Although tumors frequently show elevated protease activities, the concept of anti-proteolytic cancer therapy has lost momentum after failure of clinical trials with broad-spectrum matrix metalloproteinase inhibitors. Thus we need to adapt our design strategies for protease inhibitors. Here, we employed a series of seven structurally fine-modulated and pharmacokinetically closely related synthetic 4-amidinobenzylamine-based inhibitors with distinct selectivity for prototypical serine proteases in a murine T cell lymphoma liver metastasis model. This in vivo screening revealed efficacy of urokinase inhibitors but no correlation between urokinase selectivity or affinity and anti-metastatic effect. In contrast, factor Xa-selective inhibitors were more potent, demonstrating factor Xa or a factor Xa-like serine protease likely to be more determinant in this model. Factor Xa selectivity, but not affinity, significantly improved anti-metastatic efficacy. For example, factor Xa inhibitors CJ-504 and CJ-510 exert similar affinity for factor Xa (K(i)=14 nM versus 8.8 nM) but CJ-504 was 70-fold more selective for factor Xa. This correlated with higher anti-metastatic efficacy (58.8% with CJ-504; 28.2% with CJ-510). Our results show that among the protease inhibitors employed that have affinities in the nanomolar range, the strategy of selectivity-optimization is superior to further improvement of affinity to significantly enhance anti-metastatic efficacy. This appreciation may be important for the future rational design of new anti-proteolytic agents for cancer therapy.

Increase of anti-metastatic efficacy by selectivity- but not affinity-optimization of synthetic serine protease inhibitors.

Obwohl Tumoren h&auml;ufig erh&ouml;hten Protease T&auml;tigkeiten zeigen, hat das Konzept des Anti-proteolytischen Krebstherapie Dynamik nach dem Ausfall von klinischen Pr&uuml;fungen mit Breitspektrum-Matrix Metalloprotease Inhibitoren verloren. Wir m&uuml;ssen daher unsere Design-Strategien f&uuml;r Protease-Inhibitoren anzupassen. The serine protease urokinase-type plasminogen activator (uPA) interacts with a specific receptor (uPAR) on the surface of various cell types, including tumor cells, and plays a crucial role in pericellular proteolysis. High levels of uPA and uPAR often correlate with poor prognosis of cancer patients. Therefore, the specific inhibition of uPA with small molecule active-site inhibitors is one strategy to decrease the invasive and metastatic activity of tumor cells. We have developed a series of highly potent and selective uPA inhibitors with a C-terminal 4-amidinobenzylamide residue. Optimization was directed toward reducing the fast elimination from circulation that was observed with initial analogues. The x-ray structures of three inhibitor/uPA complexes have been solved and were used to improve the inhibition efficacy. One of the most potent and selective derivatives, benzylsulfonyl-D-Ser-Ser-4-amidinobenzylamide (inhibitor 26), inhibits uPA with a Ki of 20 nm. This inhibitor was used in a fibrosarcoma model in nude mice using lacZ-tagged human HT1080 cells, to prevent experimental lung metastasis formation. Compared with control (100%), an inhibitor dose of 2 x 1.5 mg/kg/day reduced the number of experimental metastases to 4.6 +/- 1%. Under these conditions inhibitor 26 also significantly prolonged survival. All mice from the control group died within 43 days after tumor cell inoculation, whereas 50% of mice from the inhibitor-treated group survived more than 117 days. This study demonstrates that the specific inhibition of uPA by these inhibitors may be a useful strategy for the treatment of cancer to prevent metastasis.

Design of novel and selective inhibitors of urokinase-type plasminogen activator with improved pharmacokinetic properties for use as antimetastatic agents.

Der Serin-Protease Urokinase Plasminogen Aktivator (uPA) interagiert mit einem spezifischen Rezeptor (uPAR) auf der Oberfl&auml;che der verschiedenen Zelltypen, einschlie&szlig;lich Tumor-Zellen, und spielt eine entscheidende Rolle in Pericellular Proteolyse. Hohe uPA und uPAR korrelieren oft mit schlechten Prognose von Krebspatienten. Daher ist die spezifische Hemmung der uPA mit niedermolekularen aktiven-Site-Inhibitoren eine Strategie, die invasive und metastasiertem Aktivit&auml;t der Tumorzellen zu verringern. Wir haben eine Reihe von sehr potenter und selektiver uPA-Inhibitoren mit einer C-terminalen-4-Amidinobenzylamide-R&uuml;ckstand entwickelt. Here, we investigated whether increased TIMP-1 levels in the liver achieved by adenoviral gene transfer will effectively inhibit liver metastasis of two independent tumor cell lines.

