High-frequency oscillatory potentials (HFOPs) have been recorded from ganglion cells in cat, rabbit, frog, and mudpuppy retina and in electroretinograms (ERGs) from humans and other primates. However, the origin of HFOPs is unknown. Based on patterns of tracer coupling, we hypothesized that HFOPs could be generated, in part, by negative feedback from axon-bearing amacrine cells excited via electrical synapses with neighboring ganglion cells. Computer simulations were used to determine whether such axon-mediated feedback was consistent with the experimentally observed properties of HFOPs. (1) Periodic signals are typically absent from ganglion cell PSTHs, in part because the phases of retinal HFOPs vary randomly over time and are only weakly stimulus locked. In the retinal model, this phase variability resulted from the nonlinear properties of axon-mediated feedback in combination with synaptic noise. (2) HFOPs increase as a function of stimulus size up to several times the receptive-field center diameter. In the model, axon-mediated feedback pooled signals over a large retinal area, producing HFOPs that were similarly size dependent. (3) HFOPs are stimulus specific. In the model, gap junctions between neighboring neurons caused contiguous regions to become phase locked, but did not synchronize separate regions. Model-generated HFOPs were consistent with the receptive-field center dynamics and spatial organization of cat alpha cells. HFOPs did not depend qualitatively on the exact value of any model parameter or on the numerical precision of the integration method. We conclude that HFOPs could be mediated, in part, by circuitry consistent with known retinal anatomy.

A model of high-frequency oscillatory potentials in retinal ganglion cells.

We use both analytical arguments and detailed numerical evidence to show that the first-order transition in the type-I two-dimensional Abelian-Higgs model is commensurate with the statistical behavior of its vortex fluctuations, which behave as an ensemble of attractive particles. The clustering instabilities of such ensembles are shown to be connected to the process of phase nucleation. Calculations of the vortex equation of state show that the temperature for the onset of clustering is in qualitative agreement with the critical temperature. The vortex description provides a general gauge invariant mesoscopic mechanism for the first-order transition and applies for arbitrary type-I couplings.

Vortex description of the first-order phase transition in the two-dimensional Abelian-Higgs model.

We show that coherent oscillations among neighboring ganglion cells in a retinal model encode global topological properties, such as size, that cannot be deduced unambiguously from their local, time-averaged firing rates. Whereas ganglion cells may fire similar numbers of spikes in response to both small and large spots, only large spots evoke coherent high frequency oscillations, potentially allowing downstream neurons to infer global stimulus properties from their local afferents. To determine whether such information might be extracted over physiologically realistic spatial and temporal scales, we analyzed artificial spike trains whose oscillatory correlations were similar to those measured experimentally. Oscillatory power in the upper gamma band, extracted on single-trials from multi-unit spike trains, supported good to excellent size discrimination between small and large spots, with performance improving as the number of cells and/or duration of the analysis window was increased. By using Poisson distributed spikes to normalize the firing rate across stimulus conditions, we further found that coincidence detection, or synchrony, yielded substantially poorer performance on identical size discrimination tasks. To determine whether size encoding depended on contiguity independent of object shape, we examined the total oscillatory activity across the entire model retina in response to random binary images. As the ON-pixel probability crossed the percolation threshold, which marks the sudden emergence of large connected clusters, the total gamma-band activity exhibited a sharp transition, a phenomena that may be experimentally observable. Finally, a reanalysis of previously published oscillatory responses from cat ganglion cells revealed size encoding consistent with that predicted by the retinal model.

We apply an information-theoretic treatment of action potential time series measured with microelectrode arrays to estimate the connectivity of mammalian neuronal cell assemblies grown in vitro. We infer connectivity between two neurons via the measurement of the mutual information between their spike trains. In addition we measure higher-point multi-information between any two spike trains, conditional on the activity of a third cell, as a means to identify and distinguish classes of functional connectivity among three neurons. The use of a conditional three-cell measure removes some interpretational shortcomings of the pairwise mutual information and sheds light on the functional connectivity arrangements of any three cells. We analyze the resultant connectivity graphs in light of other complex networks and demonstrate that, despite their ex vivo development, the connectivity maps derived from cultured neural assemblies are similar to other biological networks and display nontrivial structure in clustering coefficient, network diameter, and assortative mixing. Specifically we show that these networks are weakly disassortative small-world graphs, which differ significantly in their structure from randomized graphs with the same degree. We expect our analysis to be useful in identifying the computational motifs of a wide variety of complex networks, derived from time series data.

A major challenge in analyzing animal behavior is to discover some underlying simplicity in complex motor actions. Here, we show that the space of shapes adopted by the nematode Caenorhabditis elegans is low dimensional, with just four dimensions accounting for 95% of the shape variance. These dimensions provide a quantitative description of worm behavior, and we partially reconstruct "equations of motion" for the dynamics in this space. These dynamics have multiple attractors, and we find that the worm visits these in a rapid and almost completely deterministic response to weak thermal stimuli. Stimulus-dependent correlations among the different modes suggest that one can generate more reliable behaviors by synchronizing stimuli to the state of the worm in shape space. We confirm this prediction, effectively "steering" the worm in real time.

