The oncogene c-maf is frequently overexpressed in multiple myeloma cell lines and patient samples and contributes to increased cellular proliferation in part by inducing cyclin D2 expression. To identify regulators of c-maf, we developed a chemical screen in NIH3T3 cells stably overexpressing c-maf and the cyclin D2 promoter driving luciferase. From a screen of 2400 off-patent drugs and chemicals, we identified glucocorticoids as c-maf-dependent inhibitors of cyclin D2 transactivation. In multiple myeloma cell lines, glucocorticoids reduced levels of c-maf protein without influencing corresponding mRNA levels. Subsequent studies demonstrated that glucocorticoids increased ubiquitination-dependent degradation of c-maf and up-regulated ubiquitin C mRNA. Moreover, ectopic expression of ubiquitin C recapitulated the effects of glucocorticoids, demonstrating regulation of c-maf protein through the abundance of the ubiquitin substrate. Thus, using a chemical biology approach, we identified a novel mechanism of action of glucocorticoids and a novel mechanism by which levels of c-maf protein are regulated by the abundance of the ubiquitin substrate.

Das Onkogen c-Maf ist h&auml;ufig in multiplen Myelom-Zelllinien und Patientenproben Pleuramesotheliom und tr&auml;gt zur zellul&auml;ren Verbreitung teilweise durch induzieren cyclin D2-Ausdruck. Um die Regler der c-Maf zu identifizieren, entwickelten wir einen chemischen Bildschirm in NIH3T3 Zellen stabil &uuml;berexprimierenden c-Maf und der cyclin D2-Projekttr&auml;ger Luciferase fahren. Von einem Bildschirm von 2400-Patent Drogen und Chemikalien haben wir als c-Maf-abh&auml;ngigen-Inhibitoren cyclin D2 Transactivation Glukokortikoide identifiziert. Multiples Myelom-Zelllinien reduziert Glukokortikoide Niveaus des Proteins c-Maf ohne entsprechenden mRNA-Stufen zu beeinflussen. Nachfolgende Studien gezeigt, dass Glukokortikoide Ubiquitination abh&auml;ngigen Verschlechterung der c-Maf und oben-geregelt Ubiquitin C erh&ouml;hte mRNA. Dar&uuml;ber hinaus rekapitulierte ektopische Expression des Ubiquitin-C die Wirkungen der Glukokortikoide, demonstrieren Regulierung des c-Maf Protein durch die F&uuml;lle der Ubiquitin-Substrat. So wurden mit einem chemischen Biologie-Ansatz, einen neuartigen Wirkmechanismus von Glukokortikoiden und einen neuen Mechanismus, durch den Niveaus des Proteins c-Maf durch das &Uuml;berangebot an das Ubiquitin-Substrat geregelt sind.

D-cyclins are regulators of cell division that act in a complex with cyclin-dependent kinases to commit cells to a program of DNA replication. D-cyclins are overexpressed in many tumors, including multiple myeloma and leukemia, and contribute to disease progression and chemoresistance. To better understand the role and impact of D-cyclins in hematologic malignancies, we conducted a high throughput screen for inhibitors of the cyclin D2 promoter and identified the drug cyproheptadine. In myeloma and leukemia cells, cyproheptadine decreased expression of cyclins D1, D2, and D3 and arrested these cells in the G(0)/G(1) phase. After D-cyclin suppression, cyproheptadine induced apoptosis in myeloma and leukemia cell lines and primary patient samples preferentially over normal hematopoietic cells. In mouse models of myeloma and leukemia, cyproheptadine inhibited tumor growth without significant toxicity. Cyproheptadine-induced apoptosis was preceded by activation of the mitochondrial pathway of caspase activation and was independent of the drug's known activity as an H1 histamine and serotonin receptor antagonist. Thus, cyproheptadine represents a lead for a novel therapeutic agent for the treatment of malignancy. Because the drug is well tolerated and already approved in multiple countries for clinical use as an antihistamine and appetite stimulant, it could be moved directly into clinical trials for cancer.