Adenovirus-mediated overexpression of tissue inhibitor of metalloproteinases-1 in the liver: efficient protection against T-cell lymphoma and colon carcinoma metastasis.

Matrix metalloproteinases (MMPs), and in particular gelatinases (MMP-2 and MMP-9), play a key role in cancer progression. However, clinical trials in which MMP inhibitors were tested in cancer patients have been disappointing. Whereas many reasons have been postulated to explain the failure of the clinical trials, lack of inhibitor selectivity was a major limitation. Thus, despite the consensus opinion that MMP-mediated proteolysis is essential for cancer progression and that certain MMPs represent important targets for intervention, effective and selective inhibition of those MMPs remains a major challenge in drug development. We previously reported the first mechanism-based MMP inhibitor, designated SB-3CT, which is a selective gelatinase inhibitor. Here we report that SB-3CT (5-50 mg/kg/d) is a potent inhibitor of liver metastasis and increases survival in an aggressive mouse model of T-cell lymphoma. This study shows that mechanism-based inhibition of gelatinases represents a novel approach to inhibitor design that promises to be a successful anticancer therapy.

Antimetastatic activity of a novel mechanism-based gelatinase inhibitor.

Klinische Studien, in denen, die MMP Inhibitoren bei Krebspatienten getestet wurden, waren jedoch entt&auml;uschend. W&auml;hrend viele Gr&uuml;nde postuliert worden, haben das Scheitern der klinischen Pr&uuml;fungen zu erkl&auml;ren, war der Mangel an Inhibitor Selektivit&auml;t eine erhebliche Einschr&auml;nkung. So bleibt trotz der Konsensus-Meinung, dass die MMP-vermittelte Proteolyse ist essenziell f&uuml;r Fortschreiten und bestimmte MMPs wichtige Ziele f&uuml;r die Intervention darstellen, wirksame und selektive Hemmung der diese MMPs eine gro&szlig;e Herausforderung in der Medikamentenentwicklung. Wir berichteten bereits die erste Mechanismus basierende MMP Inhibitor, bezeichnete SB-3 CT, die eine selektive Gelatinase-Inhibitor. Hier berichten wir, die SB-3 CT (5-50 mg/kg/d) ist ein potenter Inhibitor der Leber Metastase und erh&ouml;ht die &uuml;berleben in einem aggressiven Mausmodell der T-Zell-Lymphom. Diese Studie zeigt, dass die Wirkmechanismen Hemmung der Gelatinases einen neuartigen Ansatz zur Inhibitor darstellt, die zu einer erfolgreichen Anti-Krebs-Therapie zu werden verspricht.

Antimetastatic T&auml;tigkeit von einem neuartigen Wirkmechanismen Gelatinase-Inhibitor.

Clinical and experimental evidence suggests that the blood coagulation system is involved in the dissemination of malignant tumors. Consequently, anticoagulant agents have been tested as metastasis suppressors in experimental models. Recently, we have found a close correlation between factor Xa (FXa)-specificity of a series of synthetic serine protease inhibitors and their anti-metastatic potential in a murine T-cell lymphoma metastasis model. Interference of such inhibitors with blood-coagulation may represent a major experimental and clinical obstacle. Here, we test anti-metastatic effects of a recently developed, highly specific 3-amidinophenylalanine-type FXa inhibitor, WX-FX4, with weaker anticoagulant activity when compared to well-established FXa inhibitors, such as DX-9065a, as measured by the activated partial thromboplastin time, prothrombin time, prothrombinase complex activity, and coagulation time. Treatment of mice with WX-FX4 (1.5 mg/kg twice daily) led to significant reduction of experimental liver metastasis of a syngeneic T-cell lymphoma in DBA/2 mice (> 90%), and of experimental lung metastasis of a human fibrosarcoma in CD1 nu/nu mice (> 60%). Due to its relatively low anticoagulant activity, daily treatment over 100 days was possible, leading to significant survival benefits without inducing bleeding anomalities. FXa-inhibitors with highly efficient anti-metastatic potential without coagulation-related side effects may represent important new tools as anticancer agents.

