Seventy-five marine bacterial strains associated with four species of sponges (Echinodictyum sp., Spongia sp., Sigmadocia fibulatus and Mycale mannarensis) were isolated from the Tuticorin coast, Gulf of Mannar region. The agar-overlay method was used to screen for antibiotic production by these strains against four bacteria, viz., Bacillus subtilis, Escherichia coli, Vibrio parahaemolyticus, and Vibrio harveyi and one fungal pathogen, viz., Candida albicans. Twenty-one per cent of the bacterial strains were found to be antibiotic producers and their activities ranged from broad spectral to species specific. A strain coded SC3 was found to be highly potent and was mass cultured. The ethyl acetate extract of the culture broth was further fractionated by reverse phase HPLC and the active fraction identified. In addition, SC3 was subjected to morphological and physiological characterization. The results of the tests showed SC3 to be a Gram-positive rod, sporulating, motile, catalase and oxidase positive. Phylogenetic analysis based on comparative analysis of sequenced 16s rRNA of the active strains indicated a preponderance of bacteria belonging to Vibrio and Bacillus genera with 95-99% sequence similarities. To our knowledge this is the first report on phylogenetic identification of antibiotic producing bacteria associated with sponges from Indian waters.

Antimicrobial activity of marine bacteria associated with sponges from the waters off the coast of South East India.

Phylogenetische Analysen basierend auf vergleichende Analyse der sequenzierten 16s rRNA der aktiven Sorten angegeben ein &Uuml;bergewicht der Bakterien Vibrio und Bacillus Gattungen mit 95-99 % Sequenz &Auml;hnlichkeiten. Unseres Wissens ist dies der erste Bericht &uuml;ber phylogenetische Kennzeichnung der Antibiotika produzierenden Bakterien, die mit Schw&auml;mmen aus indischen Gew&auml;ssern verbunden.

Five bacterial isolates were screened for resistance to organotin compound, i.e. tributyltin chloride (TBTC) up to 2 mM. The optimum pH, temperature and salinity for the growth of the isolates were found to be 7, 28 degrees C and 2.5%, respectively. The isolates were tested for survival tolerance to heavy metals (mercury, cadmium and zinc) and co-resistance to antibiotics viz. ampicillin, kanamycin, rifampicin, streptomycin, penicillin, chloramphenicol, tetracycline, nalidixic acid and neomycin. Although our earlier study reported that these five bacterial strains are of different species of Pseudomonas, our present 16S rRNA gene sequence analysis revealed that all the strains are Pseudomonas aeruginosa. One of five isolates P. aeruginosa strain 25W could grow in mineral salt medium with 2 mM of TBTC as a sole source of carbon and survive up to 5 mM of TBTC. In presence of 2 mM of TBTC there was comparable up-regulation of 45 kDa protein in the cell extract of the 25W isolate was found indicating involvement of certain enzymes in TBTC resistance.

Biodiversity of organotin resistant Pseudomonas from west coast of India.

Isolate wurden f&uuml;r &Uuml;berleben Toleranz gegen&uuml;ber Schwermetallen (Quecksilber, Cadmium und Zink) und Co-resistance gegen Antibiotika, n&auml;mlich Ampicillin, Kanamycin, Rifampicin, Streptomycin, Penicillin, Chloramphenicol, Tetracyclin, Nalidixins&auml;ure und Neomycin getestet. Obwohl unsere fr&uuml;heren Studie berichtet, dass diese f&uuml;nf Bakterienst&auml;mme verschiedene Arten von Pseudomonas sind, ergab unsere derzeitigen 16S rRNA-gen Sequenzanalyse, dass alle die St&auml;mme Pseudomonas Aeruginosa sind. Einer der f&uuml;nf Isolate p. Aeruginosa Belastungen 25W konnte Mineral Salz Medium mit 2 mM TBTC als alleinige Quelle f&uuml;r Kohlenstoff wachsen und &Uuml;berleben bis zu 5 mM TBTC. In Anwesenheit von 2 mM der TBTC gab es vergleichbare bis-Verordnung 45 kDa-Protein in der Zelle-Auszug aus der 25W Isolat fand, Einbeziehung von bestimmten Enzymen im TBTC Widerstand.

