Chronic exposure to tobacco smoke leads to an increase in the frequency of infections and in the number of CD8(+) and CD4(+) cells as well as the CD4(+) chemoattractant cytokine IL-16 in the airways. Here, we investigated whether tobacco smoke depletes intracellular IL-16 protein and inhibits de novo production of IL-16 in CD8(+) cells from human airways and blood while increasing extracellular IL-16 and whether oxygen free radicals (OFR) are involved. Intracellular IL-16 protein in CD8(+) cells and mRNA in all cells was decreased in bronchoalveolar lavage (BAL) samples from chronic smokers. This was also the case in human blood CD8(+) cells exposed to water-soluble tobacco smoke components in vitro, in which oxidized proteins were markedly increased. Extracellular IL-16 protein was increased in cell-free BAL fluid from chronic smokers and in human blood CD8(+) cells exposed to water-soluble tobacco smoke components in vitro. This was not observed in occasional smokers after short-term exposure to tobacco smoke. A marker of activation (CD69) was slightly increased, whereas other markers of key cellular functions (membrane integrity, apoptosis, and proliferation) in human blood CD8(+) cells in vitro were negatively affected by water-soluble tobacco smoke components. An OFR scavenger prevented these effects, whereas a protein synthesis inhibitor, a Î²-adrenoceptor, a glucocorticoid receptor agonist, a phosphodiesterase, a calcineurin phosphatase, and a caspase-3 inhibitor did not. In conclusion, tobacco smoke depletes preformed intracellular IL-16 protein, inhibits its de novo synthesis, and distorts key cellular functions in human CD8(+) cells. OFR may play a key role in this context.

Effects of tobacco smoke on IL-16 in CD8+ cells from human airways and blood: a key role for oxygen free radicals?

Chronischer Exposition gegen&uuml;ber Tabakrauch f&uuml;hrt zu einer Zunahme der H&auml;ufigkeit von Infektionen und die Anzahl der Zellen CD8(+) und CD4(+) sowie die CD4(+) Chemoattractant Cytokine IL-16 in den Atemwegen. Hier wir gepr&uuml;ft, ob Tabakrauch intrazellul&auml;re IL-16-Protein ersch&ouml;pft und hemmt die de-Novo-Produktion von IL-16 in CD8(+) Zellen aus menschlichen Atemwege und Blut w&auml;hrend gleichzeitig die extrazellul&auml;re IL-16 und ob freie Sauerstoff-Radikale (OFR) beteiligt sind. Intrazellul&auml;re IL-16-Protein in den CD8(+) Zellen und mRNA in allen Zellen wurde in Bronchoalveol&auml;re Lavage (BAL) Proben von chronischen Rauchern verringert. Dies war auch der Fall in menschlichem Blut CD8(+) Zellen, wasserl&ouml;slichen Tabakrauch-Komponenten in vitro ausgesetzt, in denen oxidierte Proteine deutlich erh&ouml;ht wurden. Extrazellul&auml;re IL-16-Protein wurde in zellfreien BAL-Fl&uuml;ssigkeit aus chronischen Raucher und in menschlichem Blut CD8(+) Zellen ausgesetzt wasserl&ouml;slichen Tabakrauch-Komponenten, die in-vitro-erh&ouml;ht. Dies wurde nicht nach kurzfristigen Exposition gegen&uuml;ber Tabakrauch in gelegentliche Raucher beobachtet. Eine Markierung der Aktivierung (CD69) wurde leicht erh&ouml;ht, w&auml;hrend die anderen Markern der wichtige Zellfunktionen (Membran Integrit&auml;t, Apoptose und Proliferation) in menschlichem Blut CD8(+) Zellen in vitro durch wasserl&ouml;sliche Tabakrauch-Komponenten negativ beeinflusst wurden. Eine Schnitzeljagd OFR verhindert diese Effekte, w&auml;hrend ein Protein-Synthese-Hemmer, ein &beta;-Adrenozeptor, ein Glukokortikoid-Rezeptor-Agonist, eine Phosphodiesterase, eine Calcineurin-Phosphatase und ein Caspase-3-Hemmer nicht tat. Abschlie&szlig;end Tabakrauch ersch&ouml;pft vorgeformte intrazellul&auml;re IL-16-Protein, die de-Novo-Synthese hemmt, und verzerrt wichtige Zellfunktionen in den menschlichen Zellen in CD8(+). OFR kann in diesem Zusammenhang eine Schl&uuml;sselrolle spielen.