Klinische und experimentelle Hinweise darauf, dass das Blut-Gerinnung-System zur Verbreitung von malignen Tumoren beteiligt ist. Folglich wurden gerinnungshemmende Mittel als Unterdr&uuml;cker der Metastasierung in experimentelle Modelle getestet. Vor kurzem haben wir eine enge Korrelation zwischen Faktor Xa (FXa) gefunden-Spezifit&auml;t einer Reihe von synthetischen Serin-Protease-Inhibitoren und ihre anti-metastatischen Potential in einem murinen T-Zell-Lymphom-Metastase-Modell. Durch seine relativ geringe gerinnungshemmende waren t&auml;gliche Behandlung &uuml;ber 100 Tage m&ouml;glich, f&uuml;hrt zu signifikanten &Uuml;berlebensvorteil ohne damit Blutungen Anomalities. FXa-Hemmer mit hoch effizienten anti-metastatischen Potential ohne Nebenwirkungen im Zusammenhang mit Koagulation k&ouml;nnen wichtige neue Instrumente als Anti-Krebs-Bevollm&auml;chtigte vertreten.

The L1 cell adhesion molecule is implicated in the control of proliferation, migration, and invasion of several tumor cell types in vitro. Recently, L1 overexpression was found to correlate with tumor progression of ovarian carcinoma, one of the most common causes of cancer-related deaths in gynecologic malignant diseases. To evaluate L1 as a potential target for ovarian cancer therapy, we investigated the effects of anti-L1 monoclonal antibodies (chCE7 and L1-11A) on proliferation and migration of L1-positive human SKOV3ip ovarian carcinoma cells in vitro and the therapeutic efficacy of L1-11A against i.p. SKOV3ip tumor growth in nude mice. In vitro, both anti-L1 antibodies efficiently inhibited the proliferation of SKOV3ip cells as well as other L1-expressing tumor cell lines (renal carcinoma, neuroblastoma, and colon carcinoma). On two cell lines, hyper-cross-linking of L1-11A with a secondary antibody was necessary for significant inhibition of proliferation, indicating that cross-linking of L1 is required for the antiproliferative effect. L1-negative prostate carcinoma cells were not influenced by antibody treatment. Biweekly treatment of ovarian carcinoma-bearing mice with L1-11A led to a dose-dependent and significant reduction of tumor burden (up to -63.5%) and ascites formation (up to -75%). This effect was associated with reduced proliferation within the tumors. L1-directed antibody-based inhibition of peritoneal growth and dissemination of human ovarian carcinoma cells represents important proof-of-principle for the development of a new therapy against one of the leading gynecologic malignant diseases.

Efficient inhibition of intra-peritoneal tumor growth and dissemination of human ovarian carcinoma cells in nude mice by anti-L1-cell adhesion molecule monoclonal antibody treatment.

Die L1-Zelladh&auml;sionsmolek&uuml;l ist bei der Bek&auml;mpfung der Proliferation, Migration und Invasion der Tumor Zelle verschiedene in-vitro-verwickelt. Vor kurzem fand L1 eine &Uuml;berexpression mit Tumor Fortschreiten der Eierstock Karzinom, eine der h&auml;ufigsten Ursachen der krebsbedingten Todesf&auml;lle bei kologischen maligne Erkrankungen korrelieren. Um L1 als m&ouml;gliches Angriffsziel f&uuml;r Eierstock-Krebs-Therapie zu bewerten, untersuchten wir die Wirkungen von Anti-L1 monoklonalen Antik&ouml;rpern (chCE7 und L1-11A) auf die Proliferation und Migration von L1-positiven menschlichen SKOV3ip Eierstock-Karzinom Zellen in vitro und die therapeutische Wirksamkeit von L1-11A gegen i.p. SKOV3ip Tumorwachstum in den nude M&auml;usen. In vitro hemmte sowohl Anti-L1-Antik&ouml;rper effizient die Vermehrung von SKOV3ip Zellen sowie andere L1-Ausdr&uuml;cken Tumor Zelllinien (renal Carcinoma, Neuroblastom und Kolonkarzinom). Auf zwei Zelllinien musste hyper-Kreuz-Verkn&uuml;pfung der L1-11A mit einen Sekund&auml;rantik&ouml;rper signifikante Hemmung der Proliferation, darauf hinweist, dass die Vernetzung der L1 f&uuml;r die Antiproliferative Wirkung erforderlich ist. L1-negativen Prostata-Karzinom-Zellen wurden von Antik&ouml;rper-Behandlung nicht beeinflusst. Konservative Behandlung von Eierstock-Karzinom-Lager-M&auml;use mit L1-11A f&uuml;hrte zu eine dosisabh&auml;ngige und erhebliche Reduzierung der Tumor Belastung (bis zu-63.5 %) und die Bildung von Aszites (bis zu-75 %). Dieser Effekt war verbunden mit geringeren Verbreitung innerhalb der Tumoren. L1-gerichteten Antik&ouml;rper-basierte Hemmung der Peritonealdialyse Wachstum und Verbreitung der menschlichen Eierstock Karzinom-Zellen darstellt wichtigen Beweis der Grundsatz f&uuml;r die Entwicklung einer neuen Therapie gegen eine der f&uuml;hrenden gyn&auml;kologischen malignen Erkrankungen.

