lich an Tumorzellen Nutzung erh&ouml;hte zellul&auml;re dNTP Konzentration als Tumor zellspezifische Host Faktor erm&ouml;glichen.

The interaction of human immunodeficiency virus type 1 (HIV-1) with CD4+ T lymphocytes is well studied and typically results in virally induced cytolysis. In contrast, relatively little is known concerning the interplay between HIV-1 and microglia. Recent findings suggest that, counter-intuitively, HIV-1 infection may extend the lifespan of microglia. We developed a novel cell line model system to confirm and mechanistically study this phenomenon. We found that transduction of a human microglial cell line with an HIV-1 vector results in a powerful cytoprotective effect following apoptotic challenge. This effect was reproduced by ectopic expression of a single virus-encoded protein, Tat. Subsequent studies showed that the pro-survival effects of intracellular Tat could be attributed to activation of the PI-3-kinase (PI3K)/Akt pathway in the microglial cell line. Furthermore, we found that expression of Tat led to decreased expression of PTEN, a negative regulator of the PI-3-K pathway. Consistent with this, decreased p53 activity and increased E2F activity were observed. Based on these findings, a model of possible regulatory circuits that intracellular Tat and HIV-1 infection engage during the cytoprotective event in microglia has been suggested. We propose that the expression of Tat may enable HIV-1 infected microglia to survive throughout the course of infection, leading to persistent HIV-1 production and infection in the central nervous system.

Das Zusammenspiel von Human-Immunodeficiency-Virustyp 1 (HIV-1) mit CD4 + T-Lymphozyten ist gut untersucht und f&uuml;hrt normalerweise Viral induzierten Zytolyse. Im Gegensatz dazu ist relativ wenig bekannt &uuml;ber das Zusammenspiel zwischen HIV-1 und Mikroglia. Neuesten Erkenntnissen zufolge, divergierende, HIV-1 Infektion kann die Lebensdauer der Mikroglia zu erweitern. Entwickelten wir eine neuartige Zelle Linie Modell um zu best&auml;tigen und dieses Ph&auml;nomen mechanistisch zu studieren. Wir fanden, dass Transduktion einer menschlichen Microglial Zelle Zeile mit einer HIV-1-Vektor-Ergebnisse in einem leistungsf&auml;higen Cytoprotective-Effekt nach Apoptotic Foulspiel. Dieser Effekt wurde durch ektopische Expression eines einzigen Virus-kodierten Proteins, Tat reproduziert. Sp&auml;tere Untersuchungen zeigten, dass die pro-survival Auswirkungen der intrazellul&auml;ren Tat Aktivierung des PI-3-Kinase (PI3K) zugeschrieben werden konnten / Akt-Signalweg in der Microglial Zelle Linie. Dar&uuml;ber hinaus fanden wir, dass Ausdruck der Tat zu verringerten Ausdruck des PTEN, ein negativer Regulator der PI-3-K-Signalwegs gef&uuml;hrt. Im Einklang mit dieser, verminderte p53 T&auml;tigkeit und erh&ouml;hte E2F-Aktivit&auml;t wurden beobachtet. Auf der Grundlage dieser Ergebnisse ist ein Modell der m&ouml;glichen regulatorischen Schaltkreise, die intrazellul&auml;re Tat und HIV-1 Infektion w&auml;hrend der Veranstaltung Cytoprotective in Mikroglia engagieren vorgeschlagen worden. Wir schlagen der Ausdruck der Tat HIV-1 infizierten Mikroglia im Verlauf der Infektion, &Uuml;berleben erm&ouml;glichen kann f&uuml;hrt zu permanenten HIV-1-Produktion und Infektion in das zentrale Nervensystem.

We characterized HIV-1 reverse transcriptase (RT) variants either with or without the (-)-2',3'-deoxy-3'-thiacytidine-resistant M184I mutation isolated from a single HIV-1 infected patient. First, unlike variants with wild-type M184, M184I RT variants displayed significantly reduced DNA polymerase activity at low dNTP concentrations, which is indicative of reduced dNTP binding affinity. Second, the M184I variant displayed a approximately 10- to 13-fold reduction in dNTP binding affinity, compared with the Met-184 variant. However, the k(pol) values of these two RTs were similar. Third, unlike HIV-1 vectors with wild-type RT, the HIV-1 vector harboring M184I RT failed to transduce cell types containing low dNTP concentrations, such as human macrophage, likely due to the reduced DNA polymerization activity of the M184I RT under low cellular dNTP concentration conditions. Finally, we compared the binary complex structures of wild-type and M184I RTs. The Ile mutation at position 184 with a longer and more rigid beta-branched side chain, which was previously known to alter the RT-template interaction, also appears to deform the shape of the dNTP binding pocket. This can restrict ground state dNTP binding and lead to inefficient DNA synthesis particularly at low dNTP concentrations, ultimately contributing to viral replication failure in macrophage and instability in vivo of the M184I mutation.