D-Cyclins sind in vielen Tumoren, darunter Multiples Myelom und Leuk&auml;mie, Pleuramesotheliom und zum Fortschreiten der Krankheit und Chemoresistance beitragen. Zum besseren Verst&auml;ndnis der Rolle und Wirkung der D-Cyclins in hematologic Malignancies wir einen hohen Durchsatz-Bildschirm f&uuml;r Inhibitoren cyclin D2-Promotor durchgef&uuml;hrt und identifiziert das Medikament Cyproheptadin. Myelom und Leuk&auml;mie-Zellen Cyproheptadin verringerte sich um Ausdruck des Cyclins D1, D2 und D3 und diese Zellen in die G(0)/G(1)-Phase verhaftet. Nach D-cyclin-Unterdr&uuml;ckung Cyproheptadin induzierte Apoptose in Myelom und Leuk&auml;mie-Zelllinien und prim&auml;re Patientenproben bevorzugt &uuml;ber normaler h&auml;matopoetischer Zellen. The nerve-cell cytoskeleton is essential for the regulation of intrinsic neuronal activity. For example, neuronal migration defects are associated with microtubule regulators, such as LIS1 and dynein, as well as with actin regulators, including Rac GTPases and integrins, and have been thought to underlie epileptic seizures in patients with cortical malformations. However, it is plausible that post-developmental functions of specific cytoskeletal regulators contribute to the more transient nature of aberrant neuronal activity and could be masked by developmental anomalies. Accordingly, our previous results have illuminated functional roles, distinct from developmental contributions, for Caenorhabditis elegans orthologs of LIS1 and dynein in GABAergic synaptic vesicle transport. Here, we report that C. elegans with function-altering mutations in canonical Rac GTPase-signaling-pathway members demonstrated a robust behavioral response to a GABA(A) receptor antagonist, pentylenetetrazole. Rac mutants also exhibited hypersensitivity to an acetylcholinesterase inhibitor, aldicarb, uncovering deficiencies in inhibitory neurotransmission. RNA interference targeting Rac hypomorphs revealed synergistic interactions between the dynein motor complex and some, but not all, members of Rac-signaling pathways. These genetic interactions are consistent with putative Rac-dependent regulation of actin and microtubule networks and suggest that some cytoskeletal regulators cooperate to uniquely govern neuronal synchrony through dynein-mediated GABAergic vesicle transport in C. elegans.

Beispielsweise der neuronalen Migration M&auml;ngel sind verbunden mit Microtubule-Regulatoren, wie LIS1 und Dynein, sowie mit dem Aktin-Regulatoren, einschlie&szlig;lich Rac GTPases und Integrine, und haben gedacht worden, um epileptische Anf&auml;lle bei Patienten mit kortikalen Fehlbildungen zugrunde liegen. Jedoch ist es plausibel, dass post-developmental Funktionen von bestimmten Zytoskelett Regulierungsbeh&ouml;rden mehr vor&uuml;bergehender Natur aberrante neuronaler Aktivit&auml;t zur und mit Entwicklungsanomalien maskiert werden konnte. Dementsprechend haben unsere bisherigen Ergebnisse beleuchtete Funktionsrollen, entwicklungsbedingte Beitr&auml;ge f&uuml;r Caenorhabditis Elegans Orthologen LIS1 und Dynein in GABAergic synaptische Vesikel Transport unterscheidet. Wir berichten hier, dass C. Elegans mit Funktion ver&auml;ndern Mutationen im kanonischen Rac GTPase-Signalisierung-Weg-Mitglieder nachgewiesen eine robuste Verhaltens Reaktion auf einen GABA(A)-Rezeptor-Antagonisten, Pentylenetetrazole. RAC Mutanten stellte auch &Uuml;berempfindlichkeit gegen ein Acetylcholinesterase-Hemmer, Aldicarb, Aufdeckung von M&auml;ngeln in inhibitorischen Neurotransmission. We discovered a high-level amplicon involving the chr19q13.41 microRNA (miRNA) cluster (C19MC) in 11/45 ( approximately 25%) primary CNS-PNET, which results in striking overexpression of miR-517c and 520g. Constitutive expression of miR-517c or 520g promotes in vitro and in vivo oncogenicity, modulates cell survival, and robustly enhances growth of untransformed human neural stem cells (hNSCs) in part by upregulating WNT pathway signaling and restricting differentiation of hNSCs. Remarkably, the C19MC amplicon, which is very rare in other brain tumors (1/263), identifies an aggressive subgroup of CNS-PNET with distinct gene-expression profiles, characteristic histology, and dismal survival. Our data implicate miR-517c and 520g as oncogenes and promising biological markers for CNS-PNET and provide important insights into oncogenic properties of the C19MC locus.

