The human genome transcribes a large amount of non-coding RNAs, including long non-coding RNAs (lncRNAs) and microRNAs. LncRNAs and microRNAs have been shown to play a critical regulatory role in tumorigenesis and progression. Competitive endogenous RNAs (ceRNAs) affect other RNAs transcription through competitively binding to common microRNAs (miRNAs). MALAT1 is a typical lncRNA that is markedly up-regulated in breast cancer. However, current understanding of the involvement of MALAT1 in breast cancer development and prognosis remains unclear. In the present study, the expression of MALAT1 in clinical samples of breast cancer tissues was found to be significantly up-regulated that was consistent with the result based on the dataset of the Cancer Genome Atlas (TCGA) at cBioportal. A negative correlation between overall survival and the expression of MALAT1 was statistically significant in the group of diagnosis age below 60 or in the group of infiltrating ductal carcinoma analyzed by TCGA database, which declared that MALAT1 might be a potentially useful prognostic factor. Furthermore, the combination of bioinformatics prediction with experimental verifications indicated that lncRNA MALAT1 can regulate BLCAP mRNA expression through binding to miR-339-5p.

The use of nanomedicines for cancer treatment has been widespread. Fullerenes have significant effects in the treatment of solid tumors. Here, we are going to study the effects of hydroxylated fullerene C(OH)(n = 18-22) treatment on chronic myeloid leukemia cell proliferation and investigate its toxicity. The results showed that hydroxylated fullerene C(OH) (n = 18-22) at low concentrations (less than 120 μM) not only had apparent toxic side effects, but also promoted the growth of K562 cells, while a high concentration of C(OH) had different degrees of inhibition on K562 cells. When the concentration is higher than 160 μM, the K562 cells showed morphological changes, the mitochondrial membrane potential decreased, the cell cycle was blocked in the stage of G2-phase, and cell apoptosis occurred, which may cause apoptosis, autophagy, and a variety of other damage leading to cell death. Meanwhile, it also indicated that its inhibition of solid tumors might be related to the tumor microenvironment; we verified the safety of fullerene without apparent cellular toxicity at a specific concentration.

The Effect of Polyhydroxy Fullerene Derivative on Human Myeloid Leukemia K562 Cells.

Deep eutectic solvents (DESs) have physicochemical characteristics similar to those of ionic liquids but are more cost-effective, easier to produce, and less harmful to the environment, making them viable alternatives to ionic liquids. In this study, various DESs have been created to assess their potential as storage media for enzymes. The impact of the DES composition and water content on the thermal and storage stability of cellulase and pectinase was also investigated. Molecular simulation was used to examine the kinetic parameters of cellulase and pectinase in DESs with varying water levels based on choline chloride. The results demonstrated that the stability of the enzymes initially increased and then decreased with an increase in water content in DESs. The enzymes experienced secondary structural changes, leading to variations in fluorescence values. Ultimately, DESs can be utilized as a stabilizers for long-term enzyme preservation, and this study provides a theoretical basis for the coapplication of DESs and enzymes.

Effect of Deep Eutectic Solvents on the Activity and Stability of Cellulases and Pectinases.

Tea saponins have high surface-active and biological activities and are widely used in chemicals, food, pharmaceuticals, and pesticides. Tea saponins are usually extracted using ethanol or water, but both methods have their disadvantages, including a negative impact on the environment, high energy consumption, and low purity. In this study, we explored an effective process for extracting tea saponins from tea meal using deep eutectic solvents combined with ultrasonic extraction and enzymatic techniques. The experimental results showed that a high extraction efficiency of 20.93 ± 0.48% could be achieved in 20 min using an ultrasonic power of 40% and a binary DES consisting of betaine and ethylene glycol (with a molar ratio of 1:3) at a material-liquid ratio of 1:35 and that the purity of the tea saponins after purification by a large-pore adsorption resin reached 95.94%, which was higher than that of commercially available standard tea saponin samples. In addition, the extracted tea saponins were evaluated for their antioxidant and bacteriostatic activities using chemical and biological methods; the results showed that the tea saponins extracted using these methods possessed antioxidant properties and displayed significant antibacterial activity. Therefore, the present study developed a method for using deep eutectic solvents as an environmentally friendly technological solution for obtaining high-purity tea saponins from tea meal oil. This is expected to replace the current organic solvent and water extraction process and has great potential for industrial development and a number of possible applications.

The Extraction Using Deep Eutectic Solvents and Evaluation of Tea Saponin.

