A major drawback of current approaches to antiangiogenic gene therapy is the lack of tissue-specific targeting. The aim of this work was to trigger endothelial cell-specific apoptosis, using adenoviral vector-mediated delivery of a chimeric death receptor derived from the modified endothelium-specific pre-proendothelin-1 (PPE-1) promoter. In the present study, we constructed an adenovirus-based vector that targets tumor angiogenesis. Transcriptional control was achieved by use of a modified endothelium-specific promoter. Expression of a chimeric death receptor, composed of Fas and TNF receptor 1, resulted in specific apoptosis of endothelial cells in vitro and sensitization of cells to the proapoptotic effect of TNF-alpha. The antitumoral activity of the vectors was assayed in two mouse models. In the model of B16 melanoma, a single systemic injection of virus to the tail vein caused growth retardation of tumor and reduction of tumor mass with central tumor necrosis. When the Lewis lung carcinoma lung-metastasis model was applied, i.v. injection of vector resulted in reduction of lung-metastasis mass, via an antiangiogenic mechanism. Moreover, by application of the PPE-1-based transcriptional control, a humoral immune response against the transgene was avoided. Collectively, these data provide evidence that transcriptionally controlled, angiogenesis-targeted gene therapy is feasible.

Transcription-controlled gene therapy against tumor angiogenesis.

Adenosine-to-inosine (A-to-I) RNA editing increases the complexity of the human transcriptome and is essential for maintenance of normal life in mammals. Most A-to-I substitutions occur within repetitive elements in the genome, mainly in Alu repeats. The phenomenon of A-to-I editing is far less abundant in mice, rats, chickens and flies than in humans, which correlates with the relative under-representation of Alu repeats in these non-primate genomes. Here, we review the recent results of bioinformatic and laboratory approaches that have estimated the extent of the editing phenomenon. We discuss the possible biological relevance of the editing pathway, its possible interaction with other cellular pathways that respond to double-stranded RNA and its possible contribution to the accelerated evolution of primates.

Oxygenases are a family of enzymes that dioxygenate unsaturated fatty acids, thus initiating membrane oxidation and signaling molecule synthesis. The lipoxygenases (LOs), a family of lipid-peroxidizing enzymes that induce structural and metabolic changes in the cell in a number of pathophysiological conditions, belong to the oxygenases family. This class of enzymes has several subgroups, named 5-, 8-, 12- and 15-LOs, and these LO-isoforms are capable of oxygenating arachidonic and linoleic acid. 15-LOs were reported to play an inhibitory role in tumor angiogenesis and, consequently, they slow down carcinogenesis. It has been suggested that its anti-carcinogenic effect is conferred by promoting cell differentiation and apoptosis. Using transgenic mice that over-express 15-LO-1 in endothelial cells under the regulation of the murine preproendothelin-1 promoter, we studied its effect on tumor and metastasis growth. We found that 15-LO-1 inhibited tumor and metastasis growth in the transgenic mice in two different models of cancer (mammary gland and Lewis lung carcinoma). This inhibition was concomitant with a higher number of apoptotic cells in the metastases of the transgenic mice and with a complicated network of multiple small blood vessels. This finding targets 15-LO as a new candidate in the treatment of carcinogenesis.

There are only a handful of concepts concerning cancer and carcinogenesis that are currently beyond dispute. One such dogma is the adenoma-carcinoma sequence and that a multistep accumulation of genetic alterations is required for transformation from a benign to a neoplastic tissue. The inevitable derivative of this dogma is that every invasive carcinoma is in fact a missed intraepithelial tumor, and furthermore, a late evolutionary stage in the sequence of development from a precursor lesion. Until fairly recently, high-grade serous ovarian carcinoma seemed to be one of the only known deviants of these concepts. In this article, we discuss the emergence of the fallopian tube fimbria as a field of origin for high-grade serous carcinomas and present a binary model of ovarian cancer pathogenesis that takes into consideration prior epidemiologic, morphologic, and genetic data. With the rise of the fallopian tube secretory epithelial cell as a cell of origin for high-grade pelvic serous carcinomas, the need to develop tools and model systems to characterize the biology and physiology of this cell is recognized.

New insights into the pathogenesis of serous ovarian cancer and its clinical impact.

