The mechanisms of the salutary effects of ECP are poorly understood, but appear to involve regulation of T-cell-mediated alloreactive responses, possibly by induction of regulatory T cells. We created a mouse model of ECP to determine the effects of ECP on T-cell responses in vivo and the contribution of CD4(+)CD25(+) T cells.

Heme oxygenase-1 (HO-1) catalyzes the conversion of heme into carbon monoxide (CO), iron, and biliverdin. In preliminary studies, we observed that the absence of HO-1 in aortic allograft recipients resulted in 100% mortality within 4 days due to arterial thrombosis. In contrast, recipients normally expressing HO-1 showed 100% graft patency and survival for more than 56 days. Abdominal aortic transplants were performed using Balb/cJ mice as donors and either HO-1(+/+) or HO-1(-/-) (C57BL/6xFVB) mice as recipients. Light and electron microscopy revealed extensive platelet-rich thrombi along the entire length of the graft in HO-1(-/-) recipients at 24 hours. Treatment of recipients with CORM-2, a CO-releasing molecule (10 mg/kg of body weight intravenously), 1 hour prior and 1, 3, and 6 days after transplantation, significantly improved survival (62% at >56 days, P < 0.001) compared with HO-1(-/-) recipients treated with inactive CORM-2 (median survival 1 day). Histological analyses revealed that CO treatment markedly reduced platelet aggregation within the graft. Adoptive transfer of wild-type platelets to HO-1(-/-) recipients also conferred protection and increased survival. Aortic transplants from either HO-1(-/-) or HO-1(+/+) C57BL/6 donors into HO-1(+/+) (Balb/cJ) mice did not develop arterial thrombosis, surviving more than 56 days. These studies demonstrate an important role for systemic HO-1/CO for protection against vascular arterial thrombosis in murine aortic allotransplantation.

Carbon monoxide rescues heme oxygenase-1-deficient mice from arterial thrombosis in allogeneic aortic transplantation.

Histologische Analysen ergaben, dass CO-Behandlung die Thrombozytenaggregation innerhalb der Graft deutlich reduziert. Overexpression of transforming growth factor-beta1 (TGF-beta1) and its downstream molecules in the rhabdomyosarcoma (RMS) RD cell line has been reported previously, but the regulatory role of TGF-beta1 on RMS has not been studied extensively. In the present study, we showed that expression of TGF-beta1 and its downstream molecules type II TGF-beta receptor (TbetaRII) and Smad4 was significantly higher in RMS than in normal skeletal muscle, and there was a significant relationship between TGF-beta1 expression and histological grade. Gene silencing with TGF-beta1 short-hairpin RNA (shRNA)-expressing vectors significantly decreased the growth of RD cells, which was confirmed by caspase-3 (in vitro) and TUNEL (in vivo) assays. Moreover, a proportion of treated rhabdomyosarcoma (RD) cells changed to a round shape from the normal fusiform or polygonal shape and expressed myofilaments. Myogenin is one of the myogenic differentiation genes (MyoD) family of myogenic regulators, and was obviously higher in TGF-beta1-shRNA-treated tumors than it in control at the mRNA and protein level. Immunohistochemical staining with myogenic differentiation markers such as myosin and desmin in subcutaneous RMS tissue showed that TGF-beta1 shRNA increased staining for myosin. These results provide new insight into the biological function of TGF-beta1 in malignant tumors, and imply that the TGF-beta1 signal pathway is a potential therapeutic target for drugs that induce differentiation of RMS.

Early pancreatic cancer response following cetuximab and/or irinotecan therapies was measured by serial dynamic contrast-enhanced magnetic resonance imaging (DCE-MRI) before and during therapy. Groups 1 to 4 (n â€Š=â€Š 6/group) of SCID mice bearing orthotopic pancreatic adenocarcinoma xenografts expressing luciferase were treated with phosphate-buffered saline, cetuximab, irinotecan, or cetuximab combined with irinotecan, respectively, twice weekly for 3 weeks. DCE-MRI was performed on days 0, 1, 2, and 3 after therapy initiation, whereas anatomic magnetic resonance imaging was performed on days 0, 1, 2, 3, 6, and 13. Bioluminescence imaging was performed on days 0 and 21. At day 21, all tumors were collected for further histologic analyses (Ki-67 and CD31 staining), whereas tumor dimensions were measured by calipers. The Ktrans values in the 0.5 mm-thick peripheral tumor region were calculated, and the changes in Ktrans during the 3 days posttherapy were compared to tumor volume changes, bioluminescent signal changes, and histologic findings. The Ktrans changes in the peripheral tumor region after 3 days of therapy were linearly correlated with 21-day decreases in tumor volume (p < .001), bioluminescent signal (p â€Š=â€Š .050), microvessel densities (p â€Š=â€Š .002), and proliferating cell densities (p â€Š=â€Š .001). This study supports the clinical use of DCE-MRI for pancreatic cancer patients for early assessment of an anti-epidermal growth factor receptor therapy combined with chemotherapy.