Exosomes are small extracellular nanovesicles of endocytic origin that mediate different signals between cells, by surface interactions and by shuttling functional RNA from one cell to another. Exosomes are released by many cells including mast cells, dendritic cells, macrophages, epithelial cells and tumour cells. Exosomes differ compared to their donor cells, not only in size, but also in their RNA, protein and lipid composition.

Exosomes sind kleine extrazellul&auml;re Nanovesicles endocytic Ursprungs, die unterschiedliche Signale zwischen Zellen, indem Oberfl&auml;che Interaktionen und pendelnden funktionalen RNA aus einer Zelle zu einer anderen zu vermitteln. Exosomes werden von vielen Zellen und Mastzellen, dendritische Zellen, Makrophagen, Epithelzellen und Tumorzellen freigegeben. Exosomes unterscheiden sich im Vergleich zu ihren Spenderzellen, nicht nur in der Gr&ouml;&szlig;e, sondern auch in ihrer Zusammensetzung RNA, Proteine und Lipide.

Exosomes are 30-100 nm membrane vesicles of endocytic origin produced by numerous cells. They can mediate diverse biological functions, including antigen presentation. Exosomes have recently been shown to contain functional RNA, which can be delivered to other cells. Exosomes may thus mediate biological functions either by surface-to-surface interactions with cells, or by the delivery of functional RNA to cells. Our aim was therefore to determine the presence of RNA in exosomes from human saliva, plasma and breast milk and whether these exosomes can be taken up by macrophages.

Sie k&ouml;nnen vielf&auml;ltige biologische Funktionen, einschlie&szlig;lich der Antigen-Pr&auml;sentation vermitteln. Exosomes wurden vor kurzem gezeigt, funktionalen RNA enthalten, die auf andere Zellen geliefert werden kann. Exosomes kann somit biologischen Funktionen durch Fl&auml;che zu Fl&auml;che Interaktionen mit Zellen oder durch die Lieferung von funktionalen RNA auf Zellen vermitteln. Unser Ziel war daher bestimmt das Vorhandensein von RNA in Exosomes von menschlichen Speichel, Plasma und Brust Milch und ob diese Exosomes durch Makrophagen aufgenommen werden k&ouml;nnen.

The field of exosome research is rapidly expanding, with a dramatic increase in publications in recent years. These small vesicles (30-100 nm) of endocytic origin were first proposed to function as a way for reticulocytes to eradicate the transferrin receptor while maturing into erythrocytes(1), and were later named exosomes. Exosomes are formed by inward budding of late endosomes, producing multivesicular bodies (MVBs), and are released into the environment by fusion of the MVBs with the plasma membrane(2). Since the first discovery of exosomes, a wide range of cells have been shown to release these vesicles. Exosomes have also been detected in several biological fluids, including plasma, nasal lavage fluid, saliva and breast milk(3-6). Furthermore, it has been demonstrated that the content and function of exosomes depends on the originating cell and the conditions under which they are produced. A variety of functions have been demonstrated for exosomes, such as induction of tolerance against allergen(7,8), eradication of established tumors in mice(9), inhibition and activation of natural killer cells(10-12), promotion of differentiation into T regulatory cells(13), stimulation of T cell proliferation(14) and induction of T cell apoptosis(15). Year 2007 we demonstrated that exosomes released from mast cells contain messenger RNA (mRNA) and microRNA (miRNA), and that the RNA can be shuttled from one cell to another via exosomes. In the recipient cells, the mRNA shuttled by exosomes was shown to be translated into protein, suggesting a regulatory function of the transferred RNA(16). Further, we have also shown that exosomes derived from cells grown under oxidative stress can induce tolerance against further stress in recipient cells and thus suggest a biological function of the exosomal shuttle RNA(17). Cell culture media and biological fluids contain a mixture of vesicles and shed fragments. A high quality isolation method for exosomes, followed by characterization and identification of the exosomes and their content, is therefore crucial to distinguish exosomes from other vesicles and particles. Here, we present a method for the isolation of exosomes from both cell culture medium and body fluids. This isolation method is based on repeated centrifugation and filtration steps, followed by a final ultracentrifugation step in which the exosomes are pelleted. Important methods to identify the exosomes and characterize the exosomal morphology and protein content are highlighted, including electron microscopy, flow cytometry and Western blot. The purification of the total exosomal RNA is based on spin column chromatography and the exosomal RNA yield and size distribution is analyzed using a Bioanalyzer.

