It was long expected and recently shown that main-chain liquid crystalline elastomers (MC-LCEs) may serve as high performance soft actuators due to a coupling of their intrinsic characteristics of high, yet labile, ordering and network strain. Here, we present the synthesis of new siloxane-based smectic MC-LCEs. These new materials exhibit a unique thermomechanical behavior known as the shape memory effect, which has never been observed before in such LCEs. To achieve targeted transition temperatures required for facile actuation at low temperatures, specifically temperatures ranging from 15 to 65 degrees C, we have designed and prepared such elastomers incorporating two distinct mesogenic groups, termed 5H and 5tB, coupled with hydride-terminated poly(dimethylsiloxane) spacers.

We performed molecular dynamics simulations of the electrostatic assembly of multilayers of flexible polyelectrolytes at a charged surface. The multilayer build-up was achieved through sequential adsorption of oppositely charged polymers in a layer-by-layer fashion from dilute polyelectrolyte solutions. The steady-state multilayer growth proceeds through a charge reversal of the adsorbed polymeric film which leads to a linear increase in the polymer surface coverage after completion of the first few deposition steps. Moreover, substantial intermixing between chains adsorbed during different deposition steps is observed. This intermixing is consistent with the observed requirement for several deposition steps to transpire for completion of a single layer. However, despite chain intermixing, there are almost perfect periodic oscillations of the density difference between monomers belonging to positively and negatively charged macromolecules in the adsorbed film. Weakly charged chains show higher polymer surface coverage than strongly charged ones.

Molecular dynamics simulations of layer-by-layer assembly of polyelectrolytes at charged surfaces: effects of chain degree of polymerization and fraction of charged monomers.

We report how the placement of nucleobase units, thymine, or N6-(4-methoxybenzoyl)adenine, onto the ends of a mesogenic core, bis-4-alkoxy-substituted bis(phenylethynyl)benzene, affects the properties of these materials. We show that addition of these bulky polar groups significantly reduces the range of liquid-crystalline behavior of these compounds. However, mixing two complementary nucleobase-containing AA- and BB-type monomer units together does result in the formation of stable, thermotropic liquid-crystalline (LC) phases. Hydrogen bonding is shown to play an important role in the formation of these LC phases, consistent with the formation of oligomeric or polymeric hydrogen-bonded aggregates. X-ray analyses of these mixed materials are consistent with the formation of smectic C phases.

The effect of the strength of electrostatic and short-range interactions on the multilayer assembly of oppositely charged polyelectrolytes at a charged substrate was studied by molecular dynamics simulations. The multilayer buildup was achieved through sequential adsorption of charged polymers in a layer-by-layer fashion from dilute polyelectrolyte solutions. The strong electrostatic attraction between oppositely charged polyelectrolytes at each deposition step is a driving force behind the multilayer growth. Our simulations have shown that a charge reversal after each deposition step is critical for steady multilayer growth and that there is a linear increase in polymer surface coverage after the first few deposition steps. Furthermore, there is substantial intermixing between chains adsorbed during different deposition steps. We show that the polymer surface coverage and multilayer structure are each strongly influenced by the strength of electrostatic and short-range interactions.

Polyelectrolyte spin assembly (PSA) of multilayers is a sequential process featuring adsorption of oppositely charged polyelectrolytes from dilute solutions undergoing spin-coating flow. Here, we report on the dependence of PSA multilayer buildup of poly(sodium 4-styrenesulfonate) and poly(allylamine hydrochloride) on solution ionic strength and spin speed. We observed that at a given spin speed, the PSA coating growth rate (thickness/bilayer) and polymer surface coverage shows a nonmonotonic dependence on salt concentration, first increasing and then decreasing with increasing solution ionic strength. This is argued to be a manifestation of two competing mechanisms responsible for the layer formation. At low salt concentrations, the electrostatic interactions control the multilayer assembly process, while at high salt concentrations it is dominated by shear flow. We explain this nonmonotonic behavior in the framework of a Flory-like theory of multilayer formation from polyelectrolyte solution under shear flow. Additionally, the PSA process led to multilayer coatings with a radial dependence on thickness at lower spin speed in the shear-dominated regime. On increasing spin speed, such radial dependence subsided, eventually leading to uniform coatings by planarization. The surface topography of the multilayered coatings adsorbed at salt concentration less than 0.1 M was flat and featureless for all studied spin speeds. Unique morphological features in the films were formed at salt concentration higher than 0.1 M, the size of which depended on the spin speed and solution ionic strength.