Early therapy evaluation of combined cetuximab and irinotecan in orthotopic pancreatic tumor xenografts by dynamic contrast-enhanced magnetic resonance imaging.

Recently, strong interest has been focused on immunostimulants to reducing the diseases in shrimp aquaculture. However, information regarding to the related immune-enhancing proteins in shrimps is not available yet. In this study, vitamin C (Vc), Chinese herbs (CH), and the mixture of vitamin C and Chinese herbs (Mix) were tested for their enhancement on shrimp's immune activity. Compared with those in the control group, values of phenoloxidase (PO), superoxide dismutase (SOD) and antibacterial (Ua) activity in the Mix-treated group were improved significantly 12 or 24 days after the treatment. The cumulative mortality was also lower in the Mix-treated group after infection with Vibrio parahemolyticus. Furthermore, comparative proteomic approach was used to assess the protein expression profile in shrimps. Approximately 220-290 and 300-400 protein spots were observed in the 2-DE gels. Among them, 29 and 28 altered proteins from hemocytes and hepatopancreas, respectively, were subjected to matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF/MS) analysis. The results revealed that the main altered proteins showed high homologies with Litopenaeus vannamei hemocyanin, hemolymph clottable protein, hemoglobin beta, cytosolic MnSOD, trypsin, cathepsin I(L) and zinc proteinase Mpc1. Together, these studies found Vc and CH were suitable immunostimulants to shrimp L. vannamei, and 7 altered proteins could be involved in the enhanced immune activities.

Toll/Toll-like receptors (TLRs), one of the most important pattern recognition receptors (PRRs), play a crucial role in innate immune responses in both invertebrates and vertebrates. In this study, we cloned and characterised a Toll gene from Scylla paramamosain (SpToll). Bioinformatic analysis predicted that SpToll contained one open reading frame of 3018bp and encoded a single-pass transmembrane domain protein of 1005 amino acids. Further, SpToll could be clustered into one branch along with other arthropod Tolls in a phylogenetic tree. SpToll transcripts could be detected by RT-PCR from all tissues examined including the heart, gill, hepatopancreas, stomach, intestine, muscle, eyestalk and hemocytes. Infection by Vibrio parahemolyticus up-regulated SpToll mRNA expression in hemocytes after 48h. The profile of single nucleotide polymorphisms (SNPs) in the leucine-rich repeats (LRRs) domain of SpToll in three healthy crabs was then evaluated. Two hundred and twenty SNPs with a frequency of about 1.0-4.0% were identified in hemocyte DNA/cDNA. Surprisingly, the adenine to guanine transition at position 1372 (c.1372A>G) had a frequency of about 50%. Finally, the results showed that challenge with V. parahemolyticus stimulated the appearance of two sets of SNPs in crabs. More importantly, the c.1372A>G mutation could contribute to a low mortality after V. parahemolyticus infection and introduce variation of charge and secondary structure into the SpToll polypeptide. In summary, these studies suggested a novel Toll homologue in crab and identified a SNP with potential pathogen-resistant activities. The result will be important for the investigation of crab immune defense mechanisms.

In dieser Studie haben wir eine Variable Region in der C-Terminus von H&auml;mocyanin aus Garnelen Litopenaeus (2288-2503bp, HcSC) von Sequenzalignments identifiziert. Insgesamt 13 SNPs wurden durch PCR-SSCP und HcSC Klon Sequenzierung identifiziert. Die SSCP-Muster des HcSC konnte in Vibro Parahaemolyticus-behandelten Garnelen moduliert werden. Ein Roman SSCP Band mit vier Standorten in SNP wurde V. Parahaemolyticus-resistenten Garnelen identifiziert. Noch wichtiger ist, drei von diesen vier SNPs Variationen in Aminos&auml;uresequenz und m&ouml;glicherweise Sekund&auml;rstruktur des HcSC-Polypeptid eingef&uuml;hrt und f&uuml;hrte zu einer h&ouml;heren Agglutinierende Aktivit&auml;t gegen sieben pathogenen Bakterien. Diese Ergebnisse legen nahe, dass der C-Terminus von Garnelen L. H&auml;mocyanin SNPs, besitzt, die im Zusammenhang mit Garnelen-Resistenz gegen verschiedene Krankheitserreger sein kann.