The nonsteroidal antiinflammatory drug sulindac displays chemopreventive activity in patients with familial adenomatous polyposis (FAP). Sulindac metabolites induce apoptosis in colon tumor cells, in part, by a polyamine-dependent mechanism that can be suppressed with exogenous putrescine. To determine the relevance of this mechanism in animals, we treated Apc(Min/+) mice, a model of human FAP, with sulindac alone or in combination with dietary putrescine. Sulindac increased steady-state RNA levels and enzymatic activity of the polyamine catabolic enzyme spermidine/spermine N(1)-acetyltransferase and intestinal levels of monoacetylspermidine, spermidine, and spermine in the small intestine of mice. Sulindac also decreased the activity of the biosynthetic enzyme ornithine decarboxylase but not adenosylmethionine decarboxylase (AMD). Dietary putrescine increased intestinal putrescine contents, whereas the combination of dietary putrescine and sulindac yielded the highest levels of intestinal putrescine and correlated with a statistically significant reduction in AMD enzyme activity. Dietary putrescine did not statistically significantly increase tumorigenesis, although it significantly increased the grade of adenoma dysplasia (P < 0.05). The effectiveness of sulindac to suppress intestinal carcinogenesis was partially abrogated by dietary putrescine. These data suggest that sulindac exerts at least some of its anticarcinogenic effects in mice via a polyamine-dependent mechanism. Because high concentrations of putrescine can be found in certain dietary components, it may be advantageous to restrict dietary putrescine consumption in patients undergoing treatment with sulindac.

Dietary putrescine reduces the intestinal anticarcinogenic activity of sulindac in a murine model of familial adenomatous polyposis.

Sulindac Metaboliten induzieren Apoptose in Tumorzellen von Doppelpunkt, teilweise durch einen Polyamin-abh&auml;ngige Mechanismus, der mit exogenen Putrescin unterdr&uuml;ckt werden kann. Um die Relevanz dieser Mechanismus bei Tieren zu bestimmen, behandelten wir Apc(Min/+) M&auml;use, ein Modell des menschlichen FAP mit Sulindac allein oder in Kombination mit di&auml;tetischen Putrescin. Sulindac erh&ouml;ht station&auml;ren-RNA-Level und enzymatische Aktivit&auml;t von der Polyamin katabolischen Enzym Spermidine/Spermin N 1-Acetyltransferase und intestinale Monoacetylspermidine, Spermidine und Spermin im D&uuml;nndarm von M&auml;usen. Sulindac sank auch die Aktivit&auml;t der biosynthetischen Enzym Ornithin-Dekarboxylase aber nicht Adenosylmethionine Decarboxylase (AMD). Endocytic pathways have been implicated in polyamine transport in mammalian cells, but specific mechanisms have not been described. We have shown that expression of a dominant negative (DN) form of the GTPase Dynamin, but not Eps15, diminished polyamine uptake in colon cancer cells indicating a caveolar and nonclathrin uptake mode. Polyamines co-sediment with lipid raft/caveolin-1 rich fractions, of the plasma membrane in a sucrose density gradient. Knock down of caveolin-1 significantly increased polyamine uptake. Conversely, ectopic expression of this protein resulted in diminished polyamine uptake. We also found that presence of an activated K-RAS oncogene significantly increased polyamine uptake by colon cancer cells. This effect is through an increase in caveolin-1 phosphorylation at tyrosine residue 14. Caveolin-1 is a negative regulator of caveolar endocytosis and phosphorylation in a K-RAS dependent manner leads to an increase in caveolar endocytosis. In cells expressing wild type K-RAS, addition of exogenous uPA was sufficient to stimulate caveolar endocytosis of polyamines. This effect was abrogated by the addition of a SRC kinase inhibitor. These data indicate that polyamine transport follows a dynamin-dependent and clathrin-independent endocytic uptake route, and this route is positively regulated by the oncogenic expression of K-RAS in a caveolin-1 dependent manner.