Liquid photonic crystals (LPCs) have great application potential in sensors, anti-counterfeiting materials and detection due to their sensitive dynamic responsiveness. Currently, the preparation of LPCs mainly relies on the supersaturation of colloidal nanospheres. However, the supersaturation method usually fails to obtain LPCs based on low-concentration colloidal nanospheres. In turn, the use of high-concentration colloidal nanospheres results in poor mobility of LPCs, which makes them inappropriate for subsequent utilization in responsive systems. In this study, self-assembly of LPCs with vibrant structural colors is achieved through the electrostatic fence effect by introducing an anionic carboxylate-containing polymer as an inducer into the system of low-concentration negatively charged colloidal nanospheres (≥1.25 wt%). It is shown that the anionic carboxylate groups on the inducer molecules and the appropriate molecular chain length are the decisive factors for the inducing effect. The obtained LPCs exhibit a typical non-close-packed structure with a face-centered cubic arrangement of nanospheres. The nearest inter-nanosphere surface distance is 12.10 nm, and the farthest one is as long as 40.30 nm. The LPCs possess good dynamic recovery performance and sensitive optical response characteristics, which are conducive to application in responsive optical sensors directly or after filling.

Electrostatic fence induced assembly of low-concentration colloidal nanospheres to form liquid photonic crystals.

Liquid Photonic Crystals (LPCs) represent a distinctive category of photonic materials that merges the ordered structure of colloidal photonic crystals with the dynamic nature of liquids, allowing for flexibility in tuning their assembly and optical properties in response to external stimuli. However, the requirement of high solid content and high stability of these LPCs continue to pose a significant challenge in their controlled synthesis, efficient assembly, and system stabilization. Herein, highly charged poly (acrylic acid)-b-polystyrene (PAA-b-PS) colloidal nanospheres are synthesized using RAFT-mediated emulsion polymerization. Under the hydration and electrostatic interactions induced by selected polymeric inducers, PAA-b-PS colloidal nanospheres with a uniform carboxylate anion surface are synthesized, capable of forming iridescent LPCs at an overall low solid content (20 wt%) containing localized areas of high solid content. Furthermore, carboxymethyl cellulose (CMC), one of the polymeric inducers, undergoes photosensitive modification to facilitate the digital light processing (DLP) 3D printed LPCs hydrogel models. This strategy offers innovative approaches for the synthesis, assembly, and 3D-printed LPC materials, promising applications in smart displays, sensory systems, and optical devices.

Synergistic Hydrophilic and Electrostatic Induction for Liquid Photonic Crystals of Poly (Acrylic Acid)-block-Polystyrene Colloidal Nanospheres From RAFT-Mediated Emulsion Polymerization.

As some of the most promising candidates available, fullerene-derived bioactive agents have been explored as new drugs with high efficacy and safety for biomedical applications. In this study, a fullerene-lysine derivative (C-Lys) was synthesized successfully and proved to be good at treating type 2 diabetes mellitus (T2DM). C-Lys could alleviate oxidative stress both in streptozotocin (STZ)-induced MIN6 cells and in STZ-induced T2DM mice subjected to a high-fat diet, and it significantly normalized glucose uptake and reduced blood glucose. In addition, C-Lys can alleviate insulin resistance, hyperinsulinemia and lipid levels in T2DM mice. It was further confirmed that C-Lys could alleviate oxidative stress by increasing the activities of antioxidant enzymes and stabilizing the mitochondrial membrane potential (MMP) of pancreatic β-cells to reduce the overproduction of ROS. The results provide compelling evidence that C-Lys possesses promising applications for T2DM treatment.

A novel fullerene-lysine derivative with noticeable ROS scavenging capabilities for improving type 2 diabetes mellitus.

Neurological diseases, which include various neurodegenerative disorders, not only impair patients' physical health but also impact their psychological and social functions. It is particularly urgent to seek effective prevention and treatment strategies for neurological diseases.  Fructus (AOF), a traditional Chinese medicinal herb, has been widely used in treating urinary, digestive, and neurological disorders. Contemporary medical research has demonstrated that AOF exerts neuroprotective effects through multiple mechanisms, including by inhibiting neuronal apoptosis, alleviating neuroinflammation, reducing oxidative stress, and regulating nerve cell dynamic balance. In recent years, substantial advancements have been achieved in investigations concerning the neuroprotective effects and underlying mechanisms of AOF, alongside significant breakthroughs in its clinical applications. This review systematically summarizes the neuroprotective effects of AOF and delineates its clinical applications, thereby offering valuable reference and guidance for the prevention and treatment of neurological diseases using AOF.