In this study, we identified a variable region in the C-terminus of hemocyanin from the shrimp Litopenaeus vannamei (2288-2503bp, HcSC) by sequence alignments. A total of 13 SNPs were identified by PCR-SSCP and HcSC clone sequencing. The SSCP patterns of HcSC could be modulated in Vibro parahaemolyticus-treated shrimps. A novel SSCP band with four SNP sites was identified in V. parahaemolyticus-resistant shrimps. More importantly, three of these four SNPs introduced variations in amino acid sequence and possibly secondary structure of the HcSC polypeptide and resulted in a higher agglutinative activity against seven pathogenic bacteria. These results suggest that the C-terminus of shrimp L. vannamei hemocyanin possesses SNPs, which may be related to shrimp resistance to different pathogens.

Acupuncture has long been employed for the treatment of chronic low back pain (CLBP). However, very few studies have characterized the effectiveness of the different acupuncture modes for CLBP.

Although short-term outcomes in kidney transplantation have improved dramatically, long-term survival remains a major challenge. A key component of long-term, chronic allograft injury in solid organ transplants is arteriosclerosis characterized by vascular neointimal hyperplasia and inflammation. Establishing a model of this disorder would provide a unique tool not only to identify mechanisms of disease but also to test potential therapeutics for late graft injury. To this end, we utilized a mouse orthotopic renal transplant model in which C57BL/6J (H-2b) recipients were given either a kidney allograft from a completely mismatched Balb/cJ mouse (H-2d) or an isograft from a littermate. A unilateral nephrectomy was performed at the time of transplant followed by a contralateral nephrectomy on post-transplant day 7. Recipients were treated with daily cyclosporine subcutaneously for 14 days and then studied 8 and 12 weeks post transplantation. Renal function was significantly worse in allograft compared with isograft recipients. Moreover, the allografts had significantly more advanced tubulointerstitial fibrosis and profound vascular disease characterized by perivascular leukocytic infiltration and neointimal hyperplasia affecting the intrarenal blood vessels. Thus, we describe a feasible and reproducible murine model of intrarenal transplant arteriosclerosis that is useful to study allograft vasculopathy.

Ein Modell dieser St&ouml;rung br&auml;chte ein einzigartiges Werkzeug, nicht nur um Mechanismen der Krankheit zu identifizieren, sondern auch potenzielle Therapeutika zur sp&auml;ten Transplantat Verletzungen zu testen. Zu diesem Zweck verwendet wir einem Mausmodell Orthotopic renal Transplantation welche C57BL/6J (H-2b) Empf&auml;nger entweder eine Niere Allograft &uuml;ber eine v&ouml;llig nicht &uuml;bereinstimmende Balb/cJ-Maus (H-2d) oder ein Isograft aus einer Littermate erhielten. Eine einseitige Nephrektomie war zum Zeitpunkt der Transplantation, gefolgt von einer kontralateralen Nephrektomie am nach der Transplantation Tag 7 durchgef&uuml;hrt. Empf&auml;nger mit t&auml;glichen Cyclosporin subkutan f&uuml;r 14 Tage behandelt wurden und studierte dann 8 und 12 Wochen Post-Transplantation. Nierenfunktion war deutlich schlechter in Allograft gegen&uuml;ber Isograft Empf&auml;nger. Dar&uuml;ber hinaus hatte die Herztransplanate deutlich tubulointerstitielle Fibrose und profunde vaskul&auml;re Krankheit, charakterisiert durch perivaskul&auml;ren leukocytic Infiltration und Neointimal Hyperplasie beeinflussen die intrarenal Blutgef&auml;&szlig;e erweiterte. Daher beschreiben wir eine durchf&uuml;hrbar und reproduzierbar murinen Modell intrarenal Transplantation Arteriosklerose, die hilfreich, Allograft Vaskulopathie zu studieren.
