In the essential oil of the liverwort Saccogyna viticulosa, collected on the island of Madeira, the new sesquiterpene hydrocarbons isogermacrene A (5) and its Cope rearrangement product iso-beta-elemene (6) were identified. 5 is proposed to act as the biogenetic precursor of several new sesquiterpenes identified in the volatiles of S. viticulosa. These include iso-alpha-humulene, alpha-gorgonene, gorgona-1,4(15), 11-triene and gorgon- 11-en-4-ol. In addition, the Cope product of zierene, isozierene, allo-aromadendra-4(15),10(14)-diene, aromadendra-4(15),10(14)-dien-1-ol and a prenylguaiane diterpene alcohol, named viticulol, were identified as new natural products.

Cell suspension cultures of Phyllanthus niruri were used to study the lignan profiles and biosynthesis. Suspension cultures yielded two lignans: the new cubebin dimethyl ether (1) and urinatetralin (2), a new lignan from P. niruri, but reported earlier from P. urinaria. This is the first report of cell suspension cultures of P. niruri that successfully produce lignans. Feeding 0.5 mM ferulic acid or 0.5 mM caffeic acid, being early precursors of lignan biosynthesis, resulted in an increase up to 0.7 mg g(-1) DW of 1 (control value 0.1 mg g(-1) DW) and up to 0.3 mg g(-1) DW of 2 (control value 0.2 mg g(-1) DW). Comparison of the lignan profiles of cell suspensions, callus cultures, aerial plant parts, roots, and seeds showed significant differences.

Aussetzung der Zellkulturen von Phyllanthus Niruri dienten die Lignan-Profile und Biosynthese zu studieren. Suspensionskulturen ergab zwei Lignane: die neue Cubebin-Dimethylether (1) und Urinatetralin (2), eine neue Lignan von p. Niruri, sondern berichtet fr&uuml;her von p. Urinaria. Dies ist der erste Bericht der Aussetzung Zellkulturen von p. Niruri, die Lignane erfolgreich zu produzieren. F&uuml;tterung, Ferulas&auml;ure 0,5 mM oder 0,5 mM Kaffees&auml;ure, ergab als fr&uuml;he Vorl&auml;ufer der Lignan-Biosynthese, eine Erh&ouml;hung bis zu 0,7 mg g(-1) DW 1 (Kontrolle Wert 0,1 mg g(-1) DW) und bis zu 0,3 mg g(-1) DW 2 (Kontrolle Wert 0,2 mg g(-1) DW). Vergleich der Lignan profile Zellsuspensionen, Hornhaut-Kulturen, Antenne Pflanzenteile, Wurzeln und Samen zeigte signifikante Unterschiede.

The essential oil of the liverwort Plagiochila bifaria was analysed by GC and GC-MS. Three eudesmane type sesquiterpenes, ent-eudesm-4-en-6-one, ent-eudesm-4(15)-en-6-one, ent-7-hydroxyeudesm-4-en-6-one were isolated and identified as new natural products. Structure elucidation and the determination of absolute configurations are described.

Drei Eudesmane Typ Sesquiterpene, ent-eudesm-4-en-6-one, ent-Eudesm-4 15-de-6-One, Ent-7-hydroxyeudesm-4-en-6-one isoliert und als neue Naturprodukte identifiziert wurden. The reaction of some 1,1'-dialkynylferrocenes with a variety of phenols in the presence as well as in the absence of [Mo(CO)(6)] yields good to high yields of phenoxy[4]ferrocenophanedienes. Similar reactivity was observed with a thiophenol and with acetic acid. Reaction under basic reaction conditions led to the formation of the [4]ferrocenophanone 17. The phenoxy[4]ferrocenophanedienes obtained show dynamic behavior as a result of a torsional twist of the carbon bridge as indicated by the (1)H and (13)C NMR spectra. The reaction mechanism is discussed in view of recent related results of Sato et al. as well as of Pudelski et al. A vinyl cation intermediate is postulated in this context, whose relative stability is evident from the mass spectra of the compounds prepared.

Die Reaktion von rund 1, 1&#39;-Dialkynylferrocenes mit einer Vielzahl von Phenole in der Gegenwart ebenso wie das Fehlen von [Mo(CO)(6)] ergibt gute, hohe Ertr&auml;ge von Phenoxy [4] Ferrocenophanedienes. &Auml;hnliche Reaktivit&auml;t wurde mit einem Thiophenol und Essigs&auml;ure beobachtet. Reaktion unter grundlegenden Reaktionsbedingungen f&uuml;hrte zur Gr&uuml;ndung von der [4] Ferrocenophanone 17. Die Phenoxy [4] Ferrocenophanedienes erhalten zeigen dynamisches Verhalten durch eine Torsionssteifigkeit Wendung der Kohlenstoff-Br&uuml;cke durch H (1) und (13) C-NMR-Spektren. Der Reaktionsmechanismus ist angesichts der j&uuml;ngsten &auml;hnliche Ergebnisse von Sato Et Al. sowie der Pudelski Et al. diskutiert. Eine Vinyl-kation Mittelstufe wird in diesem Zusammenhang postuliert, deren relativer Stabilit&auml;t aus der Massenspektren der Verbindungen vorbereitet ist.