Polypeptides such as polylysine have been shown to catalyze the condensation and direct the structure of silica from precursor solutions under ambient conditions. Several of the reaction parameters have been shown to mediate this activity. Specifically, mechanical perturbation seems to play a role in the formation of hierarchical structures. Most studies have been conducted in solution, but biomedical and particularly dental applications will likely require control of biosilicified coatings, films or particle formation on surfaces. Tetraethylorthosilicate was reacted with polylysine and then spin coated onto a surface. The process parameters catalyst structure, pH, buffer: ethanol ratio and percentage of cocatalyst polyethyleneimine were varied to determine their effects on the formed silica. The chemical nature and morphology of the silica were investigated with FTIR and SEM, respectively and reaction rates were monitored with a colorimetric assay. Our results show that these process parameters had only minor effects on composition, but the catalyst conformation influenced the degree of hydration while the pH, choice of solvent and cocatalyst strongly influenced morphology. We also found that perturbation from spin coating significantly influences the silicification dynamics. The ability to catalyze nano- to micron-sized mineral with different morphologies using polypeptides could have numerous dental applications including, sealing of dentin tubules, in situ reinforcement of resin interfaces or preparation of implant surfaces.

Polypeptid-katalysierte Kiesels&auml;ure f&uuml;r Zahnmedizinische Anwendungen.

A new hybrid thermoplastic polyurethane (TPU) system that incorporates an organic, biodegradable poly(D, L-lactide) soft block with a hard block bearing the inorganic polyhedral oligosilsesquioxane (POSS) moiety is introduced and studied. Changes in the polyol composition made through variation of the hydrophilic initiator molecular weight show direct control of the final transition temperatures. Incorporating POSS into the hard segments allows for excellent elasticity above T(g), as evidenced with dynamic mechanical analysis, not seen in most other biodegradable materials. This elasticity is attributed to physical cross-links formed in the hard block through POSS crystallization, as revealed with wide-angle X-ray diffraction. Increasing the POSS incorporation level in the TPU hard block was observed to increase crystallinity and also the rigidity of the material. The highest incorporation, using a statistical average of three POSS units per hard block, demonstrated one-way shape memory with excellent shape fixing capabilities. In vitro degradation of this sample was also investigated during a two month period. Moderate water uptake and dramatic molecular weight decrease were immediately observed although large mass loss (approximately 20 wt %) was not observed until the two month time point.

Biodegradable thermoplastic polyurethanes incorporating polyhedral oligosilsesquioxane.

Biologisch abbaubaren Thermoplastischen Polyurethanen polyedrischen Oligosilsesquioxane einbinden.

Highly adjustable and precisely controllable drug release from a biodegradable stent coating was achieved using a unique family of nanostructured hybrid polyurethanes. These polyurethanes are polyhedral oligosilsesquioxane thermoplastic polyurethanes (POSS TPUs) featuring alternating multiblock structures formed by nanostructured hard segments of POSS and biodegradable soft segments of a polylactide/caprolactone copolymer (P(DLLA-co-CL)) incorporating polyethylene glycol (PEG) covalently. POSS aggregated to form crystals serving as physical crosslinks on the nanometer scale, while the soft segments were designed carefully to modulate the drug release rate from the POSS TPU stent coatings in PBS buffer solution, with 90% of the drug releasing from within half a day to about 90 days. In order to interpret the underlying drug release mechanisms, an approximation model capable of describing the entire drug release process was developed. This model is based on Fickian diffusional transport, but also takes into account the polymer degradation and/or swelling of the coating, depending on the dominance of the degradation/swelling behavior compared to that of the diffusion characteristics. A general methodology was utilized for statistically fitting the drug release curves from the POSS TPU stent coatings using the model. We observed that the fitted initial drug release diffusion coefficient covered more than three orders of magnitude, depending on the polymer glass transition temperature (T(g)), according to a modified Williams-Landel-Ferry (WLF) equation. In addition, two additional rate constants describing the impact of degradation and swelling on drug elution were determined and found to be consistent with independent measurements. Our results clearly show that the studied hybrid polyurethane family allows a drug release rate that is effectively manipulated through variation in polymer T(g), degradation rate, and thickness increment rate.

Tailored drug release from biodegradable stent coatings based on hybrid polyurethanes.

