We present a rapid and non-photolithographic approach to microfluidic pattern generation by leveraging the inherent shrinkage properties of biaxially oriented polystyrene thermoplastic sheets. This novel approach yields channels deep enough for mammalian cell assays, with demonstrated heights up to 80 microm. Moreover, we can consistently and easily achieve rounded channels, multi-height channels, and channels as thin as 65 microm in width. Finally, we demonstrate the utility of this simple microfabrication approach by fabricating a functional gradient generator. The whole process--from device design conception to working device--can be completed within minutes.

Wir pr&auml;sentieren eine rasche und nicht-fotolithografische Vorgehensweise mikrofluidische Mustererzeugung durch Nutzung die inh&auml;rente Schrumpf-Eigenschaften der Biaxial orientierte Polystyrol thermoplastische Sheets. Dieser neuartige Ansatz ergibt Kan&auml;le tief genug f&uuml;r S&auml;ugetier-Zellkultur-Assays mit nachgewiesenen Hubh&ouml;hen bis 80 Microm. Dar&uuml;ber hinaus k&ouml;nnen wir konsequent und einfach erreichen abgerundeten Kan&auml;le, Plattenset Kan&auml;le und Kan&auml;le als d&uuml;nne als 65 Microm in der Breite. Schlie&szlig;lich zeigen wir die N&uuml;tzlichkeit dieser einfachen Microfabrication Ansatz durch Herstellung eines funktionalen Gradienten Generators. Der gesamte Prozess--von Ger&auml;te-Design-Konzeption, funktionierendes Ger&auml;t--kann innerhalb weniger Minuten abgeschlossen werden.

Shrinky Dink Biochips: schnelles Erzeugung von tiefen und abgerundeten Muster.

Embryoid body (EB) formation closely recapitulates early embryonic development with respect to lineage commitment. Because it is greatly affected by cell-cell and cell-substrate interactions, the ability to control the initial number of cells in the aggregates and to provide an appropriate substrate are crucial parameters for uniform EB formation. Here we report of an ultra-rapid fabrication and culture method utilizing a laser-jet printer to generate closely arrayed honeycomb microwells of tunable sizes for the induction of uniform EBs from single cell suspension. By printing various microwell patterns onto pre-stressed polystyrene sheets, and through heat induced shrinking, high aspect micromolds are generated. Notably, we achieve rounded bottom polydimethylsiloxane (PDMS) wells not easily achievable with standard microfabrication methods, but critical to achieve spherical EBs. Furthermore, by simply controlling the size of the microwells and the concentration of the cell suspension we can control the initial size of the cell aggregate, thus influencing lineage commitment. In addition, these microwells are easily adaptable and scalable to most standard well plates and easily integrated into commercial liquid handling systems to provide an inexpensive and easy high throughput compound screening platform.

Hier berichten wir einer Vorbereiter Fabrikation und Kultur-Methode unter Verwendung eines Laser-Jet-Druckers, eng zu generieren die Waben Inkubation einstellbare Gr&ouml;&szlig;en f&uuml;r die Induktion von einheitlichen EBs aus einzelnen Zellsuspension angetreten. Durch Drucken verschiedene Microwell-Muster auf vorgespannten Polystyrol-Platten und durch Hitze induzierte schrumpfen, werden hohe Aspekt Micromolds generiert. Bemerkenswert ist, erreichen wir abgerundeten unteren Polydimethylsiloxan (PDMS) Brunnen nicht so einfach umsetzbar sind mit standard Microfabrication Methoden, aber entscheidend f&uuml;r sph&auml;rische EBs zu erreichen. Einfach zu steuern, die Gr&ouml;&szlig;e der die Vertiefungen und die Konzentration der Zellsuspension k&ouml;nnen wir dar&uuml;ber hinaus die Anfangsgr&ouml;&szlig;e des Geldmengenaggregats Zelle steuern somit beeinflusst Linie Engagement. Dar&uuml;ber hinaus sind diese Vertiefungen, leicht anpassbar und skalierbar, um die meisten standard-well-Platten und leicht integriert in kommerziellen Liquid handling Systemen um eine billig und einfach Hochdurchsatz-Verbindungs-Screening-Plattform anzubieten.

As advances in microfluidics continue to make contributions to diagnostics and life sciences, broader awareness of this expanding field becomes necessary. By leveraging low-cost microfabrication techniques that require no capital equipment or infrastructure, simple, accessible, and effective educational modules can be made available for a broad range of educational needs from middle school demonstrations to college laboratory classes. These modules demonstrate key microfluidic concepts such as diffusion and separation as well as "laboratory on-chip" applications including chemical reactions and biological assays. These modules are intended to provide an interdisciplinary hands-on experience, including chip design, fabrication of functional devices, and experiments at the microscale. Consequently, students will be able to conceptualize physics at small scales, gain experience in computer-aided design and microfabrication, and perform experiments-all in the context of addressing real-world challenges by making their own lab-on-chip devices.

Fortschritte in der Mikrofluidik weiterhin Beitr&auml;ge zu Diagnosen und Biowissenschaften machen, wird breiteres Bewusstsein dieses expandierenden Feldes notwendig. Durch Nutzung von kosteng&uuml;nstigen Microfabrication k&ouml;nnen Techniken, die keine Hauptstadt Ausr&uuml;stung und Infrastruktur, einfache, zug&auml;ngliche und effektive p&auml;dagogische Module erfordern f&uuml;r eine Vielzahl von p&auml;dagogischen Bed&uuml;rfnissen von Mittelschule Demonstrationen auf College-Labor-Klassen verf&uuml;gbar gemacht werden. Diese Module veranschaulichen wichtige mikrofluidische Konzepte wie Verbreitung sowie Trennung und &quot;Labor auf dem Chip&quot;-Anwendungen, einschlie&szlig;lich chemische Reaktionen und biologische Proben. Diese Module sollen eine interdisziplin&auml;re praktische Erfahrung, einschlie&szlig;lich Chipdesign, Herstellung von funktionalen Ger&auml;te und Experimente in der Microscale bilden. Studenten werden folglich Physik auf kleinen Skalen zu konzipieren, sammeln Erfahrungen in CAD und Microfabrication und Experimente-alle im Zusammenhang mit realen Herausforderungen durch eine eigene Lab-on-Chip-Ger&auml;te durchf&uuml;hren k&ouml;nnen.