The human tissue kallikrein family of serine proteases (hK1-hK15 encoded by the genes KLK1-KLK15) is involved in several cancer-related processes. Accumulating evidence suggests that certain tissue kallikreins are part of an enzymatic cascade pathway that is activated in ovarian cancer and other malignant diseases. In the present study, OV-MZ-6 ovarian cancer cells were stably co-transfected with plasmids expressing hK4, hK5, hK6, and hK7. These cells displayed similar proliferative capacity as the vector-transfected control cells (which do not express any of the four tissue kallikreins), but showed significantly increased invasive behavior in an in vitro Matrigel invasion assay (p<0.01; Mann-Whitney U-test). For in vivo analysis, the cancer cells were inoculated into the peritoneum of nude mice. Simultaneous expression of hK4, hK5, hK6, and hK7 resulted in a remarkable 92% mean increase in tumor burden compared to the vector-control cell line. Five out of 14 mice in the 'tissue kallikrein overexpressing' group displayed a tumor/situs ratio greater than 0.198, while this weight limit was not exceeded at all in the vector control group consisting of 13 mice (p=0.017; chi2 test). Our results strongly support the view that tumor-associated overexpression of tissue kallikreins contributes to ovarian cancer progression.

Overexpression of the human tissue kallikrein genes KLK4, 5, 6, and 7 increases the malignant phenotype of ovarian cancer cells.

More effective treatment of patients with metastasizing osteosarcoma (OS) with a mean 5-year survival rate of <20% requires more detailed knowledge on the complex mechanisms of metastasis for the design of new drugs, which selectively target metastasizing cells. Moreover, novel diagnostic imaging technology for early detection of metastases is needed. Mouse models, which reproduce human metastasizing OS and allow visualization of single metastatic cells are instrumental for preclinical testing of new pharmaceuticals and diagnostic instruments. Here, the low metastatic Dunn cell line and its highly metastatic LM8 subline, both equipped with a constitutively expressed lacZ gene, were used to improve the well-established OS models in syngeneic C3H mice to achieve ex vivo visualization of single metastatic cells in affected organs by X-gal staining. These models, combined with a technique for in situ high quality lung tissue-maintaining perfusion revealed, as a novel finding, single metastasizing Dunn cells in lung and liver. Importantly, constitutive lacZ gene expression did not affect in vitro and in vivo tumorigenic and metastatic properties of Dunn and LM8 cells. Thus, these improved Dunn and LM8 OS mouse models will in the future serve as a benchmark for the development of new metastasis-targeting drugs and metastasis-imaging technology.

Eine effektivere Behandlung von Patienten mit metastasierenden Osteosarkom (OS) mit eine mittlere 5-Jahres-&Uuml;berlebensrate von &lt; 20 % erfordert detaillierte Kenntnisse &uuml;ber die komplexen Mechanismen der Metastasierung f&uuml;r die Gestaltung neuer Medikamente, die selektiv metastasierenden Zellen abzielen. Dar&uuml;ber hinaus ist die neuartige diagnostische imaging-Technologie f&uuml;r die Fr&uuml;herkennung von Metastasen erforderlich. Maus-Modelle, die menschliche metastasierenden OS zu reproduzieren und erm&ouml;glichen die Visualisierung von metastasierendem Einzelzellen sind ma&szlig;geblich f&uuml;r pr&auml;klinische Tests von neuen Pharma und Diagnostik-Instrumente. Hier wurden die niedrigen metastasierendem Dunn-Zelllinie und seine h&ouml;chst metastasierendem LM8-Subline, beide ausgestattet mit einem konstitutiv exprimierten LacZ Gen, zur verbessern die etablierten OS-Modelle im syngenen C3H M&auml;use, ex Vivo Visualisierung von metastasierendem Einzelzellen in betroffenen Organen zu erreichen von X-gal-F&auml;rbung. Diese Modelle, kombiniert mit einer Technik f&uuml;r in-situ hochwertige Lunge Gewebe-erhaltende Perfusion aufgedeckt, als Roman finden, metastasierenden Dunn Einzelzellen in Lunge und Leber. Wichtiger ist, konstitutive LacZ Gen-Ausdrucks nicht in-vitro und in vivo tumorigenic und metastasiertem Eigenschaften von Dunn und LM8 Zellen beeinflussen. So diese verbessert Dunn und LM8 OS Mausmodelle dient in Zukunft als Ma&szlig;stab f&uuml;r die Entwicklung neuer Metastasierung Zielversionen Medikamente und Metastasierung-Imaging-Technologie.