Wir zeichnen sich HIV-1 Reverse Transkriptase (RT) Varianten mit oder ohne (-)-2&#39;,3&#39;-deoxy-3&#39;-thiacytidine-resistant M184I Mutation aus einem einzigen HIV-1 infizierten Patienten isoliert. Zuerst angezeigt anders als Varianten mit Wildtyp M184, M184I RT-Varianten erheblich verringerter DNA-Polymeraseaktivit&auml;t bei niedrige dNTP Konzentration, das zeugt von reduzierten dNTP Bindungsaffinit&auml;t. Zweitens die M184I-Variante eine ca. 10 - bis 13 - 8-fach-Reduzierung in dNTP Bindungsaffinit&auml;t, verglichen mit der Met-184-Variante angezeigt. Allerdings waren die k(pol) Werte von diesen zwei RTs &auml;hnlich. Drittens konnte der HIV-1-Vektor beherbergen M184I RT im Gegensatz zu HIV-1-Vektoren mit Wildtyp RT, mit niedrigen dNTP-Konzentrationen, wie menschliche Makrophagen, wahrscheinlich wegen der reduzierten DNA-Polymerisation-Aktivit&auml;t des M184I RT unter niedrigen zellul&auml;ren dNTP Konzentration Bedingungen Zelltypen zur&uuml;ckwirken. Schlie&szlig;lich, als wir die bin&auml;re komplexen Strukturen der Wildtyp und M184I RTs. MicroRNAs (miRNA) have emerged as key regulators of cell lineage differentiation and cancer. We used precursor miRNA profiling by a novel real-time QPCR method (i) to define progressive stages of endothelial cell transformation cumulating in Kaposi sarcoma (KS) and (ii) to identify specific miRNAs that serve as biomarkers for tumor progression. We were able to compare primary patient biopsies to well-established culture and mouse tumor models. Loss of mir-221 and gain of mir-15 expression demarked the transition from merely immortalized to fully tumorigenic endothelial cells. Mir-140 and Kaposi sarcoma-associated herpesvirus viral miRNAs increased linearly with the degree of transformation. Mir-24 emerged as a biomarker specific for KS.

MicroRNAs (MiRNA) entstanden als wichtige Regulatoren der &Uuml;bertragungslinie Zelldifferenzierung und Krebs. Wir verwendet Vorl&auml;ufer MiRNA Profilerstellung durch eine neuartige Echtzeit QPCR-Methode (i), progressive Phasen der Endothelzellen Transformation Kumulierung in Kaposi-Sarkom (KS) zu definieren und (Ii) bestimmte MiRNAs zu identifizieren, die als Biomarker f&uuml;r Tumor Fortschreiten dienen. Wir konnten uns prim&auml;re Patienten Biopsien etablierte Kultur und Maus-Tumor-Modelle zu vergleichen. Verlust von Mir-221 und Gewinn von Mir-15 Ausdruck gekennzeichnet den &Uuml;bergang von lediglich verewigt, voll tumorigenic Endothelzellen. Mir-140 und Kaposi Sarkom-assoziierte Herpesvirus virale MiRNAs erh&ouml;ht sich linear mit dem Grad der Transformation. Mir-24 wurde als Biomarker spezifische KS untersucht.

The acute phase of immunodeficiency virus infection plays a crucial role in determining steady-state virus load and subsequent progression of disease in both humans and nonhuman primates. The acute period is also the time when vaccine-mediated effects on host immunity are likely to exert their major effects on virus infection. Recently we developed a Monte-Carlo (MC) simulation with mathematical analysis of viral evolution during primary HIV-1 infection that enables classification of new HIV-1 infections originating from multiple versus single transmitted viral strains and the estimation of time elapsed following infection.

Their expression of oxidants, proteases, and other mediators present in granules is well documented, but their ability to produce mediators through transcription and translation after migration to an inflammatory site has been appreciated only more recently.