The need exists for biomaterials that prevent biofilm formation and associated infections. In this report, we have studied the synthesis, processing, and antimicrobial behavior of new silver-containing thermoplastic hydrogel nanofibrous webs. Thermoplastic hydrogels were synthesized from multiblock PEG-POSS polyurethanes (PEG: poly(ethylene gylcol); POSS: polyhedral oligosilsesquioxane) and electrospun into nanofibrous webs (diameter approximately 150 nm), with or without AgNO(3). The nanofibrous hydrogels exhibited unusual shrinkage during water uptake, yielding a uniquely dense structure compared to hydrogels prepared from cast films. Antimicrobial activity was examined using exposure to Escherichia coli with daily refreshment of medium and inoculation to a controlled cell density. Nanofibrous hydrogels without silver featured the most rapid and most extensive biofilm formation, while the silver-containing nanofibrous hydrogel featured outstanding biofilm resistance, with biofilm formation taking hold only after 14 days of incubation in daily refreshed bacterial cultures. We envision application of the unique antimicrobial hydrogels as wound dressings that combine sustained bactericidal properties and lack of volumetric swelling during water uptake.

Antimicrobial properties of nanostructured hydrogel webs containing silver.

In this paper, we report on the development of a new and broadly applicable strategy to produce thermally mendable polymeric materials, demonstrated with an epoxy/poly(-caprolactone) (PCL) phase-separated blend. The initially miscible blend composed of 15.5 wt % PCL undergoes polymerization-induced phase separation during cross-linking of the epoxy, yielding a "bricks and mortar" morphology wherein the epoxy phase exists as interconnected spheres (bricks) interpenetrated with a percolating PCL matrix (mortar). The fully cured material is stiff, strong, and durable. A heating-induced "bleeding" behavior was witnessed in the form of spontaneous wetting of all free surfaces by the molten PCL phase, and this bleeding is capable of repairing damage by crack-wicking and subsequent recrystallization with only minor concomitant softening during that process. The observed bleeding is attributed to volumetric thermal expansion of PCL above its melting point in excess of epoxy brick expansion, which we term differential expansive bleeding (DEB). In controlled thermal-mending experiments, heating of a cracked specimen led to PCL extrusion from the bulk to yield a liquid layer bridging the crack gap. Upon cooling, a "scar" composed of PCL crystals formed at the site of the crack, restoring a significant portion of the mechanical strength. When a moderate force was applied to assist crack closure, thermal-mending efficiencies exceeded 100%. We further observed that the DEB phenomenon enables strong and facile adhesion of the same material to itself and to a variety of materials, without any requirement for macroscopic softening or flow.

In diesem Beitrag berichten wir &uuml;ber die Entwicklung einer neuen und breit anwendbare Strategie, thermisch mendable Polymere Werkstoffe, demonstrierten mit einer epoxy/poly(-caprolactone) (PCL) Phase getrennte Mischung zu produzieren. Die anf&auml;nglich mischbar Mischung bestehend aus 15,5 Gew.-% erf&auml;hrt PCL induziert durch Polymerisation Phase, die Trennung bei der Vernetzung von Epoxy, woraus sich eine &quot;Ziegel und M&ouml;rtel&quot; Morphologie worin die Epoxy-Phase als miteinander verbundene Kugeln (Ziegel) interpenetrated mit einer durchsickernd PCL-Matrix (M&ouml;rtel) vorhanden ist. Das vollst&auml;ndig ausgeh&auml;rtete Material ist steif, stark und langlebig. Eine Heizung-induzierte &quot;blutende&quot; Verhalten wurde in Form von spontanen Befeuchten der alle freien Fl&auml;chen von der geschmolzenen PCL-Phase bezeugt, und diese Blutungen kann der Behebung von Sch&auml;den durch Sprung-Feuchtigkeitsregulierung und nachfolgende Rekristallisation mit nur geringf&uuml;gigen damit einhergehende Aufweichung w&auml;hrend dieses Vorgangs. Die beobachteten Blutung ist thermische Volumenausdehnung PCL &uuml;ber seinen Schmelzpunkt &uuml;ber Epoxy Backstein-Erweiterung zugeordnet, wir differenzielle expansive Blutungen (DEB) bezeichnen. In kontrollierten Thermal-Ausbessern-Experimente f&uuml;hrte Heizung eines gebrochenen Exemplars zu PCL-Extrudieren von der Masse, eine fl&uuml;ssige Schicht, die die Sprung-L&uuml;cke zu erbringen. Bei der K&uuml;hlung, ein &quot;Scar&quot;, komponiert von PCL-Kristallen gebildet am Standort des Risses, Wiederherstellen einen signifikanten Anteil an die mechanische Festigkeit. Wenn eine m&auml;&szlig;ige Kraft angewendet wurde, um Riss Schlie&szlig;ung zu unterst&uuml;tzen, &uuml;berschritten Thermal-Ausbessern-Effizienz 100 % haben. Polyester polyurethanes incorporating polyhedral oligosilsesquioxane (POSS) as the crystalline hard block were evaluated for biocompatibility and degradation over 24 weeks in vivo. In vitro studies were also used to predict the onset of mass loss. The molecular weight of each sample was found to decrease quickly over an 8 week period and then became constant due to the nondegrading POSS hard block. Kinetic analysis of the initial molecular weight change indicated that the degradation rate was dependent on the soft block composition. Crystallinity, melting temperature, and heat of fusion of the polyurethanes were found to increase during degradation as the amorphous polyester soft segments were hydrolyzed. The histological analysis of each polymer demonstrated rapid resolution of the acute and chronic inflammatory responses and the development of expected, normal foreign body reaction, consisting of adherent macrophages and foreign body giant cells on the surface of the polymers, and fibrous capsule formation around the polymer. No acute and/or chronic inflammation was seen after 3 weeks, indicating that the polymers in film form and biodegraded form, that is, particles, were biocompatible and did not elicit inflammatory responses expected for toxic or nonbiocompatible materials.

