Sod2-/- mice, which are deficient in the mitochondrial form of superoxide dismutase (MnSOD), have a short survival time that is strongly affected by genetic background. This suggests the existence of genetic modifiers that are capable of modulating the degree of mitochondrial oxidative damage caused by the MnSOD deficiency, thereby altering longevity. To identify these modifier(s), we generated recombinant congenic mice with quantitative trait loci (QTL) containing the putative genetic modifiers on the short-lived C57BL/6J genetic background. MnSOD deficient C57BL/6J mice with a QTL from the distal region of chromosome 13 from DBA/2J were able to survive for as long as those generated on the long-lived DBA/2J background. Within this region, the gene encoding nicotinamide nucleotide transhydrogenase (Nnt) was found to be defective in C57BL/6J mice, and no mature NNT protein could be detected. The forward reaction of NNT, a nuclear-encoded mitochondrial inner membrane protein, couples the generation of NADPH to proton transport and provides NADPH for the regeneration of two important antioxidant compounds, glutathione and thioredoxin, in the mitochondria. This action of NNT could explain its putative protective role in MnSOD-deficient mice.

Genetic modifiers of the phenotype of mice deficient in mitochondrial superoxide dismutase.

Sod2-/-M&auml;usen, die einen Mangel an mitochondrialen Form von Superoxiddismutase (MnSOD), haben eine kurze &Uuml;berlebenszeit, die stark von genetischen Hintergrund beeinflusst wird. Innerhalb dieser Region gen f&uuml;r Nicotinamid Nukleotid Transhydrogenase (Nnt) wurde festgestellt, dass in den M&auml;usen C57BL/6J defekt sein und kein Reifen NNT-Protein nachgewiesen werden konnte. Konstruierten wir einen bacterial artificial Chromosome (BAC)-basierte physische Karte der Chromosomen 2 und 3 von Drosophila Melanogaster, die 81 % des Genoms darstellen. We constructed a bacterial artificial chromosome (BAC)-based physical map of chromosomes 2 and 3 of Drosophila melanogaster, which constitute 81% of the genome. Sequence tagged site (STS) content, restriction fingerprinting, and polytene chromosome in situ hybridization approaches were integrated to produce a map spanning the euchromatin. Three of five remaining gaps are in repeat-rich regions near the centromeres. A tiling path of clones spanning this map and STS maps of chromosomes X and 4 was sequenced to low coverage; the maps and tiling path sequence were used to support and verify the whole-genome sequence assembly, and tiling path BACs were used as templates in sequence finishing.

A BAC-based physical map of the major autosomes of Drosophila melanogaster.

A moderate resolution single nucleotide polymorphism (SNP) map of the genome of Drosophila melanogaster that is designed for use in quantitative genetic mapping is described. Seventeen approximately 500 nucleotide gene sequences spaced at 10 to 20 centimorgan intervals were combined with 49 shorter sequence tag sites (STSs) at 5 to 10 centimorgan intervals to generate a map that should not leave any gaps greater than one half of a chromosome arm when any two wild type lines are compared. Of 20 markers with sufficient polymorphism to construct haplotype cladograms, 13 showed evidence for two divergent classes of haplotype. The possible mechanisms for and implications of the unexpected finding that two thirds of all short gene sequences in D. melanogaster may be dimorphic are discussed, including the suggestion that admixture between two separate lineages may have been a major event in the history of the species.

For nearly a century, genetic analysis in Drosophila melanogaster has been a powerful tool for analyzing gene function, yet Drosophila lacks the molecular genetic mapping tools that recently have revolutionized human, mouse, and plant genetics. Here, we describe the systematic characterization of a dense set of molecular markers in Drosophila by using a sequence tagged site-based physical map of the genome. We identify 474 biallelic markers in standard laboratory strains of Drosophila that span the genome. Most of these markers are single nucleotide polymorphisms and sequences for these variants are provided in an accessible format. The average density of the new markers is one per 225 kb on the autosomes and one per megabase on the X chromosome. We include in this survey a set of P-element strains that provide additional use for high-resolution mapping. We show one application of the new markers in a simple set of crosses to map a mutation in the hedgehog gene to an interval of <1 Mb. This new map resource significantly increases the efficiency and resolution of recombination mapping and will be of immediate value to the Drosophila research community.

Seit fast einem Jahrhundert wurde die genetische Analyse in Drosophila Melanogaster ein leistungsf&auml;higes Werkzeug f&uuml;r die Analyse der Genfunktion, noch Drosophila fehlt die molekulare genetische Mapping-Tools, die vor kurzem menschliche revolutioniert haben, Maus und Pflanzengenetik. Hier beschreiben wir die systematische Charakterisierung einer dichten Reihe von molekularer Marker in Drosophila, mithilfe einer Sequenz markierte standortbasierte physische Karte des Genoms. Wir identifizieren 474 Biallelic Markierungen im standard Labor St&auml;mme von Drosophila, die das Genom zu erstrecken. Die meisten dieser Marker sind Einzel-Nukleotid-Polymorphismen und Sequenzen f&uuml;r diese Varianten werden in einem barrierefreien Format bereitgestellt. Single Nucleotide Polymorphismus Marker f&uuml;r genetische Zuordnung in Drosophila Melanogaster.

