Primary Sjogren's syndrome (pSS) is a slowly progressive, systemic autoimmune disorder characterized by gradual lymphocytic infiltration of exocrine glands. However, the spatially profiling the immune microenvironment in pSS is largely unclear, limiting the understanding of the complex interplay among cells within the microenvironment. Based on imaging mass cytometry (IMC) analysis of clinical pSS samples, we first revealed that labial salivary gland (LSG) comprised of epithelial, immune cells and stromal cells, and epithelial was the main cell type in LSG. Eight immune cells populations were identified, including CD8 T, CD4 T, Treg, B, NK cells, neutrophils, resident macrophages and a mixed immune cell cluster. We found that CD8 T cells, but not CD4 T cells, were the most prominent T cells in immune infiltrates of pSS LSG. With the increase of pSS disease activity and severity, the infiltration abundance of CD8 T cells gradually increased and was accompanied by the activation of inflammatory response. sc-RNA-seq analysis based on the GSE272409 dataset confirmed that CD8 T cells were the main immune cells, and dominated the most intercellular ligand-receptor interactions. CD8 T cells were further clustered into five cell subsets, of which CD160CD8 T cells subset appeared to present only in pSS patients. Further experiments demonstrated that CD160 expression on CD8 T cells was associated with an enhanced expression of proinflammatory and cytotoxic cytokines IFN-γ, GZMB and TNF-α, and the injury of salivary gland epithelial cells. Besides, proportion of GZMKCD8 T cells subset was increased in pSS patients. Trajectory analysis confirmed an enhanced frequency of CD8 T cell differentiation and activation during the progression of pSS. This study provided single cell profile with spatial information for analyzing the LSG immune microenvironment in pSS, which could not be achieved by conventional immunofluorescence and immunohistochemistry assays.

Application of imaging mass cytometry for spatially profiling the microenvironment of salivary glands in primary Sjögren's syndrome.

The role of MDM Two Binding Protein (MTBP) in tumor regulation is controversial; METHODS: We gathered MTBP's genomic location and GTEx expression data from University of California Santa Cruz (UCSC) and utilized the Human Protein Atlas (HPA) to evaluate its expression. Survival prognosis and immune infiltration analysis were meticulously evaluated by downloading and processing TCGA RNAseq data. Genetic alterations in MTBP were explored using cBioPortal; (3) Our findings revealed a prevailing trend of MTBP overexpression in numerous cancer types, associating with adverse prognoses and implicating MTBP as an oncogenic facilitator. Notably, in certain cancers, reduced MTBP expression correlated with improved patient survival, reflecting a tumor suppressive function of MTBP. By analyzing the tumor immunological environment, we observed positive correlations between MTBP and specific T helper cell subsets, suggesting its potential role in immune evasion strategies. Phosphorylation emerged as a pivotal post-translational modification regulating MTBP's function, influencing DNA replication process frequently exploited by cancer cells. Functional enrichment analyses highlighted MTBP's involvement in nuclear division, organelle fission, and cell cycle regulation, reinforcing its centrality in tumor proliferation and differentiation; CONCLUSIONS: Our results position MTBP as a critical regulator of cancer progression, with implications for immune modulation and DNA replication control.

Unveiling the enigmatic role of MTBP in pan-cancer: A bioinformatics perspective.

S100 protein family members S100A8 and S100A9 function primarily as a heterodimer complex (S100A8/A9) in vivo. This complex has been implicated in various cancers, including gastric cancer (GC). Recent studies suggest that these proteins play significant roles in tumor progression, inflammation, and metastasis. However, the exact mechanisms by which S100A8/A9 contributes to GC pathogenesis remain unclear. This study investigates the role of S100A8/A9 and its receptor in GC. Immunohistochemical analysis was performed on GC tissue samples to assess the expression of the S100A8/A9 receptor melanoma cell adhesion molecule (MCAM). In vitro transwell migration and invasion assays were used to evaluate the motility and invasiveness of GC cells. Cell proliferation was assessed using a growth assay, and Western blotting (WB) was employed to examine downstream signaling pathways, including ERK and the transcription factor c-Jun, in response to S100A8/A9-MCAM interaction. S100A8/A9 stimulation enhanced both proliferation and migration through MCAM binding in GC cell lines. These cellular events were accompanied by ERK activation and c-Jun induction. Downregulation of MCAM suppressed both ERK phosphorylation and c-Jun expression, highlighting the importance of the S100A8/A9‒MCAM‒ERK‒c-Jun axis in promoting GC progression. These findings indicate that S100A8/A9 contributes to GC progression via MCAM, which activates the ERK‒c-Jun pathway. The S100A8/A9‒signaling axis may represent a novel therapeutic target in GC.