Activated K-RAS increases polyamine uptake in human colon cancer cells through modulation of caveolar endocytosis.

Diese Daten zeigen, dass Polyamin-Transport eine Dynamin-abh&auml;ngige und Clathrin-unabh&auml;ngige endocytic Aufnahme-Route folgt, und diese Route ist positiv durch Onkogene Ausdruck des K-RAS Caveolin-1 abh&auml;ngigen Weise geregelt.

Co-infections with HIV-1 and the human T leukemia virus types 1 and 2 (HTLV-1, HTLV-2) occur frequently, particularly in large metropolitan areas where injection drug use is a shared mode of transmission. Recent evidence suggests that HIV-HTLV co-infections are associated with upregulated HTLV-1/2 virus expression and disease. An in vitro model of HIV-1 and HTLV-1/2 co-infection was utilized to determine if cell free HIV-1 virions or recombinant HIV-1 Tat protein (200-1,000 ng/ml) upregulated HTLV-1/2 expression and infectivity. Exposure to HIV-1 increased the number of HTLV-1 antigen expressing cells, from 6% at baseline to 12% at 24 hr, and 20% at 120 hr (P < 0.05) post-exposure. A similar, although less robust response was observed in HTLV-2 infected cells. HIV-1 co-localized almost exclusively with HTLV-1/2 positive cells. Exposure to HIV-1 Tat protein (1,000 ng/ml) increased HTLV-1 p19 expression almost twofold by 48 hr, and cells co-stimulated with 10 nM phorbol myristate acetate (PMA) showed almost a fourfold increase over baseline. It is concluded that HIV-1 augments HTLV-1/2 infectivity in vitro. The findings also suggest a role for the HIV-1 Tat protein and PMA-inducible cellular factors, in HIV-1 induced HTLV-1/2 antigen expression.

Hochregulation von HTLV-1 und HTLV-2 Ausdruck von HIV-1 in-vitro.

Kallikreins are secreted proteases that may play a functional role and/or serve as a serum biomarker for the presence or progression of certain types of cancers. Kallikrein 6 (KLK6) has been shown to be upregulated in several types of cancers, including colon. The aims of this study were to elucidate pathways that influence KLK6 gene expression and KLK6 protein secretion in the HCT116 human colon cancer cells. Our data indicate a central role for caveolin-1 (CAV-1), the main structural protein of caveolae, in both KLK6 gene expression and protein secretion. Sucrose gradient subcellular fractionation reveals that CAV-1 and KLK6 colocalize to lipid raft domains in the plasma membrane of HCT116 cells. Furthermore, we show that CAV-1, although it does not directly interact with the KLK6 molecule, enhances KLK6 secretion from the cells. Deactivation of CAV-1, through SRC-mediated phosphorylation, decreased KLK6 secretion. We also demonstrate that, in colon cancer cells, CAV-1 increased the amount of phosphorylated AKT in cells by inhibiting the activity of the AKT-negative regulators PP1 and PP2A. This study demonstrates that proteins such as CAV-1 and AKT, which are known to be altered in colon cancer, affect KLK6 expression and KLK6 secretion.

Kallikrein-6 (KLK6) hat sich herraufreguliert in verschiedene Arten von Krebs, einschlie&szlig;lich Doppelpunkt werden gezeigt. Die Ziele dieser Studie waren Wege zu erhellen, die KLK6 Genexpression und KLK6 Protein Sekretion in den HCT116 menschlichen Doppelpunkt-Krebszellen beeinflussen. Caveolin-1-vermittelte Expression und Sekretion von Kallikrein 6 in Doppelpunkt Krebszellen.