Dimensionality and dynamics in the behavior of C. elegans.

We introduce optogenetic investigation of neurotransmission (OptIoN) for time-resolved and quantitative assessment of synaptic function via behavioral and electrophysiological analyses. We photo-triggered release of acetylcholine or gamma-aminobutyric acid at Caenorhabditis elegans neuromuscular junctions using targeted expression of Chlamydomonas reinhardtii Channelrhodopsin-2. In intact Channelrhodopsin-2 transgenic worms, photostimulation instantly induced body elongation (for gamma-aminobutyric acid) or contraction (for acetylcholine), which we analyzed acutely, or during sustained activation with automated image analysis, to assess synaptic efficacy. In dissected worms, photostimulation evoked neurotransmitter-specific postsynaptic currents that could be triggered repeatedly and at various frequencies. Light-evoked behaviors and postsynaptic currents were significantly (P <or= 0.05) altered in mutants with pre- or postsynaptic defects, although the behavioral phenotypes did not unambiguously report on synaptic function in all cases tested. OptIoN facilitates the analysis of neurotransmission with high temporal precision, in a neurotransmitter-selective manner, possibly allowing future investigation of synaptic plasticity in C. elegans.

Verbal communication is a joint activity; however, speech production and comprehension have primarily been analyzed as independent processes within the boundaries of individual brains. Here, we applied fMRI to record brain activity from both speakers and listeners during natural verbal communication. We used the speaker's spatiotemporal brain activity to model listeners' brain activity and found that the speaker's activity is spatially and temporally coupled with the listener's activity. This coupling vanishes when participants fail to communicate. Moreover, though on average the listener's brain activity mirrors the speaker's activity with a delay, we also find areas that exhibit predictive anticipatory responses. We connected the extent of neural coupling to a quantitative measure of story comprehension and find that the greater the anticipatory speaker-listener coupling, the greater the understanding. We argue that the observed alignment of production- and comprehension-based processes serves as a mechanism by which brains convey information.

We consider words as a network of interacting letters, and approximate the probability distribution of states taken on by this network. Despite the intuition that the rules of English spelling are highly combinatorial and arbitrary, we find that maximum entropy models consistent with pairwise correlations among letters provide a surprisingly good approximation to the full statistics of words, capturing âˆ¼92% of the multi-information in four-letter words and even "discovering" words that were not represented in the data. These maximum entropy models incorporate letter interactions through a set of pairwise potentials and thus define an energy landscape on the space of possible words. Guided by the large letter redundancy we seek a lower-dimensional encoding of the letter distribution and show that distinctions between local minima in the landscape account for âˆ¼68% of the four-letter entropy. We suggest that these states provide an effective vocabulary which is matched to the frequency of word use and much smaller than the full lexicon.

Organisms move through the world by changing their shape, and here we explore the mapping from shape space to movements in the nematode Caenorhabditis elegans as it crawls on an agar plate. We characterize the statistics of the trajectories through the correlation functions of the orientation angular velocity, orientation angle and the mean-squared displacement, and we find that the loss of orientational memory has significant contributions from both abrupt, large amplitude turning events and the continuous dynamics between these events. Further, we discover long-time persistence of orientational memory in the intervals between abrupt turns. Building on recent work demonstrating that C. elegans movements are restricted to a low-dimensional shape space, we construct a map from the dynamics in this shape space to the trajectory of the worm along the agar. We use this connection to illustrate that changes in the continuous dynamics reveal subtle differences in movement strategy that occur among mutants defective in two classes of dopamine receptors.

What fascinates us about animal behavior is its richness and complexity, but understanding behavior and its neural basis requires a simpler description. Traditionally, simplification has been imposed by training animals to engage in a limited set of behaviors, by hand scoring behaviors into discrete classes, or by limiting the sensory experience of the organism. An alternative is to ask whether we can search through the dynamics of natural behaviors to find explicit evidence that these behaviors are simpler than they might have been. We review two mathematical approaches to simplification, dimensionality reduction and the maximum entropy method, and we draw on examples from different levels of biological organization, from the crawling behavior of Caenorhabditis elegans to the control of smooth pursuit eye movements in primates, and from the coding of natural scenes by networks of neurons in the retina to the rules of English spelling. In each case, we argue that the explicit search for simplicity uncovers new and unexpected features of the biological system and that the evidence for simplification gives us a language with which to phrase new questions for the next generation of experiments. The fact that similar mathematical structures succeed in taming the complexity of very different biological systems hints that there is something more general to be discovered.

Searching for simplicity in the analysis of neurons and behavior.