Einer solchen Dogma der Adenom-Karzinom-Sequenz ist und dass eine multistep Anh&auml;ufung von genetischen Ver&auml;nderungen f&uuml;r die Umwandlung von einer gutartigen eine neoplastische Gewebe erforderlich ist. Die unvermeidliche Ableitung dieses Dogma ist, dass jede invasive Karzinom in der Tat eine verpasste intraepitheliale Tumor, und dar&uuml;ber hinaus eine sp&auml;te Evolutionsstufe in der Reihenfolge der Entwicklung von einer Vorstufe L&auml;sion. Bis ziemlich vor kurzem hochwertige ser&ouml;se Eierstock Karzinom schien man die einzigen bekannten Diktatoren dieser Konzepte. In diesem Artikel haben wir die Entstehung der Eileiter Fimbria als Feld Ursprungs f&uuml;r hochwertige ser&ouml;se Karzinome zu diskutieren und pr&auml;sentieren ein bin&auml;res Modell des Eierstockkrebs-Pathogenese, die &Uuml;berlegung vorherige epidemiologische, morphologische und genetische Daten ber&uuml;cksichtigt. High-grade serous ovarian carcinoma (HGSOC) is a lethal disease for which improved screening and treatment strategies are urgently needed. Progress in these areas is impeded by our poor understanding of HGSOC pathogenesis. Most ovarian cancer research is based on the hypothesis that HGSOC arises from ovarian surface epithelial cells. However, recent studies suggest that >50% of high-grade serous carcinomas involving the ovary likely arise from fallopian tube epithelium. Therefore, limiting HGSOC research to modeling based on ovarian surface epithelium alone is inadequate. To address the need for a fallopian tube-based model of HGSOC, we have developed a system for studying human fallopian tube secretory epithelial cell (FTSEC) transformation. Our model is based on (i) immortalization of FTSECs isolated from primary samples of normal, nondiseased human fallopian tubes, (ii) transformation of FTSECs with defined genetic elements, and (iii) xenograft-based tumorigenic assays. We use our model to show that FTSECs immortalized with human telomerase reverse transcriptase (hTERT) plus SV40 large T and small T antigens are transformed by either oncogenic Ras (H-Ras(V12)) or c-Myc expression, leading to increased proliferation, clonogenicity, and anchorage-independent growth. Additionally, we demonstrate that FTSECs remain susceptible to c-Myc-mediated transformation in the absence of viral oncoproteins, by replacing SV40 large T and small T antigens with sh-p53, mutant CDK4 (CDK4(R24C)), and sh-PP2A-B56Î³. Importantly, all transformed FTSECs gave rise to high-grade MÃ¼llerian carcinomas that were grossly, histologically, immunophenotypically, and genomically similar to human HGSOC. With this model, we will now be able to assess the transformative effects of specific genetic alterations on FTSECs in order to characterize their respective roles in HGSOC development.

Modeling high-grade serous ovarian carcinogenesis from the fallopian tube.

Hochwertige ser&ouml;se Eierstock-Karzinom (HGSOC) ist eine t&ouml;dliche Krankheit, die f&uuml;r die verbesserte Fr&uuml;herkennung, die und Behandlung Strategien dringend gebraucht werden. Fortschritte in diesen Bereichen ist durch unsere Arme Verst&auml;ndnis der Pathogenese der HGSOC behindert. Die meisten Eierstockkrebs-Forschung basiert auf der Hypothese, da&szlig; HGSOC aus ovarielle Oberfl&auml;che Epithelzellen ergibt. Neuere Studien legen jedoch nahe, die &gt; 50 % der hochwertigen ser&ouml;se Karzinome, die mit dem Eierst&ouml;cke wahrscheinlich ergeben sich aus der Eileiter Epithel. Deshalb begrenzen HGSOC Forschung zur Modellierung basierend auf ovarielle Oberfl&auml;che Epithel allein nicht ausreichend. Um die Notwendigkeit f&uuml;r ein Eileiter basierendes Modell des HGSOC zu begegnen, haben wir ein System f&uuml;r das Studium der menschlichen Eileiter sekretorische Epithelzellen (FTSEC) Transformation entwickelt. Unser Modell basiert auf a Immortalization des FTSECs von Prim&auml;rproben normal, nondiseased menschlichen Eileiter, isoliert (Ii) die Umwandlung der FTSECs mit definierten genetische Elemente und (Iii) Xenograft--basierte tumorigenic Assays. Wir nutzen unser Modell um zu zeigen, dass FTSECs verewigt mit menschlichen Telomerase Reverse Transkriptase (hTERT) zuz&uuml;glich SV40 gro&szlig;en T und kleine T-Antigene werden transformiert von beiden Onkogene Ras (H-Ras(V12)) oder c-Myc Ausdruck, zu Verbreitung, Clonogenicity und Ankerplatz-unabh&auml;ngigen Wachstum f&uuml;hrt. Dar&uuml;ber hinaus zeigen wir, dass FTSECs anf&auml;llig f&uuml;r c-Myc-vermittelten Transformation in Ermangelung des viralen Oncoproteinen bleiben, durch das Ersetzen SV40 gro&szlig;en T und kleine T-Antigene mit sh-p53, Mutant CDK4 (CDK4(R24C)) und sh-PP2A-B56&gamma;. Wichtig ist, f&uuml;hrte alle transformierten FTSECs zu hochwertigen M&uuml;ller-Karzinome, die waren grob, histologisch, Immunophenotypically und genomically &auml;hnlich dem menschlichen HGSOC. Bei diesem Modell werden wir nun in der Lage, die Transformationswirkung des spezifischen genetischen Ver&auml;nderungen auf FTSECs zu beurteilen, um ihre jeweilige Rolle in der Entwicklung der HGSOC zu charakterisieren.

Most high-grade pelvic serous carcinomas (HGPSCs) arise from fallopian tube epithelium (FTE). To date, few markers have been shown to characterize FTE transformation. Stathmin 1 (STMN1) is a candidate oncogene whose activity is influenced by p53, p27Kip1 (p27), and PI3K/Akt pathway activation. As a microtubule destabilizing protein, STMN1 regulates cytoskeletal dynamics, cell cycle progression, mitosis, and cell migration. This study examines the expression of STMN1 and its negative regulator p27 along the morphologic continuum from normal FTE to invasive carcinoma.

Stathmin 1, a marker of PI3K pathway activation and regulator of microtubule dynamics, is expressed in early pelvic serous carcinomas.