Genetic evidence suggests that caveolin-1, an essential component of membrane caveolae, acts as a tumor promoter in some, and a tumor suppressor in other cancers. The role of caveolin-1 in colon carcinogenesis is controversial. We report here, for the first time, that caveolin-1 is transcriptionally induced in colon cancer cells in response to conditional expression of a full length adenomatous polyposis coli (APC) gene. This induction of caveolin-1 by APC is mediated by both FOXO1a, a member of the Forkhead family of transcription factor, and c-myc. The FOXO1a protein, which is increased by wild-type APC expression, induces caveolin-1 promoter-reporter activity and binds directly to a FKHR consensus binding sequence in the caveolin-1 promoter. The c-myc protein, which is reduced in the presence of wild-type APC, acts to repress caveolin-1 expression by acting at non-E-box containing elements in the caveolin-1 promoter. These data predict that caveolin-1 protein expression would be decreased early in colonic carcinogenesis, which is associated with loss of wild-type APC. Our results would be consistent with the interpretation that caveolin-1 may have tumor suppressing functions during early stages of colon carcinogenesis.

Wild-type APC regulates caveolin-1 expression in human colon adenocarcinoma cell lines via FOXO1a and C-myc.

The constituents of highly active anti-retroviral therapy (HAART) include HIV-1 protease inhibitors (HPIs) and nucleoside reverse transcriptase inhibitors (NRTIs). Endothelial cell (EC) barriers, especially the blood-brain-barrier (BBB) suppresses the entry of HAART drugs to subendothelial HIV-1 reservoirs. The ATP binding cassette (ABC) transporter family members, multidrug resistant-1 (MDR-1) and multidrug resistance-associated proteins (MRPs) can efflux both HPIs and NRTIs from intracellular compartments. Using brain derived ECs from non-human sources, previous studies suggested a dominant role for MDR-1 in HAART efflux from the BBB. However, due to species variations in ABC-transporter expression, drug-efflux functions using human brain ECs need to be investigated. Furthermore, roles of ABC-transporters in drug-efflux from systemic EC barriers need to be studied. We monitored the expression of ABC-transporters in primary human ECs obtained from brain (HBMVECs), aorta (HAECs), pulmonary-artery (HPAECs), dermal-microvessel (HDMVECs) and umbilical vein (HUVECs). Gene expression for MDR-1 and MRPs (MRP-1 to MRP-5) were analyzed by reverse transcriptase polymerase chain reaction (RT-PCR). Drug efflux functions were determined by calcein retention assays. Intracellular accumulation of both 3H-saquinavir (an HPI) and 3H-zidovudine (an NRTI) were also monitored in HAECs and HBMVECs. Both assays were carried out in presence of verapamil (20-60 microM) or MK-571 (12.5-50 microM) inhibitors of MDR-1 and MRPs, respectively in presence of verapamil or MK-571. The HBMVECs expressed higher levels of MRPs than MDR-1 and only MK-571 significantly (P<0.01) suppressed calcein efflux from these cells. However, both HAECs and HPAECs showed MDR-1 and MRP expression and calcein efflux was inhibited by both verapamil and MK-571. Both inhibitors suppressed 3H-saqubinavir efflux from HAECs, but only MK-571 suppressed saquinavir efflux from HBMVECs. In both ECs, 3H-zidovudine efflux was only suppressed by MK-571. Thus, primary human ECs, especially brain derived ECs, predominantly express MRPs and their specific inhibition may enhance HAART efficacy in subendothelial HIV-1 reservoirs.

ATP verbindliche Kassette (ABC) Transporter Familienmitglieder, erfolgt mit breiter resistent-1 (MDR-1) und erfolgt mit breiter Widerstand-assoziierte Proteine (MRPs) k&ouml;nnen Efflux HPIs und NRTIs aus intrazellul&auml;ren Depots. Mit Gehirn abgeleitet ECs aus nichtmenschlichen Quellen, fr&uuml;here Studien vorgeschlagen eine dominierende Rolle f&uuml;r MDR-1 im HAART Efflux von der BBB. Die HBMVECs ausgedr&uuml;ckt deutlich h&ouml;here MRPs als MDR-1 und nur MK-571 (P &lt; 0,01) unterdr&uuml;ckt Calcein Efflux aus diesen Zellen. Jedoch wurde sowohl die HAECs als auch die HPAECs zeigte, MDR-1 und MRP Ausdruck und Calcein Efflux durch Verapamil und MK-571 gehemmt. Beide Inhibitoren unterdr&uuml;ckt 3H-Saqubinavir Efflux von HAECs, aber nur MK-571 unterdr&uuml;ckt Saquinavir Efflux von HBMVECs. In beiden ECs war 3 H-Zidovudin Efflux von MK-571 nur unterdr&uuml;ckt. So prim&auml;ren menschlichen ECs, vor allem brain abgeleiteten ECs, &uuml;berwiegend ausdr&uuml;ckliche MRPs und ihre spezifische Hemmung HAART Wirksamkeit bei Subendothelial HIV-1-Stauseen verbessern kann.