Wir &uuml;berpr&uuml;fen zwei mathematische Ans&auml;tze zur Vereinfachung, Reduzierung der Dimensionalit&auml;t und die maximale Entropie-Methode, und wir ziehen auf Beispiele aus verschiedenen Ebenen der biologischen Organisation, vom Crawlen Verhalten von Caenorhabditis Elegans die Kontrolle der glatte Verfolgung Augenbewegungen bei Primaten und die Kodierung der nat&uuml;rlichen Szenen durch Netzwerke von Nervenzellen in der Netzhaut an die Regeln der englischen Schreibweise. In jedem Fall behaupten wir, dass die explizite Suche nach Einfachheit neue und unerwartete Funktionen des biologischen Systems deckt und die Beweise f&uuml;r eine Vereinfachung uns eine Sprache mit der gibt Formulierung der neue Fragen f&uuml;r die n&auml;chste Generation von Experimenten. Tierische Verhaltensweisen sind oft zerlegbaren in diskreter, Stereotype Elemente, gut in der Zeit getrennt. In einem Modell werden solche Verhaltensweisen durch spezifische Befehle ausgel&ouml;st; im Extremfall ist das Discreteness Verhalten auf das Discreteness der Aktionspotentiale in den Neuronen der einzelnen Befehl zur&uuml;ckgef&uuml;hrt. Hier wird das Crawlen Verhalten der Fadenwurm Caenorhabditis elegans verwendet, um die entgegengesetzte Ansicht zu demonstrieren, in der Discreteness, Klischeehaftigkeit und langen Zeitskalen der kollektive Dynamik des Verhaltens selbst hervorgehen. In fr&uuml;heren Arbeiten fanden wir, dass als C. Elegans crawlen, seinen K&ouml;rper bewegt sich durch einen &quot;Form-Raum&quot;, in dem vier Dimensionen etwa 95 % der Varianz in K&ouml;rperform erfassen. Entstehung der langen Zeitskalen und Stereotypen Verhaltensweisen in Caenorhabditis Elegans.

Animal behaviors often are decomposable into discrete, stereotyped elements, well separated in time. In one model, such behaviors are triggered by specific commands; in the extreme case, the discreteness of behavior is traced to the discreteness of action potentials in the individual command neurons. Here, we use the crawling behavior of the nematode Caenorhabditis elegans to demonstrate the opposite view, in which discreteness, stereotypy, and long timescales emerge from the collective dynamics of the behavior itself. In previous work, we found that as C. elegans crawls, its body moves through a "shape space" in which four dimensions capture approximately 95% of the variance in body shape. Here we show that stochastic dynamics within this shape space predicts transitions between attractors corresponding to abrupt reversals in crawling direction. With no free parameters, our inferred stochastic dynamical system generates reversal timescales and stereotyped trajectories in close agreement with experimental observations. We use the stochastic dynamics to show that the noise amplitude decreases systematically with increasing time away from food, resulting in longer bouts of forward crawling and suggesting that worms can use noise to modify their locomotory behavior.

Emergence of long timescales and stereotyped behaviors in Caenorhabditis elegans.

The ability to respond to chemical stimuli is fundamental to the survival of motile organisms, but the strategies underlying odour tracking remain poorly understood. Here we show that chemotaxis in Drosophila melanogaster larvae is an active sampling process analogous to sniffing in vertebrates. Combining computer-vision algorithms with reconstructed olfactory environments, we establish that larvae orient in odour gradients through a sequential organization of stereotypical behaviours, including runs, stops, lateral head casts and directed turns. Negative gradients, integrated during runs, control the timing of turns. Positive gradients detected through high-amplitude head casts determine the direction of individual turns. By genetically manipulating the peripheral olfactory circuit, we examine how orientation adapts to losses and gains of function in olfactory input. Our findings suggest that larval chemotaxis represents an intermediate navigation strategy between the biased random walks of Escherichia Coli and the stereo-olfaction observed in rats and humans.

Aktive Probenahme und Decision Making in Drosophila Chemotaxis.

The nervous functions of an organism are primarily reflected in the behavior it is capable of. Measuring behavior quantitatively, at high-resolution and in an automated fashion provides valuable information about the underlying neural circuit computation. Accordingly, computer-vision applications for animal tracking are becoming a key complementary toolkit to genetic, molecular and electrophysiological characterization in systems neuroscience.

Die nerv&ouml;sen Funktionen des Organismus spiegeln sich vor allem im Verhalten, die, denen es f&auml;hig ist. Verhalten quantitativ, bei hoher Aufl&ouml;sung und automatisiertes Messen bietet wertvolle Informationen &uuml;ber die zugrunde liegenden neuronalen Schaltkreisberechnung. Dementsprechend werden Computer-Vision-Anwendungen f&uuml;r das Tier Tracking eine wichtige erg&auml;nzende Toolkit auf genetische, Molekulare und elektrophysiologische Charakterisierung der Systeme Neurowissenschaften.