Anaphylaxis mediated by carbohydrate structures is a controversially discussed phenomenon. Nevertheless, IgE with specificity for the xenotransplantation antigen Î±1,3-Gal (Î±-Gal) are associated with a delayed type of anaphylaxis, providing evidence for the clinical relevance of carbohydrate epitopes in allergy. The aim of this study was to dissect immunoreactivity, interaction, and fine epitope of Î±-Gal-specific antibodies to obtain insights into the recognition of carbohydrate epitopes by IgE antibodies and their consequences on a molecular and cellular level. The antigen binding moiety of an Î±-Gal-specific murine IgM antibody was employed to construct chimeric IgE and IgG antibodies. Reactivity and specificity of the resulting antibodies were assessed by means of ELISA and receptor binding studies. Using defined carbohydrates, interaction of the IgE and human serum was assessed by mediator release assays, surface plasmon resonance (SPR), and saturation transfer difference NMR analyses. The Î±-Gal-specific chimeric IgE and IgG antibodies were proven functional regarding interaction with antigen and Fc receptors. SPR measurements demonstrated affinities in the micromolar range. In contrast to a reference antibody, anti-Gal IgE did not induce mediator release, potentially reflecting the delayed type of anaphylaxis. The Î±1,3-Gal epitope fine structures of both the recombinant IgE and affinity-purified serum were defined by saturation transfer difference NMR, revealing similar contributions of carbohydrate residues and participation of both galactose residues in interaction. The antibodies generated here constitute the principle underlying Î±1,3-Gal-mediated anaphylaxis. The complementary data of affinity and fine specificity may help to elucidate the recognition of carbohydrates by the adaptive immune response and the molecular requirements of carbohydrate-based anaphylaxis.

Anaphylaxie, vermittelt durch Kohlenhydrat-Strukturen ist ein kontrovers diskutierte Ph&auml;nomen. Trotzdem IgE mit Spezifit&auml;t f&uuml;r die Xenotransplantation Antigen &alpha;1, 3-Gal (&alpha;-Gal) sind einem verz&ouml;gerten Typ der Anaphylaxie, den Nachweis f&uuml;r die klinische Relevanz der Kohlenhydrat-Epitope im Allergie zugeordnet. Ziel dieser Studie war, Immunoreactivity, Interaktion und feine Epitop &alpha;-Gal-spezifische Antik&ouml;rper erhalten Sie Einblicke in die Anerkennung der Kohlenhydrat-Epitope durch IgE-Antik&ouml;rper und deren Folgen auf molekularer und zellul&auml;rer Ebene zu zergliedern. Die Antigen-Bindung-Gruppe von einer &alpha;-Gal-spezifische murinen IgM-Antik&ouml;rper wurde angestellt, um Chim&auml;re IgE und IgG-Antik&ouml;rpern zu konstruieren. Die &alpha;-Gal-spezifische Chim&auml;ren IgE und IgG-Antik&ouml;rper wurden funktionale hinsichtlich Interaktion mit Antigen und Fc-Rezeptoren nachgewiesen. SPR-Messungen zeigten Affinit&auml;ten im micromolar Bereich. Im Gegensatz zu einem Referenz-Antik&ouml;rper Anti-Gal IgE nicht Mediator Freigabe, induzieren, potenziell reflektieren den verz&ouml;gerten Typ der Anaphylaxie. Das &alpha;1, 3-Gal-Epitop feine Strukturen der beiden die rekombinante IgE und Affinit&auml;t gereinigt Serum wurden durch S&auml;ttigung Transfer Differenz NMR, offenbart &auml;hnliche Beitr&auml;ge von Kohlenhydrat-R&uuml;ckst&auml;nden und Beteiligung der beiden Galaktose-R&uuml;ckst&auml;nde in Interaktion definiert. Die hier erzeugten Antik&ouml;rper bilden das Prinzip zugrunde liegenden &alpha;1, 3-Gal-vermittelten Anaphylaxie. Die erg&auml;nzenden Daten der Affinit&auml;t und feine Spezifit&auml;t k&ouml;nnen helfen, um die Anerkennung der Kohlenhydrate durch die adaptive Immunantwort und die molekularen Anforderungen der Kohlenhydrat-basierte Anaphylaxie herauszustellen.

(1)H NMR spectroscopy was used to follow the cleavage of sucrose by invertase. The parameters of the enzyme's kinetics, K(m) and V(max), were directly determined from progress curves at only one concentration of the substrate. For comparison with the classical Michaelis-Menten analysis, the reaction progress was also monitored at various initial concentrations of 3.5 to 41.8mM. Using the Lambert W function the parameters K(m) and V(max) were fitted to obtain the experimental progress curve and resulted in K(m)=28mM and V(max)=13Î¼M/s. The result is almost identical to an initial rate analysis that, however, costs much more time and experimental effort. The effect of product inhibition was also investigated. Furthermore, we analyzed a much more complex reaction, the conversion of farnesyl diphosphate into (+)-germacrene D by the enzyme germacrene D synthase, yielding K(m)=379Î¼M and k(cat)=0.04s(-1). The reaction involves an amphiphilic substrate forming micelles and a water insoluble product; using proper controls, the conversion can well be analyzed by the progress curve approach using the Lambert W function.

