Rett syndrome (RTT) is one of the most prevalent female neurodevelopmental disorders that cause severe mental retardation. Mutations in methyl CpG binding protein 2 (MeCP2) are mainly responsible for RTT. Patients with classical RTT exhibit normal development until age 6-18 mo, at which point they become symptomatic and display loss of language and motor skills, purposeful hand movements, and normal head growth. Murine genetic models and postmortem human brains have been used to study the disease and enable the molecular dissection of RTT. In this work, we applied a recently developed reprogramming approach to generate a novel in vitro human RTT model. Induced pluripotent stem cells (iPSCs) were derived from RTT fibroblasts by overexpressing the reprogramming factors OCT4, SOX2, KLF4, and MYC. Intriguingly, whereas some iPSCs maintained X chromosome inactivation, in others the X chromosome was reactivated. Thus, iPSCs were isolated that retained a single active X chromosome expressing either mutant or WT MeCP2, as well as iPSCs with reactivated X chromosomes expressing both mutant and WT MeCP2. When these cells underwent neuronal differentiation, the mutant monoallelic or biallelelic RTT-iPSCs displayed a defect in neuronal maturation consistent with RTT phenotypes. Our in vitro model of RTT is an important tool allowing the further investigation of the pathophysiology of RTT and the development of the curative therapeutics.

Neuronal maturation defect in induced pluripotent stem cells from patients with Rett syndrome.

Rett-Syndrom (RTT) ist eine der am weitesten verbreitete weibliche Neurodevelopmental St&ouml;rungen, die zu schweren geistiger Retardierung f&uuml;hren. Mutationen im Methyl CpG verbindliches Protein 2 (MeCP2) sind haupts&auml;chlich verantwortlich f&uuml;r RTT. Patienten mit klassischen RTT zeigen normale Entwicklung bis Alter 6-18-Mo, an welcher Stelle sie werden symptomatisch und Verlust der Sprache und Motorik, gezielte Handbewegungen und normale Kopf Wachstum zeigen. Murine genetische Modelle und Post-mortem menschliche Gehirne wurden eingesetzt, die Krankheit zu studieren und aktivieren die molekulare Genetik der RTT. In dieser Arbeit angewendet wir einen neu entwickelten Neuprogrammierung Ansatz um ein neuartiges in vitro menschliche RTT Modell zu generieren. Induzierten pluripotenten Stammzellen (iPSCs) stammen aus RTT Fibroblasten durch die Neuprogrammierung Faktoren OCT4, SOX2, KLF4 und MYC. Interessanterweise &uuml;berexprimierenden w&auml;hrend einige iPSCs x-Chromosom-Inaktivierung, in anderen beibehalten, die das x-Chromosom reaktiviert wurde. So waren die iPSCs isoliert, beibehalten einer einzigen aktiven x-Chromosom auszudr&uuml;cken entweder Mutant oder WT MeCP2, sowie iPSCs mit reaktivierten X-Chromosomen Mutant und WT MeCP2 auszudr&uuml;cken. Wenn diese Zellen neuronale Differenzierung unterzog, angezeigt der mutant monoallelisches oder Biallelelic RTT-iPSCs einen Defekt im Einklang mit RTT Ph&auml;notypen neuronale Reifung. Unsere in-vitro-Modell der RTT ist ein wichtiges Instrument, so dass die weitere Untersuchung der Pathophysiologie von RTT und die Entwicklung des heilenden Therapeutics.

X-chromosome inactivation (XCI) is an important mechanism employed by mammalian XX female cells to level X-linked gene expression with that of male XY cells. XCI occurs early in development as the pluripotent cells of the inner cell mass (ICM) in blastocysts successively differentiate into cells of all three germ layers. X-chromosome reactivation (XCR), the reversal of XCI, is critical for germ cell formation as a mechanism to diversify the X-chromosome gene pool. Here we review the characterization of XCR, and further explore its natural occurrence during development and the in vitro models of cellular reprogramming. We also review the key regulators involved in XCI for their role in suppressing the active histone marks and the genes in the active chromosome for their inhibition of X inactivation signals.

Wissenschaftlerhaus Inaktivierung (XCI) ist ein wichtiger Mechanismus, der von S&auml;ugetierzellen XX weiblich, Ebene, X-chromosomale Genexpression mit der m&auml;nnlichen XY-Zellen eingesetzt. XCI tritt fr&uuml;h in der Entwicklung, wie die pluripotenten Zellen der inneren Zelle Masse (ICM) im Blastozysten sukzessive in Zellen aller drei Ebenen der Keim zu unterscheiden. Darstellung Reaktivierung (XCR), die Umkehrung der XCI, ist entscheidend f&uuml;r Keim Zellbildung als Mechanismus, der Wissenschaftlerhaus-gen-Pool zu diversifizieren. Hier &uuml;berpr&uuml;fen wir die Charakterisierung der XCR, und sein nat&uuml;rliche Vorkommen w&auml;hrend der Entwicklung und der in-vitro-Modelle der zellul&auml;ren Umprogrammierung weiter zu erforschen. Wir &uuml;berpr&uuml;fen auch die wichtigsten Regulatoren, die f&uuml;r ihre Rolle bei der Unterdr&uuml;ckung der aktiven Histon-Marken und den Genen im aktiven Chromosom f&uuml;r ihre Hemmung der X Inaktivierung Signale XCI beteiligt.