In vivo kinetic degradation analysis and biocompatibility of aliphatic polyester polyurethanes.

Cell culture substrates of defined topography have emerged as powerful tools with which to investigate cell mechanobiology, but current technologies only allow passive control of substrate properties. Here we present a thermo-responsive cell culture system that uses shape memory polymer (SMP) substrates that are programmed to change surface topography during cell culture. Our hypothesis was that a shape-memory-activated change in substrate topography could be used to control cell behavior. To test this hypothesis, we embossed an initially flat SMP substrate to produce a temporary topography of parallel micron-scale grooves. After plating cells on the substrate, we triggered shape memory activation using a change in temperature tailored to be compatible with mammalian cell culture, thereby causing topographic transformation back to the original flat surface. We found that the programmed erasure of substrate topography caused a decrease in cell alignment as evidenced by an increase in angular dispersion with corresponding remodeling of the actin cytoskeleton. Cell viability remained greater than 95% before and after topography change and temperature increase. These results demonstrate control of cell behavior through shape-memory-activated topographic changes and introduce the use of active cell culture SMP substrates for investigation of mechanotransduction, cell biomechanical function, and cell soft-matter physics.

Dynamic cell behavior on shape memory polymer substrates.

Diese Ergebnisse zeigen, Kontrolle der Zelle Verhalten durch Form-Ged&auml;chtnis-aktiviert topographische Ver&auml;nderungen und stellen die Verwendung der aktiven Zelle Kultur SMP Substrate f&uuml;r Untersuchung von Mechanotransduction, Zelle biomechanische Funktion und Physik der Zelle-Weiche Materie.

Dynamische Zelle Verhalten auf Shape Memory Polymer Substraten.

Self-healing (SH) polymers are responsive polymeric materials that can repair mechanical damage such as cracks in an autonomous fashion. In most SH polymers studies reported to date, crack closure was either unaddressed or achieved by manual intervention. Here, we report a new strategy that utilizes shape memory (SM) to prepare novel SH polymers that are capable of simultaneously closing and rebonding cracks with a simple thermal trigger. This strategy, termed "shape memory assisted self-healing (SMASH)", is demonstrated in a blend system consisting of cross-linked poly(Îµ-caprolactone) network (n-PCL) with linear poly(Îµ-caprolactone) (l-PCL) interpenetrating the network, and exhibits a combination of SM response from the network component and SH capacity from the linear component. Thermomechanical analysis revealed that the thermoset, n-PCL, demonstrates reversible plasticity -a form of shape memory where large plastic deformation at room temperature is fully recoverable upon heating. This SM action assists to close any cracks formed during deformation and/or damage while l-PCL chains tackify the crack surfaces by diffusion to the free surface and ultimately across the area of damage during the same heating step as used for SM. In our study, we investigated the controlled damage and SMASH healing of blends with varying composition using tensile testing of essential work of fracture film specimens. The healing component, l-PCL used had a high M(w) (M(w) âˆ¼65k g/mol) to enable re-entanglement after diffusion across the interface while the shape memory component, n-PCL was prepared from PCL telechelic diacrylates and a tetrathiol cross-linker, yielding excellent shape memory. We found excellent self-healing of films by the SMASH mechanism, with near complete healing for l-PCL contents exceeding 25 wt %. Applications are envisioned in the area of self-healing bladders, inflated structure membranes, and architectural building envelopes.