(1)H-NMR-Spektroskopie wurde die Spaltung von Saccharose zu folgen von Invertase verwendet. Die Parameter der Enzym-Kinetik, K(m) und V(max), wurden direkt vom Fortschritt Kurven nur eine Konzentration des Substrats bestimmt. Zum Vergleich mit der klassischen Michaelis-Menten-Analyse war der Reaktion-Fortschritt auch in unterschiedlichen anf&auml;ngliche Konzentrationen von 3.5 bis 41,8 mM beobachtet. Mithilfe der lambertschen W-Funktion die Parameter K(m) und V(max) wurden eingebaut, um die experimentelle Fortschritte-Kurve zu erhalten und f&uuml;hrte zu K (m) = 28 mM und V (Max) = 13&mu;M/s. Das Ergebnis ist fast identisch mit dem ersten Satz Analyse, die jedoch sehr viel mehr Zeit und experimentelle Aufwand kostet. Die Wirkung der Produkt-Hemmung wurde auch untersucht. Wir analysierten dar&uuml;ber hinaus eine viel komplexere Reaktion, die Umwandlung von Farnesyl-Diphosphat in (+)-Germacrene D durch das Enzym Germacrene D-Synthase, woraus K (m) = 379&mu;M und k(cat)=0.04s(-1). Die Reaktion erfolgt eine AMPHIPHILE-Substrat, die Bildung von Mizellen und ein Wasser unl&ouml;sliche Produkt; verwenden geeignete Kontrollen, kann die Konvertierung auch durch den Fortschritt-Kurve-Ansatz mit der lambertschen W-Funktion analysiert werden.

The first step of any molecular phylogenetic analysis is the selection of the species and sequences to be included, the taxon sampling. Already here different pitfalls exist. Sequences can contain errors, annotations in databases can be inaccurate and even the taxonomic classification of a species can be wrong. Usually, these artefacts become evident only after calculation of the phylogenetic tree. Following, the taxon sampling has to be corrected iteratively. This can become tedious and time consuming, as in most cases the taxon sampling is de-coupled from the further steps of the phylogenetic analysis. Here, we present the ITS2 Workbench (http://its2.bioapps.biozentrum.uni-wuerzburg.de/), which eliminates this problem by a tight integration of taxon sampling, secondary structure prediction, multiple alignment and phylogenetic tree calculation. The ITS2 Workbench has access to more than 280,000 ITS2 sequences and their structures provided by the ITS2 database enabling sequence-structure based alignment and tree reconstruction. This allows the interactive improvement of the taxon sampling throughout the whole phylogenetic tree reconstruction process. Thus, the ITS2 Workbench enables a fast, interactive and iterative taxon sampling leading to more accurate ITS2 based phylogenies.

Der erste Schritt eine molekulare phylogenetische Analyse ist die Auswahl der Arten und Sequenzen zugerechnet werden das Taxon-Sampling. Bereits existieren hier verschiedene Fallstricke. Sequenzen k&ouml;nnen Fehler enthalten, Anmerkungen in Datenbanken k&ouml;nnen ungenau und sogar die taxonomische Klassifikation einer Spezies kann falsch sein. In der Regel werden diese Artefakte erst nach Berechnung der phylogenetische Struktur deutlich. Nach, hat die Probenahme Taxon iterativ korrigiert werden. Dies kann werden m&uuml;hsam und zeitaufw&auml;ndig, wie in den meisten F&auml;llen wird die Taxon-Stichproben aus den weiteren Schritten der phylogenetischen Analyse abgekoppelt ist. Hier pr&auml;sentieren wir die ITS2 Workbench (http://its2.bioapps.biozentrum.uni-wuerzburg.de/), die dieses Problem durch eine enge Integration von Taxon Probenahme, Sekund&auml;rstruktur Vorhersage, mehrere Ausrichtung und phylogenetische Stammbaum Berechnung eliminiert. Die Workbench ITS2 hat Zugang zu mehr als 280.000 ITS2 Sequenzen und deren Strukturen, bereitgestellt durch die ITS2 Datenbank basierende Sequenz-Struktur Ausrichtung und Struktur Wiederaufbau aktivieren. Dies erm&ouml;glicht die interaktive Verbesserung der Taxon Probenahme w&auml;hrend des gesamten phylogenetischen Baum-Wiederaufbauprozesses. Die ITS2-Workbench erm&ouml;glicht, eine schnelle, interaktive und iterative Taxon-Probenahme f&uuml;hrt zu genaueren ITS2 basierte am&ouml;benartigen.