Isolation and Characterization of RNA-Containing Exosomes.

Bereich der Exosome Forschung ist, mit einem drastischen Anstieg der Ver&ouml;ffentlichungen in den letzten Jahren rasch. Diese kleinen Bl&auml;schen (30-100 nm) endocytic Ursprungs wurden zuerst die Funktion als eine M&ouml;glichkeit f&uuml;r Retikulozyten des Transferrin-Rezeptors zu beseitigen, w&auml;hrend der Reifung in erythrocytes(1) vorgeschlagen, und wurden sp&auml;ter benannt Exosomes. Exosomes werden gebildet, indem nach innen, die Knospen der sp&auml;ten Endosomen, multivesicular Karosserien (MVBs), und durch die Fusion von MVBs mit der Plasma-membrane(2) in die Umwelt freigesetzt werden. Seit der ersten Entdeckung der Exosomes wurde eine Vielzahl von Zellen angezeigt, diese Bl&auml;schen freizugeben. Exosomes wurden auch in mehreren biologischen Fl&uuml;ssigkeiten, einschlie&szlig;lich Plasma, nasale Lavage-Fl&uuml;ssigkeit, Speichel und Brust milk(3-6) nachgewiesen. Dar&uuml;ber hinaus wurde nachgewiesen, dass die Inhalte und die Funktion des Exosomes h&auml;ngt von der urspr&uuml;nglichen Zelle und die Bedingungen, unter denen sie produziert werden. Eine Vielzahl von Funktionen f&uuml;r Exosomes, z. B. Induktion von Toleranz gegen&uuml;ber allergen(7,8), Beseitigung der etablierten Tumoren in mice(9), Hemmung und Aktivierung der nat&uuml;rlichen killer-cells(10-12), F&ouml;rderung der Differenzierung in regulatorischen T-cells(13), Stimulation der T-Zell-proliferation(14) und Induktion von T-Zell-apoptosis(15) nachgewiesen worden. Jahr 2007 wir bewiesen, dass Exosomes aus Mastzellen freigesetzt Boten-RNA (mRNA) und MicroRNA (MiRNA) enthalten und die RNA von einer Zelle zur anderen &uuml;ber Exosomes pendelte sein kann. In den Empf&auml;nger Zellen zeigte die mRNA von Exosomes pendelte in Protein &uuml;bersetzt werden vorgeschlagen eine regulatorische Funktion der &uuml;bertragenen RNA(16). Dar&uuml;ber hinaus haben wir auch gezeigt, dass Exosomes von Zellen, die unter oxidativem Stress gewachsen abgeleitet k&ouml;nnen Toleranz gegen weitere Stress im Empf&auml;nger Zellen induzieren und so eine biologische Funktion des Exosomal Shuttle RNA(17) suggerieren. Zellkulturmedien und biologischen Fl&uuml;ssigkeiten enthalten eine Mischung aus Bl&auml;schen und Schuppen Fragmente. Eine hochwertige Isolation Methode f&uuml;r Exosomes, gefolgt von Charakterisierung und Identifizierung der Exosomes und ihres Inhalts, ist daher entscheidend f&uuml;r die Exosomes von anderen Bl&auml;schen und Teilchen zu unterscheiden. Hier pr&auml;sentieren wir eine Methode f&uuml;r die Isolierung von Exosomes von Zellkulturmedium und K&ouml;rperfl&uuml;ssigkeiten. Diese Isolation-Methode basiert auf wiederholte Zentrifugierung und Filtrierung Steps, gefolgt von einer endg&uuml;ltigen Ultrazentrifugation Schritt, in denen die Exosomes pelleted sind. Wichtige Methoden zur Ermittlung der Exosomes und Charakterisierung des Exosomal Morphologie und Protein-Inhalts, einschlie&szlig;lich Elektronenmikroskopie, Durchflusszytometrie und Western-Blot, werden hervorgehoben. Die Reinigung der gesamten Exosomal RNA basiert auf Spin-S&auml;ulenchromatographie und die Exosomal RNA-Ausbeute und Gr&ouml;&szlig;e-Verteilung ist mit einem Bioanalyzer analysiert.
