A novel protein domain with dual affinity has been created by randomization and selection. The small alkali-stabilized albumin-binding domain (ABD*), used as scaffold to construct the library, has affinity to human serum albumin (HSA) and is constituted of 46 amino acids of which 11 were randomized. To achieve a dual binder, the binding site of the inherent HSA affinity was untouched and the randomization was made on the opposite side of the molecule. Despite its small size and randomization of almost a quarter of its amino acids, a bifunctional molecule, ABDz1, with ability to bind to both HSA and the Z2 domain/protein A was successfully selected using phage display. Moreover, the newly selected variant showed improved affinity for HSA compared to the parental molecule. This novel protein domain has been characterized regarding secondary structure and affinity to the two different ligands. The possibility for affinity purification on two different matrices has been investigated using the two ligands, the HSA matrix and the protein A-based, MabSelect SuRe matrix, and the new protein domain was purified to homogeneity. Furthermore, gene fusions between the new domain and three different target proteins with different characteristics were made. To take advantage of both affinities, a purification strategy referred to as orthogonal affinity purification using two different matrices was created. Successful purification of all three versions was efficiently carried out using this strategy.

A small bispecific protein selected for orthogonal affinity purification.

Eine neuartige Protein-Dom&auml;ne mit dual Affinit&auml;t wurde durch Randomisierung und Auswahl erstellt. Die kleine Alkali stabilisierten Albumin-bindende Dom&auml;ne (ABD *), als Ger&uuml;st verwendet, um die Bibliothek zu konstruieren hat Affinit&auml;t zum menschlichen Serum-Albumin (HSA) und konstituiert sich aus 46 Aminos&auml;uren, von denen 11 randomisiert wurden. Um eine duale-Sammelmappe zu erreichen, wurde die Bindungsstelle der inh&auml;renten HSA-Affinit&auml;t unber&uuml;hrt und die Randomisierung wurde auf der gegen&uuml;berliegenden Seite des Molek&uuml;ls. Trotz seiner geringen Gr&ouml;&szlig;e und Randomisierung von fast ein Viertel seiner Aminos&auml;uren, ein Bifunktionelles Molek&uuml;l, ABDz1, mit M&ouml;glichkeit zum Binden an HSA und Z2 Dom&auml;ne/Protein A erfolgreich w&auml;hlte Phagen-Display verwenden. Dar&uuml;ber hinaus zeigte die neu ausgew&auml;hlte Variante verbesserte Affinit&auml;t f&uuml;r HSA im Vergleich zu den elterlichen Molek&uuml;l. Diese neuartige Protein-Dom&auml;ne war bez&uuml;glich Sekund&auml;rstruktur und Affinit&auml;t zu zwei unterschiedlichen Liganden charakterisiert. Die M&ouml;glichkeit der Affinit&auml;tsreinigung auf zwei verschiedenen Matrizen wurde untersucht mit zwei Liganden, der HSA-Matrix und das Protein A-basierte, MabSelect SuRe Matrix und die neue Proteindom&auml;ne zur Homogenit&auml;t gereinigt wurde. Dar&uuml;ber hinaus erfolgten gen Fusionen zwischen der neuen Dom&auml;ne und drei unterschiedliche Ziel-Proteine mit unterschiedlichen Eigenschaften. Um beide Affinit&auml;ten zu nutzen, entstand eine Reinigung-Strategie als orthogonale Affinit&auml;tsreinigung mit zwei verschiedene Matrizen bezeichnet. Erfolgreiche Reinigung aller drei Versionen wurde effizient durchgef&uuml;hrt, mit dieser Strategie.