Frequent amplification of a chr19q13.41 microRNA polycistron in aggressive primitive neuroectodermal brain tumors.

Chronic lung disease (CLD) is a major cause of long-term morbidity in extremely LBW infants with respiratory distress syndrome. Parenteral vitamin A administration decreases the risk of CLD. We tested the hypothesis that intratracheal vitamin A administration with surfactant is systemically bioavailable without interfering with the functional properties of exogenous surfactant. Newborn piglets were ventilated with 100% FiO2 and sequential saline lavage induced respiratory distress syndrome. During lung injury induction, ventilator changes were allowed, but none were made following treatment allocation. Animals were assigned by chance in a blinded control trial to three groups: I=control; II=surfactant; III=surfactant+vitamin A. Hemodynamics, lung mechanics, and blood gases were measured following instrumentation, pre- and posttreatment for 4 h, at which time the liver was sampled for retinol determination. All parameters improved in animals receiving surfactant. A significant interaction existed between time and group for PaO2 and alveolar-arterial oxygen difference (A-aDO2). Hepatic levels of retinol were higher (p<0.001) in animals receiving retinyl acetate. Intratracheal administration of surfactant+vitamin A did not alter the beneficial effects of surfactant on lung compliance and gas exchange. Intratracheal Vitamin A was associated with rapid hepatic uptake. Further studies are warranted.

Non-surgical, antiviral treatment options are desirable for HPV-related lesions within the genitourinary and upper digestive tract. We compared the toxicity of three zinc finger-ejecting (ZFE) compounds (4,4-dithiodimorpholine, azodicarbonamide, and diamide) to the HIV protease inhibitor lopinavir using HPV-positive SiHa, CaSki, HeLa, ME180, and HPV-negative C33A cervical carcinoma cell lines as well as primary human foreskin keratinocytes (PHFKs). Colorimetric growth assays revealed selective toxicity when treated with lopinavir. All carcinoma cell lines, except CaSki, were sensitive to 20 Î¼M lopinavir whereas primary PHFKs were highly resistant. In contrast, 4,4-dithiodimorpholine was uniformly toxic to all cells tested while azodicarbonamide and diamide showed no effect at all. It is concluded that lopinavir may be an attractive candidate to treat pre-cancerous and cancerous HPV-positive lesions.

Konservative, antivirale Behandlungsoptionen sind w&uuml;nschenswert f&uuml;r HPV-&auml;hnliche L&auml;sionen innerhalb der urogenitalen und oberen Verdauungstrakt. Wir verglichen die Toxizit&auml;t von drei Zink Finger-auswerfen (ZFE) Verbindungen (4,4-Dithiodimorpholine Azodicarbonamid und Diamide) auf die HIV-Protease-Inhibitor Lopinavir mit HPV-positiven SiHa, CaSki, HeLa, ME180 und HPV-negativen C33A Geb&auml;rmutterhals-Karzinom-Zelllinien sowie prim&auml;re menschliche Vorhaut Keratinozyten (PHFKs). Kolorimetrische Wachstum Proben ergab selektive Toxizit&auml;t nach Behandlung mit Lopinavir. Alle Karzinom-Zelllinien, au&szlig;er CaSki, waren empfindlich auf 20 &mu;M Lopinavir w&auml;hrend prim&auml;re PHFKs sehr resistent waren. Wird der Schluss gezogen, dass Lopinavir eine attraktive Kandidat pr&auml;kanzer&ouml;se und krebsartige HPV-positiven Hautver&auml;nderungen zu behandeln sein kann.

Lopinavir zeigt h&ouml;here Spezifit&auml;t als Zinkfinger Auswerfen Verbindungen als potenzielle Behandlung f&uuml;r menschliche Papillomavirus-&auml;hnliche L&auml;sionen.
