Anti-apoptotic Bcl2 family proteins such as Bcl-x(L) protect cells from death by sequestering apoptotic molecules, but also contribute to normal neuronal function. We find in hippocampal neurons that Bcl-x(L) enhances the efficiency of energy metabolism. Our evidence indicates that Bcl-x(L)interacts directly with the Î²-subunit of the F(1)F(O)Â ATP synthase, decreasing an ion leak within the F(1)F(O) ATPase complex and thereby increasing net transport of H(+) by F(1)F(O) during F(1)F(O) ATPase activity. By patch clamping submitochondrial vesicles enriched in F(1)F(O) ATP synthase complexes, we find that, in the presence of ATP, pharmacological or genetic inhibition of Bcl-x(L) activity increases the membrane leak conductance. In addition, recombinant Bcl-x(L) protein directly increases the level of ATPase activity of purified synthase complexes, and inhibition of endogenous Bcl-x(L) decreases the level of F(1)F(O) enzymatic activity. Our findings indicate that increased mitochondrial efficiency contributes to the enhanced synaptic efficacy found in Bcl-x(L)-expressingÂ neurons.

In Neuronen finden wir, dass Bcl-x(L) die Effizienz des Energiestoffwechsels erh&ouml;ht. Unsere Beweise gibt, dass Bcl-X (L) direkt mit der &beta;-Untereinheit des F(1)F(O) ATP-Synthase, interagiert, sinkende ein Ion-Leck in der F(1)F(O)-ATPase komplexe und wodurch net Transport von H(+) von F(1)F(O) w&auml;hrend der F(1)F(O) ATPase-Aktivit&auml;t. Von Patch spannen Submitochondrial Bl&auml;schen angereichertes F(1)F(O) ATP-Synthase-komplexe finden wir, dass in Gegenwart von ATP, pharmakologische oder genetische Hemmung der Bcl-x(L) Aktivit&auml;t den Membran-Leck-Leitwert erh&ouml;ht. Dar&uuml;ber hinaus rekombinantes Bcl-x(L) Protein erh&ouml;ht direkt ATPase-Aktivit&auml;t des gereinigten Synthase-komplexe und Hemmung der endogenen Bcl-x(L) verringert F(1)F(O) enzymatische Aktivit&auml;t. Unsere Ergebnisse zeigen, dass die mitochondriale Effizienzsteigerung auf die verbesserte synaptische Wirksamkeit gefunden in Bcl-X (L) tr&auml;gt-Neuronen zum Ausdruck zu bringen.

Cellular stress or injury can result in mitochondrial dysfunction, which has been linked to many chronic neurological disorders including amyotrophic lateral sclerosis (ALS) and Parkinson's disease (PD). Stressed and dysfunctional mitochondria exhibit an increase in large conductance mitochondrial membrane currents and a decrease in bioenergetic efficiency. Inefficient energy production puts cells, and particularly neurons, at risk of death when energy demands exceed cellular energy production. Here we show that the candidate ALS drug dexpramipexole (DEX; KNS-760704; ((6R)-4,5,6,7-tetrahydro-N6-propyl-2,6-benzothiazole-diamine) and cyclosporine A (CSA) inhibited increases in ion conductance in whole rat brain-derived mitochondria induced by calcium or treatment with a proteasome inhibitor, although only CSA inhibited calcium-induced permeability transition in liver-derived mitochondria. In several cell lines, including cortical neurons in culture, DEX significantly decreased oxygen consumption while maintaining or increasing production of adenosine triphosphate (ATP). DEX also normalized the metabolic profile of injured cells and was protective against the cytotoxic effects of proteasome inhibition. These data indicate that DEX increases the efficiency of oxidative phosphorylation, possibly by inhibition of a CSA-sensitive mitochondrial conductance.

Effects of dexpramipexole on brain mitochondrial conductances and cellular bioenergetic efficiency.

Betont und gest&ouml;rte Mitochondrien zeigen eine Zunahme der gro&szlig;en Leitwert mitochondriale Membran Str&ouml;mungen und eine Abnahme der bioenergetische Effizienz. Ineffizienten Energieerzeugung gef&auml;hrdet Zellen und vor allem Neuronen des Todes beim Energiebedarf zellul&auml;ren Energieproduktion &uuml;berschreiten. Hier zeigen wir, dass der Kandidat ALS Droge Dexpramipexole (DEX; KNS-760704; ((6R)-4,5,6,7-tetrahydro-N6-propyl-2,6-benzothiazole-diamine) und Cyclosporin A (CSA) gehemmt erh&ouml;ht in Ionen Leitwert in ganze Ratte Brain-derived Mitochondrien induziert durch Kalzium oder Behandlung mit einem Proteasom-Inhibitor, obgleich nur CSA Kalzium-induzierte Permeabilit&auml;t &Uuml;bergang in Leber abgeleitete Mitochondrien gehemmt. In mehreren Zelllinien, kortikale Neuronen in der Kultur einschlie&szlig;lich, sank DEX deutlich Sauerstoffverbrauch beim Erhalt oder Ausbau der Produktion von Adenosintriphosphat (ATP). DEX auch das metabolische Profil der verletzten Zellen normalisiert und Schutzfunktion gegen den zytotoxischen Effekt Proteasom-Hemmung war. Transient global ischemia in rats induces delayed death of hippocampal CA1 neurons. Early events include caspase activation, cleavage of anti-death Bcl-2 family proteins and large mitochondrial channel activity. However, whether these events have a causal role in ischemia-induced neuronal death is unclear. We found that the Bcl-2 and Bcl-x(L) inhibitor ABT-737, which enhances death of tumor cells, protected rats against neuronal death in a clinically relevant model of brain ischemia. Bcl-x(L) is prominently expressed in adult neurons and can be cleaved by caspases to generate a pro-death fragment, Î”N-Bcl-x(L). We found that ABT-737 administered before or after ischemia inhibited Î”N-Bcl-x(L)-induced mitochondrial channel activity and neuronal death. To establish a causal role for Î”N-Bcl-x(L), we generated knock-in mice expressing a caspase-resistant form of Bcl-x(L). The knock-in mice exhibited markedly reduced mitochondrial channel activity and reduced vulnerability to ischemia-induced neuronal death. These findings suggest that truncated Bcl-x(L) could be a potentially important therapeutic target in ischemic brain injury.

Diese Forschungsergebnisse legen nahe, dass abgeschnittene Bcl-x(L) potenziell wichtige therapeutische Ziel in isch&auml;mische Gehirnverletzung sein k&ouml;nnte.