Bispecific antibodies as well as non-immunoglobulin based bispecific affinity proteins are considered to have a very high potential in future biotherapeutic applications. In this study, we report on a novel approach for generation of extremely small bispecific proteins comprised of only a single structural domain. Binding to tumor necrosis factor-Î± (TNF-Î±) was engineered into an albumin-binding domain while still retaining the original affinity for albumin, resulting in a bispecific protein composed of merely 46 amino acids. By diversification of the non albumin-binding side of the three-helix bundle domain, followed by display of the resulting library on phage particles, bispecific single-domain proteins were isolated using selections with TNF-Î± as target. Moreover, based on the obtained sequences from the phage selection, a second-generation library was designed in order to further increase the affinity of the bispecific candidates. Staphylococcal surface display was employed for the affinity maturation, enabling efficient isolation of improved binders as well as multiparameter-based sortings with both TNF-Î± and albumin as targets in the same selection cycle. Isolated variants were sequenced and the binding to albumin and TNF-Î± was analyzed. This analysis revealed an affinity for TNF-Î± below 5 nM for the strongest binders. From the multiparameter sorting that simultaneously targeted TNF-Î± and albumin, several bispecific candidates were isolated with high affinity to both antigens, suggesting that cell display in combination with fluorescence activated cell sorting is a suitable technology for engineering of bispecificity. To our knowledge, the new binders represent the smallest engineered bispecific proteins reported so far. Possibilities and challenges as well as potential future applications of this novel strategy are discussed.

Engineering bispecificity into a single albumin-binding domain.

Bispezifischer Antik&ouml;rper als auch nicht-Immunglobulin basierend Bispezifischer Affinit&auml;t Proteine werden als eine sehr hohe k&uuml;nftig biotherapeutische Anwendungsm&ouml;glichkeiten haben. In dieser Studie berichten wir &uuml;ber einen neuen Ansatz zur Generierung von extrem kleinen Bispezifischer Proteine bestehen aus nur einer einzigen strukturellen Dom&auml;ne. Bindung an Tumor-Nekrose-Faktor-&alpha; (TNF-&alpha;) wurde in eine Dom&auml;ne im Eiwei&szlig;-Bindung entwickelt, w&auml;hrend noch die Beibehaltung der urspr&uuml;nglichen Affinit&auml;t f&uuml;r Albumin, wodurch ein Bispezifischer Protein setzt sich aus lediglich 46 Aminos&auml;uren. Durch Diversifikation der unverbindlichen Albumin-Seite des drei-Helix-B&uuml;ndel Dom&auml;ne, gefolgt vom Display der resultierende Bibliothek auf Bakteriophagenpartikel wurden Bispezifischer einzelnen Dom&auml;ne Proteine isoliert mit Auswahlen mit TNF-&alpha; als Ziel. Dar&uuml;ber hinaus wurde auf der Grundlage der erhaltenen Sequenzen aus der Phage-Auswahl, eine zweite Generation Bibliothek entworfen um die Affinit&auml;t der Bispezifischer Kandidaten zu steigern. Staphylokokken Oberfl&auml;che Display war f&uuml;r die Affinit&auml;t-Reifung, erm&ouml;glicht effiziente Isolierung verbesserte Bindemittel sowie Multiparameter-basierte Sortings mit TNF-&alpha; und Albumin als Ziele in der gleichen Auswahl Zyklus besch&auml;ftigt. Isolierte Varianten wurden sequenziert und die Bindung an Albumin und TNF-&alpha; wurde analysiert. Diese Analyse ergab eine Affinit&auml;t f&uuml;r TNF-&alpha; unter 5 nM f&uuml;r die st&auml;rkste Bindemittel. Aus der Multiparameter sortieren, dass mehrere Bispezifischer Kandidaten gleichzeitig gezielte TNF-&alpha; und Albumin, isoliert mit hoher Affinit&auml;t zu beide Antigene waren, was darauf hindeutet, dass Zelle ist Display in Kombination mit Fluoreszenz aktivierten Zellsortierung eine geeignete Technologie f&uuml;r Engineering von Bispecificity. Unseres Wissens darstellen der neuen Bindemittel die kleinste ver&auml;nderter Bispezifischer Proteine wurden bisher beschrieben. M&ouml;glichkeiten und Herausforderungen sowie m&ouml;gliche zuk&uuml;nftige Anwendungen dieser neuartige Strategie werden diskutiert.
