Malaria continues to be a devastating parasitic disease that causes the death of 2 million individuals annually. The increase in multi-drug resistance together with the absence of an efficient vaccine hastens the need for speedy and comprehensive antimalarial drug discovery and development. Throughout history, traditional herbal remedies or natural products have been a reliable source of antimalarial agents, e.g. quinine and artemisinin. Today, one emerging source of small molecule drug leads is the world's oceans. Included among the source of marine natural products are marine microorganisms such as the recently described actinomycete. Members of the genus Salinispora have yielded a wealth of new secondary metabolites including salinosporamide A, a molecule currently advancing through clinical trials as an anticancer agent. Because of the biological activity of metabolites being isolated from marine microorganisms, our group became interested in exploring the potential efficacy of these compounds against the malaria parasite.

Im Laufe der Geschichte wurden traditionelle pflanzliche Heilmittel oder nat&uuml;rliche Produkte eine zuverl&auml;ssige Quelle f&uuml;r Malariamedikamente Agenten, z. B. Chinin und Artemisinin. Heute ist einer, den aufstrebenden Quelle der kleinen Molek&uuml;l-Drogen f&uuml;hrt die Weltmeere. Unter der Quelle der marine Naturprodukte enthalten sind marine Mikroorganismen wie der k&uuml;rzlich beschriebene Actinomycete. Arten der Gattung Salinispora haben eine F&uuml;lle von neuen Sekund&auml;rmetaboliten einschlie&szlig;lich Salinosporamid A, ein Molek&uuml;l, das derzeit voran durch klinische Tests als Anti-Krebs-Agent gef&uuml;hrt. Weil die biologische Aktivit&auml;t von Metaboliten von marine Mikroorganismen isoliert interessierte sich unsere Fraktion in der Erforschung der potenziellen Wirksamkeit dieser Substanzen gegen die Malaria-Parasiten.

The epigenetic activator Mixed lineage leukemia 1 (MLL1) is paramount for embryonic development and hematopoiesis. Here, we demonstrate that the long, noncoding RNA (lncRNA) Mistral (Mira) activates transcription of the homeotic genes Hoxa6 and Hoxa7 in mouse embryonic stem cells (mESC) by recruiting MLL1 to chromatin. The Mira gene is located in the spacer DNA region (SDR) separating Hoxa6 and Hoxa7, transcriptionally silent in mESCs, and activated by retinoic acid. Mira-mediated recruitment of MLL1 to the Mira gene triggers dynamic changes in chromosome conformation, culminating in activation of Hoxa6 and Hoxa7 transcription. Hoxa6 and Hoxa7 activate the expression of genes involved in germ layer specification during mESC differentiation in a cooperative and redundant fashion. Our results connect the lncRNA Mira with the recruitment of MLL1 to target genes and implicate lncRNAs in epigenetic activation of gene expression during vertebrate cell-fate determination.

Die epigenetische Aktivator Mixed Lineage Leuk&auml;mie 1 (MLL1) ist von gr&ouml;&szlig;ter Bedeutung f&uuml;r die embryonale Entwicklung und Blutbildung. Hier zeigen wir, dass der lange, noncoding RNA (LncRNA) Mistral (Mira) Transkription der HOM&Ouml;OTISCHE Gene Hoxa6 und Hoxa7 in M&auml;useder embryonalen Stammzellen (mESC) aktiviert durch die Rekrutierung von MLL1 gegen Chromatin. Das Mira-Gen liegt in der Trennung von Hoxa6 und Hoxa7, in mESCs, transcriptionally leisen Abstandhalter DNA Region (SDR) und durch Retinoic S&auml;ure aktiviert. Mira-vermittelten Rekrutierung von MLL1 an das Mira-gen l&ouml;st dynamische &Auml;nderungen im Chromosom Konformation, die ihren H&ouml;hepunkt in der Aktivierung der Transkription von Hoxa6 und Hoxa7. Aktivieren die Expression von Genen Germ Layer Spezifikation w&auml;hrend mESC Differenzierung in eine kooperative und redundante Weise beteiligt, Hoxa6 und Hoxa7. Analysis of the evolution of drug target genes under changing drug policy is needed to assist monitoring of Plasmodium falciparum drug resistance in the field. Here we genotype Pfcrt and Pfdmr1 of 700 isolates collected in French Guiana from 2000 (5 years after withdrawal of chloroquine) to 2008, i.e., the period when the artemether-lumefantrine combination was progressively introduced and mefloquine was abandoned. Gene sequencing showed fixation of the 7G8-type Pfcrt SMVNT resistance haplotype and near fixation of the NYCDY Pfdmr1 haplotype. Pfdmr1 gene copy number correlated with 50% inhibitory concentrations of mefloquine and halofantrine (r = 0.64 and 0.47, respectively, n = 547); its temporal changes paralleled changes in in vitro mefloquine susceptibility. However, the molecular parameters studied did not account for the regained in vitro susceptibility to chloroquine and showed a poor correlation with susceptibility to artemether, lumefantrine, or quinine. Identification of novel markers of resistance to these antimalarials is needed in this South American area.

Discordant Temporal Evolution of Pfcrt and Pfmdr1 Genotypes and Plasmodium falciparum In Vitro Drug Susceptibility to 4-Aminoquinolines after Drug Policy Change in French Guiana.

Analyse der Entwicklung der Droge Zielgene unter wechselnden Drogenpolitik ist Unterst&uuml;tzung von Plasmodium Falciparum-Resistenzen im Bereich &Uuml;berwachung erforderlich. Hier wir Genotyp Pfcrt und Pfdmr1 von 700 Isolate zusammengestellt in Franz&ouml;sisch-Guayana ab 2000 (5 Jahre nach Abzug der Chloroquin) bis 2008, d. h., der Zeit, wann die Artemether-Lumefantrin-Kombination wurde schrittweise eingef&uuml;hrt, und Mefloquin wurde aufgegeben. Gen Sequenzierung zeigte Fixierung der 7 G-8-Typ Pfcrt SMVNT Widerstand Haplotype und in der N&auml;he von Fixierung des NYCDY Pfdmr1-Haplotyp. Pfdmr1 gen Exemplarnummer mit 50 % hemmende Konzentrationen von Mefloquin und Halofantrin korreliert (R = 0,64 bzw., 0.47, n = 547); in &auml;hnlicher Weise seine zeitlichen Ver&auml;nderungen wurden &Auml;nderungen in in-vitro-Mefloquin Anf&auml;lligkeit. Jedoch studierte Molekulare Parameter die wiedergewonnene in-vitro Anf&auml;lligkeit gegen&uuml;ber Chloroquin nicht ber&uuml;cksichtigt und zeigte eine schlechte Korrelation mit Anf&auml;lligkeit f&uuml;r Artemether und Lumefantrin Chinin. Identifizierung neuartiger Marker des Widerstandes gegen diese Malariamittel ist in diesem s&uuml;damerikanischen Bereich erforderlich.