Metastasizing osteosarcoma has a mean 5-year survival rate of only 20% to 30%. Therefore, novel chemotherapeutics for more effective treatment of this disease are required.

Antimetastatic Aktivit&auml;t von Honokiol in Osteosarkom.

Osteosarcoma (OS) is the most frequent primary bone tumor. Despite multiagent neoadjuvant chemotherapy, patients with metastatic disease have a poor prognosis. Moreover, currently used chemotherapeutics have severe toxic side effects. Thus, novel agents with improved antimetastatic activity and reduced toxicity are needed. Taurolidine, a broad-spectrum antimicrobial, has recently been shown to have antineoplastic properties against a variety of tumors and low systemic toxicity. Consequently, we investigated in our study the antineoplastic potential of taurolidine against OS in two different mouse models. Although both OS cell lines, K7M2 and LM8, were sensitive for the compound in vitro, intraperitoneal application of taurolidine failed to inhibit primary tumor growth. Moreover, it enhanced the metastatic load in both models 1.7- to 20-fold and caused severe liver deformations and up to 40% mortality. Thus, systemic toxicity was further investigated in tumor-free mice histologically, by electron microscopy and by measurements of representative liver enzymes. Taurolidine dose-dependent fibrous thickening of the liver capsule and adhesions and atrophies of the liver lobes were comparable in healthy and tumor-bearing mice. Liver toxicity was further indicated by up to eightfold elevated levels of the liver enzymes alanine transaminase, aspartate transaminase and GLDH in the circulation. Ultrastructural analysis of affected liver tissue showed swollen mitochondria with cristolysis and numerous lipid vacuoles in the cytoplasm of hepatocytes. The findings of our study question the applicability of taurolidine for OS treatment and may suggest the need for caution regarding the widespread clinical use of taurolidine as an antineoplastic agent.

Das Osteosarkom (OS) ist der h&auml;ufigste prim&auml;re Knochentumor. Trotz Multiagenten Neoadjuvante Chemotherapie haben Patienten mit Metastasen eine schlechte Prognose. Dar&uuml;ber hinaus haben die aktuell verwendeten Chemotherapeutika toxischen Nebenwirkungen. Daher sind die neue Wirkstoffen mit verbesserter antimetastatic Aktivit&auml;t und verminderte Toxizit&auml;t erforderlich. Taurolidine, eine Breitspektrum antimikrobielle hat k&uuml;rzlich gezeigt, Antineoplastische Eigenschaften gegen eine Vielzahl von Tumoren und geringe systemische Toxizit&auml;t aufweist. Daher untersuchten wir in unserer Studie das Antineoplastische Potenzial der Taurolidine gegen OS in zwei verschiedene Maus-Modellen. Obwohl beide OS-Zell-Linien, K7M2 und LM8, waren empfindlich f&uuml;r das in-vitro-Compound, intraperitoneale Anwendung von Taurolidine konnte Prim&auml;rtumorwachstum hemmen. &Uuml;berdies verbessert es die metastasierendem Last in beide Modelle 1.7 und 20 8-fach und verursachte schwere Leber Verformungen und bis zu 40 % Mortalit&auml;t. So wurde systemische Toxizit&auml;t weiter im Tumor-freie M&auml;use histologisch durch Elektronenmikroskopie und durch Messungen der repr&auml;sentativen Leberenzyme untersucht. Taurolidine Dosis-abh&auml;ngige fibr&ouml;se Verdickung der Leber Kapsel und Adh&auml;sionen und verk&uuml;mmert der Leber Lappen waren bei gesunden und Tumor-tragenden M&auml;usen vergleichbar. Lebertoxizit&auml;t wurde weiter durch bis zu Achtfache erh&ouml;hte Werte der Leberenzyme Alanin-Aminotransferase, Aspartat-Aminotransferase und GLDH im Kreislauf angezeigt. Ultrastrukturelle Analyse der betroffenen Lebergewebe zeigte geschwollene Mitochondrien mit Cristolysis und zahlreichen Lipid-Vakuolen im Zytoplasma der Hepatozyten. Die Ergebnisse unserer Studie die Anwendbarkeit der Taurolidine f&uuml;r OS-Behandlung in Frage und k&ouml;nnen die Notwendigkeit der Vorsicht in Bezug auf den weitverbreiteten klinischen Gebrauch von Taurolidine als Antineoplastische Vertreter vorschlagen.