MRP (ABCC) Transporter-vermittelten Efflux von Anti-HIV-Medikamente, Saquinavir und Zidovudin, von menschlichen Endothelzellen.

Since lectins have been proposed to function as non-self recognizing molecules, it is important to access the specificity and diversity of lectin molecules that play an important role as pattern recognition receptors (PRRs). Like vertebrates, several lectins containing fibrinogen-like (FBG) domains have been reported as PRRs from different invertebrate species, however, in case of mosquito, very little information is available. In this study, we report identification and characterization of a novel family of fibrinogen-related proteins which exhibits (43-68%) similarities to that of A. gambiae FREPs, isolated from the immune challenged cell line of mosquito Aedes albopictus. Analysis of the longest (790bp) cDNA (AF-1) appears to encode a 263 deduced amino acid sequence that contains a C-terminal fibrinogen-like domain. Amino acid sequence similarity and phylogenetic analysis showed a high degree of conservation with other known insect fibrinogen-related proteins, suggesting that mosquito A. albopictus FREPs may play a similar role in an innate immunity as pattern recognition receptor molecules; however, further characterization is needed to understand their transcriptional regulation and behavior during microbial infection.

Activating mutations of FGFRs1-3 cause craniosynostosis (CS), the premature fusion of cranial bones, in man and mouse. The mechanisms by which such mutations lead to CS have been variously ascribed to increased osteoblast proliferation, differentiation, and apoptosis, but it is not always clear how these disturbances relate to the process of suture fusion. We have reassessed coronal suture fusion in an Apert Fgfr2 (S252W) mouse model. We find that the critical event of CS is the early loss of basal sutural mesenchyme as the osteogenic fronts, expressing activated Fgfr2, unite to form a contiguous skeletogenic membrane. A mild increase in osteoprogenitor proliferation precedes but does not accompany this event, and apoptosis is insignificant. On the other hand, the more apical coronal suture initially forms appropriately but then undergoes fusion, albeit at a slower rate, accompanied by a significant decrease in osteoprogenitor proliferation, and increased osteoblast maturation. Apoptosis now accompanies fusion, but is restricted to bone fronts in contact with one another. We correlated these in vivo observations with the intrinsic effects of the activated Fgfr2 S252W mutation in primary osteoblasts in culture, which show an increased capacity for both proliferation and differentiation. Our studies suggest that the major determinant of Fgfr2-induced craniosynostosis is the failure to respond to signals that would halt the recruitment or the advancement of osteoprogenitor cells at the sites where sutures should normally form.

Early onset of craniosynostosis in an Apert mouse model reveals critical features of this pathology.