Hier berichten wir &uuml;ber eine neue Strategie, die nutzt Formged&auml;chtnis (SM), Roman SH Polymere vorzubereiten, die gleichzeitig schlie&szlig;en und rebonding Risse mit einem einfachen thermischen Ausl&ouml;ser sind. Diese Strategie, genannt &quot;Formged&auml;chtnis unterst&uuml;tzt die Selbstheilung (SMASH)&quot;, zeigt sich in einer Mischung-System, bestehend aus vernetztem poly(&epsilon;-caprolactone) Netzwerk (n-PCL) mit linearen poly(&epsilon;-caprolactone) (l-PCL) unvorstellbarer im Netzwerk und zeigt eine Kombination von SM-Antwort aus der Netzwerkkomponente und SH-Kapazit&auml;t aus der linearen Komponente. Thermomechanische Analyse ergab, dass die duroplastischen, n-PCL, veranschaulicht umkehrbare Plastizit&auml;t - eine Form von Formged&auml;chtnis, wo gro&szlig;e plastische Verformung bei Raumtemperatur beim Erhitzen vollst&auml;ndig wiederherstellbar ist. Diese SM-Aktion hilft schlie&szlig;en Risse bildeten w&auml;hrend der Verformung und/oder Sch&auml;den w&auml;hrend l-PCL-Ketten die Sprung-Oberfl&auml;chen tackify durch Diffusion auf der freien Oberfl&auml;che und letztlich &uuml;ber den Bereich der Besch&auml;digung w&auml;hrend des gleichen Heizung Schritt als gebraucht f&uuml;r SM. In unserer Studie untersuchten wir die kontrollierte und SMASH Heilungsboni von Mischungen mit unterschiedlicher Zusammensetzung mit Zugversuche wesentliche Arbeiten der Fraktur Film Exemplare. Die heilende Komponente, l-PCL verwendet hatte eine hohe M(w) (M(w)/&sim;65k g/Mol) Re-entanglement aktivieren nach Verbreitung &uuml;ber die Schnittstelle w&auml;hrend der Shape-Speicherkomponente, n-PCL wurde vorbereitet von PCL-Telechelic-Diacrylates und ein Tetrathiol Cross-Linker, woraus ausgezeichnete Formged&auml;chtnis. Wir fanden hervorragende Selbstheilung Filme durch den SMASH-Mechanismus mit nahe vollst&auml;ndige Heilung f&uuml;r l-PCL-Inhalte, die von mehr als 25 Gew.-%. Anwendungen sind im Bereich der Selbstheilung, Blasen, aufgeblasene Struktur Membranen und architektonische Geb&auml;udeh&uuml;llen vorgestellt.

In this Article, we studied the enzymatic hydrolytic biodegradation behavior of a novel multiblock thermoplastic polyurethane (TPU) system, which incorporates polyhedral oligomeric silsesquioxane (POSS) into linear biodegradable thermoplastic polyurethanes containing poly(Îµ-caproactone) (PCL) and polyethylene glycol (PEG) blocks. The biodegradation behavior of POSS-PCL-PEG TPUs was characterized by proton nuclear magnetic resonance spectroscopy ((1)H NMR), differential scanning calorimetry (DSC), tensile tests, scanning electron microscopy (SEM), and wavelength dispersive X-ray spectrometry (WDS) after enduring 22-day accelerated enzymatic hydrolytic degradation tests. POSS incorporation significantly suppressed in vitro enzymatic hydrolytic degradation of PCL-PEG-based multiblock TPUs by a surface passivation mechanism. WDS observations revealed that the covalently bonded POSS moieties developed a near-continuous and robust POSS-layer after initial degradation, which prevented ester bonds of PCL from enzymatic attack, thereby inhibiting further degradation. These striking results provide a new strategy to fabricate the polyester-based biostable thermoplastic polyurethanes (TPUs) of potential use in long-term surgical implants.

WDS Beobachtungen ergeben, dass die kovalent gebundenen POSS Adsorptionseigenschaften eine nahezu kontinuierliche und robuste POSS-Schicht nach anf&auml;nglichen Verschlechterung,, die Ester Anleihen PCL von enzymatischen Angriff entwickelt, was wiederum behindert weitere Verschlechterung zu verhindern. Diese auff&auml;llige Ergebnisse bieten eine neue Strategie um die Polyester-basierte Biostable Thermoplastische Polyurethane (TPU) der m&ouml;glichen Verwendung in langfristige chirurgische Implantate zu fabrizieren.