To understand the role of oxidative stress and mitochondrial defects in the development of neurodegeneration, we examined the age-related pathological changes and corresponding gene expression profiles in homozygous mutant mice deficient in the mitochondrial form of superoxide dismutase (MnSOD, SOD2). These Sod2-/- mice, generated on a B6D2F1 background, developed ataxia at Postnatal Day (P) 11 and progressively deteriorated with frequent seizures by P14. Histopathological examination revealed neurodegenerative changes consistent with the neurological signs. Vacuolar degeneration was observed in neurons and neuropil throughout the brainstem and rostral cortex. The motor trigeminal nucleus in brainstem and the deeper layers of the motor cortex were the earliest regions to degenerate, with the thalamus and hippocampus affected at later stages. Oligonucleotide microarrays were used to compare gene expression profiles in the brainstem and thalamus of Sod2+/+ and -/- mice from birth to P18. Notably, a large set of heat-shock protein genes was transcriptionally down regulated, and this was most likely due to a reduction in the heat-shock transcription factor 1 (HSF1). Other major classes of differentially expressed genes include lipid biosynthesis and ROS metabolism.

Um die Rolle des oxidativen Stress und mitochondriale Defekte in der Entwicklung der Neurodegeneration zu verstehen, haben wir die altersbedingte krankhafte Ver&auml;nderungen und entsprechenden Gen-Ausdruck-Profile in homozygote Mutante M&auml;use einen Mangel an mitochondrialen Form von Superoxiddismutase (MnSOD, SOD2) gepr&uuml;ft. Diese Sod2-/-M&auml;usen, generiert auf B6D2F1 Hintergrund, entwickelt Ataxie bei postnatalen Tag (P) 11 und schrittweise Verschlechterung mit h&auml;ufigen Anf&auml;llen von P14. Histopathologische Untersuchung ergab Neurodegenerative Ver&auml;nderungen mit neurologischen Symptomen. Insbesondere ein gro&szlig;er Satz von Hitzeschock Protein-gen war transcriptionally nach unten geregelt, und dies wurde wahrscheinlich durch eine Reduzierung der Hitze-Schock-Transkriptionsfaktor 1 (HSF1). Anderen Hauptgruppen von differentiell exprimierten Genen geh&ouml;ren Lipid-Biosynthese und ROS-Stoffwechsel.

Selektive neuronalen Verwundbarkeit und unzureichende Stressantwort in Superoxid-Dismutase-mutant-M&auml;use.

To identify biomarkers associated with the development of hepatocellular carcinoma (HCC) in CuZn superoxide dismutase (CuZnSOD, Sod1) deficient mice, 2-DE followed by MS analysis was carried out with liver samples obtained from 18-month-old Sod1-/- and +/+ mice. The intracellular Ca binding protein, regucalcin (RGN), showed a divergent alteration in Sod1-/- samples. Whereas elevated RGN levels were observed in -/- samples with no obvious neoplastic changes, marked reduction in RGN was observed in -/- samples with fully developed HCC. GST mu1 (GSTM1), on the other hand, showed a significant increase only in the neoplastic regions obtained from Sod1-/- livers. No change in GSTM1 was observed in the surrounding normal tissues. Marked reduction was observed in two intracellular lipid transporters, fatty acid binding protein 1 (FABP1) and major urinary protein 11 and 8 (MUP 11&8), in Sod1-/- samples. Analysis of additional samples at 18-22 months of age showed a three-fold increase in enolase activities in Sod1-/- livers. Consistent with previous findings, carbonic anhydrase 3 (CAIII) levels were significantly reduced in Sod1-/- samples, and immunohistochemical analysis revealed that the reduction was not homogenous throughout the lobular structure in the liver.

Identification of biomarkers associated with the development of hepatocellular carcinoma in CuZn superoxide dismutase deficient mice.

Zur Identifizierung von Biomarkern, die die Entwicklung des hepatozellul&auml;ren Karzinom (HCC) in CuZn Superoxid-Dismutase (CuZnSOD, Sod1) unzul&auml;ngliche M&auml;usen, 2-DE gefolgt von MS zugeordnet wurden untersucht mit Leber Proben aus 18 Monate alten Sod1-/- und + / + M&auml;use. Die intrazellul&auml;re Ca verbindliches Protein, Regucalcin (RGN), zeigte eine abweichende Ver&auml;nderung im Sod1-/-Samples. W&auml;hrend die erh&ouml;hten RGN in beobachtet wurden- / - samples ohne offensichtliche neoplastischen &Auml;nderungen, deutliche Reduktion RGN in festgestellt- / - samples mit voll entwickelten HCC. GST mu1 (GSTM1), auf der anderen Seite zeigte eine deutliche Steigerung nur in neoplastischen Regionen Sod1-/-Lebern entnommen. Keine &Auml;nderung im GSTM1 wurde in das umliegende normale Gewebe beobachtet. Deutliche Reduktion wurde in zwei intrazellul&auml;re Lipid-Transporter, Fetts&auml;ure-verbindliches Protein 1 (FABP1) und gro&szlig;e Urin Protein 11 und 8 (MUP 11 &amp; 8), in Sod1-/-Samples beobachtet. Weitere Beispiele bei 18-22 Monate alt zeigte eine Verdreifachung der Zahl der Enolase-Aktivit&auml;ten im Sod1-/-Lebern. Fr&uuml;here Ergebnisse, kohlenstoffhaltige Anhydrase 3 entsprechen (CAIII) wurden deutlich reduziert im Sod1-/-Samples und Immunohistochemical Analyse ergab, dass die Reduzierung nicht homogen im gesamten lobul&auml;res Struktur in der Leber.