Aktivierende Mutationen des FGFRs1-3 verursachen Kraniosynostose (CS), die vorzeitige Verschmelzung von Sch&auml;delknochen, in Mensch und Maus. The ATP binding cassette (ABC)-transporters are energy dependent efflux pumps which regulate the pharmacokinetics of both anti-cancer chemotherapeutic agents, e.g. taxol, and of human immunodeficiency virus-1 (HIV-1) protease inhibitors (HPIs), e.g. saquinavir. Increased expression of several ABC-transporters, especially P-glycoprotein (P-gp) and multidrug resistance protein 2 (MRP2), are observed in multidrug resistant (MDR) tumor cells and on HIV-1 infected lymphocytes. In addition, due to their apical expression on vascular endothelial barriers, both P-gp and MRP2 are of crucial importance towards dictating drug access into sequestered tissues. However, although a number of P-gp inhibitors are currently in clinical trials, possible inhibitors of MRP2 are not being thoroughly investigated. The experimental leukotriene receptor antagonist (LTRA), MK-571 is known to be a potent inhibitor of MRP transporters. Using the MRP2 over-expressing Madin-Darby canine kidney cell line, MDCKII-MRP2, we evaluated whether the clinically approved LTRAs, e.g. montelukast (Singulair) and zafirlukast (Accolate), can similarly suppress MRP2-mediated efflux. We compared the efficacy of increasing concentrations (20-100 microM) of MK-571, montelukast, and zafirlukast, in suppressing the efflux of calcein-AM, a fluorescent MRP substrate, and the radiolabeled [(3)H-] drugs, taxol and saquinavir. Montelukast was the most potent inhibitor (p<0.01) of MRP2-mediated efflux of all three substrates. Montelukast also increased (p<0.01) the duration of intracellular retention of both taxol and saquinavir. More than 50% of the drugs were retained in cells even after 90 min post removal of montelukast from the medium. Our findings implicate that montelukast, a relatively safe anti-asthmatic agent, may be used as an adjunct therapy to suppress the efflux of taxol and saquinavir from MRP2 overexpressing cells.

Long-term antiretroviral therapy (ART) for human immunodeficiency virus type one (HIV-1) infection shows limitations in pharmacokinetics and biodistribution while inducing metabolic and cytotoxic aberrations. In turn, ART commonly requires complex dosing schedules and leads to the emergence of viral resistance and treatment failures. We posit that the development of nanoformulated ART could preclude such limitations and affect improved clinical outcomes. To this end, we wet-milled 20 nanoparticle formulations of crystalline indinavir, ritonavir, atazanavir, and efavirenz, collectively referred to as "nanoART," then assessed their performance using a range of physicochemical and biological tests. These tests were based on cell-nanoparticle interactions using monocyte-derived macrophages and their abilities to uptake and release nanoformulated drugs and affect viral replication. We demonstrate that physical characteristics such as particle size, surfactant coating, surface charge, and most importantly shape are predictors of cell uptake and antiretroviral efficacy. These studies bring this line of research a step closer to developing nanoART that can be used in the clinic to affect the course of HIV-1 infection.

Tissue resident mesenchymal stem cells (MSCs) are multipotent, self-renewing cells known for their differentiation potential into cells of mesenchymal lineage. The ability of single cell clones isolated from adipose tissue resident MSCs (ASCs) to differentiate into cells of hematopoietic lineage has been previously demonstrated. In the present study, we investigated if the hematopoietic differentiated (HD) cells derived from ASCs could productively be infected with HIV-1.

Mesenchymal stem cell derived hematopoietic cells are permissive to HIV-1 infection.

Nanoformulations of crystalline indinavir, ritonavir, atazanavir, and efavirenz were manufactured by wet milling, homogenization or sonication with a variety of excipients. The chemical, biological, immune, virological, and toxicological properties of these formulations were compared using an established monocyte-derived macrophage scoring indicator system. Measurements of drug uptake, retention, release, and antiretroviral activity demonstrated differences amongst preparation methods. Interestingly, for drug cell targeting and antiretroviral responses the most significant difference among the particles was the drug itself. We posit that the choice of drug and formulation composition may ultimately affect clinical utility.

Comparative manufacture and cell-based delivery of antiretroviral nanoformulations.

Nanoformulierungen kristalline Indinavir, Ritonavir, Atazanavir und Efavirenz waren von nassen Mahlen, Homogenisierung oder Sonorisierung mit einer Vielzahl von Hilfsstoffen hergestellt. Die chemischen, biologischen, immun, virologischen und toxikologischen Eigenschaften dieser Formulierungen wurden verglichen mit einer etablierten Monozyte abgeleiteten Makrophagen Indikator Bewertungssystem.
